  • DOI 10.2460/javma.2004.225.414
  • PMID 15328718

DOAJ: no — l'interrogazione per ISSN 0003-1488 restituisce nessun risultato.

Licenza dell'articolo originale: nessuna licenza registrata.

Quanto pesa. Crossref registra 20 citazioni, OpenAlex 25 (letti il 31/08/2026). In ventidue anni.

Europe PMC reports not open access, not in PubMed Central, no PDF available; OpenAlex instead classifies the article as «bronze» OA and points to a PDF on the publisher's site. That address, queried on 08/31/2026, returned HTTP 500; a second display address returned HTTP 200 with 1,444 bytes, i.e. a block page and not an article. This card has not read the full text and does not claim it as available.

  • Article open access status: yes, but without a license (free to read, but no declared license). Unpaywall, queried on 09/03/2026: the full text is readable free of charge on the publisher's site, which however declares no open license. It is a revocable courtesy, not a right: it can be read, not redistributed.

Journal status (verified 08/31/2026): Journal of the American Veterinary Medical Association, ISSN 0003-1488 (print) and 1943-569X (electronic); publisher American Veterinary Medical Association; NLM ID 7503067. PubMed: yes. DOAJ: no — the query for ISSN 0003-1488 returns no results. OpenAlex: 28,634 indexed works, not open access. It is the journal of the U.S. veterinary association: a natural home for clinical work on non-conventional species in North America.

License of the original article: no license registered. The license field of the Crossref response is null, and this is not the same as «open license»: it means the publisher has not deposited any reuse terms, so publisher copyright applies. This card may reproduce: factual data and short quoted excerpts from the structured abstract; no figures, no tables.

How much it weighs. Crossref records 20 citations, OpenAlex 25 (both read 08/31/2026). Over twenty-two years. This is a measure of the field's size more than of the work's value: this is camelid vaccine literature, and it is made of handfuls of articles cited by handfuls of articles.

Specie e soggetti

Alpaca, lama e cavallo insieme, in tre gruppi con calendari diversi.

  • 28 alpaca, sani;
  • 56 lama, sani. ⚠️ La sierologia non è stata fatta su tutti e 56: l'abstract dichiara «Samples from 29 randomly selected vaccinated llamas were used». Il denominatore delle percentuali sui lama è quindi 29, non 56, e ogni cifra di questa scheda lo rispetta;

Species and subjects

Alpacas, llamas and horses together, in three groups with different schedules.

  • 28 alpacas, healthy;
  • 56 llamas, healthy. ⚠️ Serology was not done on all 56: the abstract states «Samples from 29 randomly selected vaccinated llamas were used». The denominator for llama percentages is therefore 29, not 56, and every figure in this card respects that;
  • 16 horses, as a species comparison group.

Not stated in the accessible records: age, sex, breed, reproductive status, site or sites, origin (single herd or multiple herds?), prior vaccination status, and the criteria by which the 84 camelids were selected → to be checked against the full text. It is only stated that they were healthy («healthy alpacas and llamas»).

Not stated is the procedure by which the 29-llama subsample was drawn, beyond the word «randomly» → to be checked against the full text.

Domanda clinica (PICO)

Clinical question (PICO)

P — in healthy adult alpacas and llamas; I — administration of the equine vaccine against the West Nile virus, in three doses three weeks apart; C — versus horses vaccinated with the label schedule, two doses four weeks apart; O — the appearance of virus-neutralizing antibodies, measured before each dose and three weeks after the last.

What the PICO does NOT contain, and is the thing this card repeats several times: the outcome is not «getting West Nile disease». It is «having antibodies». No animal was exposed to the virus.

Gov't.

Study type and level of evidence

Comparative clinical trial, non-randomized, open-label, with a cross-species comparison. The structured abstract states the design as «Clinical trial»; PubMed classifies it as Journal Article · Research Support, Non-U.S. Gov't.

It is not a randomized controlled trial, and the distinction must be made precisely because the design's name is close: there is no group of unvaccinated camelids; the comparison is between different species subjected to different schedules, which means the «species» factor and the «number of doses» factor are confounded by construction. There is no randomization of assignment, no blinding, no placebo. In the accessible records there is not even a statistical analysis: there are proportions.

Level of evidence: uncontrolled prospective clinical study on a surrogate outcome. Consequence for the reader: it establishes how many administrations are needed to bring about the appearance of neutralizing antibodies in alpacas and llamas with a vaccine licensed for horses; it does not establish that those antibodies protect against disease, because there was no exposure to the virus and because — the authors themselves write — the protective titer in the camelid is not known.

Relevant reporting standard: REFLECT, the reporting checklist for RCTs in food-producing animals, obtained by modifying CONSORT; and it must be applied as a declared adaptation, because the study is not randomized and REFLECT was built for randomized trials. Alpacas and llamas in Europe sit on both sides of the food/companion-animal distinction (chapter 14, §3.2), and the reader of this card in Italy probably has them on the companion-animal and fiber side: there is no checklist for companion animals, and this card uses REFLECT by adapting it rather than inventing one. Declared by the authors: no — in 2004 REFLECT did not exist (it dates from 2010), and demanding it would be an anachronistic reproach. The checklist serves to say what is missing, not to give a grade.

In breve

In brief

In 2002–2003 the West Nile virus swept across North America, and camelid owners found themselves facing a problem the Italian veterinarian knows well for other diseases: there is no vaccine licensed for their species. There is a vaccine licensed for the horse. It is used off-label, and no one knows whether it works, nor with what schedule.

This work measures the only thing that can be measured without exposing animals to the virus: whether and when neutralizing antibodies appear. And the result takes a shape that recurs, in this issue, in an almost systematic way: after the first dose the camelid is nearly stalled, and it becomes seropositive only after the booster.

Why this study matters

Because West Nile is endemic in Italy, and because the reasoning this article dismantles is the one that comes naturally: he has the horse vaccine, I'll give it the horse schedule. The horse schedule, in this study, after the first dose brings the titer to 7 of 16 horses, and 4 of 28 alpacas and 4 of 29 llamas.

And because the authors' conclusion is a model of how to write an honest vaccine finding — so much so that it is worth reading in full, which the Interpretation field below does.

Obiettivo

Objective

Stated by the authors: to determine the humoral response to an equine vaccine against the West Nile virus in healthy alpacas and llamas, and to compare the response of alpacas and llamas with that of horses.

Metodi / Protocollo

Ciò che l'abstract strutturato dichiara:

  • cavalli: 2 somministrazioni a 4 settimane di distanza;
  • alpaca e lama: 3 somministrazioni a 3 settimane di distanza;
  • 55 lama hanno ricevuto una quarta somministrazione tre settimane dopo la terza;
  • prelievi: immediatamente prima di ogni somministrazione; 3 settimane dopo l'ultima per alpaca e lama; 4 settimane dopo l'ultima per i cavalli;

  • quale vaccino, di quale produttore, di quale tipo (spento? vettoriale? DNA?) e con quale numero di lotto → da verificare sul testo pieno;
  • quale dose (volume) e quale via di somministrazione → da verificare sul testo pieno. Nessuna delle due compare nell'abstract. Questa scheda non scrive un numero di dose perché non ne ha uno, e non lo deduce dall'etichetta equina;
  • quale prova di sieroneutralizzazione, con quale soglia di positività → da verificare sul testo pieno;
  • approvazione etica e consenso dei proprietari; finanziamento; conflitti.

Methods / Protocol

What the structured abstract states:

  • horses: 2 administrations 4 weeks apart;
  • alpacas and llamas: 3 administrations 3 weeks apart;
  • 55 llamas received a fourth administration three weeks after the third;
  • sampling: immediately before each administration; 3 weeks after the last for alpacas and llamas; 4 weeks after the last for horses;
  • outcome measured: presence of virus-neutralizing antibodies.

Not stated in the accessible records, and this is why this card's clinical utility is kept low:

  • which vaccine, from which manufacturer, of which type (killed? vectored? DNA?) and with which lot number → to be checked against the full text;
  • which dose (volume) and which route of administration → to be checked against the full text. Neither appears in the abstract. This card does not write a dose number because it does not have one, and does not infer it from the equine label;
  • which assay for seroneutralization, with which positivity threshold → to be checked against the full text;
  • ethical approval and owner consent; funding; conflicts.

Risultati (dati)

Sicurezza: «None of the animals developed any local or systemic adverse reactions.»

Results (data)

The proportions reported in the abstract, each with its denominator and its species:

After the FIRST administration (serology at 3 weeks for camelids, at 4 for horses):

SpeciesSeropositiveProportion
alpaca4 of 2814 %
llama4 of 2914 %
horse7 of 1644 %

After the SECOND administration:

SpeciesSeropositiveProportion
alpaca27 of 2896 %
llama26 of 2990 %
horse15 of 1694 %

After the THIRD administration (camelids only; horses, per protocol, had received two):

SpeciesSeropositiveProportion
alpaca28 of 28100 %
llama28 of 2997 %

After the FOURTH administration (55 llamas): no serologic data in the accessible records → to be checked against the full text. It is the abstract's most curious omission: fifty-five animals received one more dose and the result is not reported.

Safety: «None of the animals developed any local or systemic adverse reactions.» No local or systemic reaction in any of the 84 camelids and the 16 horses. Not stated is for how long and by what method the animals were observed → to be checked against the full text, and the difference between «we saw none» and «we looked and there were none» lies entirely there.

No numerical titer is reported in the abstract — no means, no ranges, no standard deviations, no threshold above which a serum was defined as positive → to be checked against the full text. This card therefore knows how many animals seroconverted and does not know by how much.

«Results suggest that vaccination with the equine WNV vaccine is safe in alpacas and llamas. Administration of 3 vaccinations generally resulted in virus-neutralizing antibody titers similar to those observed following 2 vaccinations in horses; however, because it is not known what antibody titer would be protective against clinical WNV disease in alpacas or llamas, we cannot conclude that the vaccine was efficacious.»

Authors' interpretation (attributed, not merged with the data)

The authors' conclusions should be reported in full, because the second half is the one the world has forgotten to cite:

«Results suggest that vaccination with the equine WNV vaccine is safe in alpacas and llamas. Administration of 3 vaccinations generally resulted in virus-neutralizing antibody titers similar to those observed following 2 vaccinations in horses; however, because it is not known what antibody titer would be protective against clinical WNV disease in alpacas or llamas, we cannot conclude that the vaccine was efficacious.»

Translated and attributed: the authors suggest that the equine vaccine is safe in the two species; the authors observe that three administrations in the camelid give titers similar to two in the horse; and the authors explicitly state that they cannot conclude that the vaccine is efficacious, because it is not known which titer protects.

That third sentence is, in substance, a voluntary renunciation of the most sellable conclusion of their own work. The card reports it as a matter of method as much as of content.

Limiti dichiarati dagli autori

Limitations declared by the authors

Only one appears in the abstract, and it is the decisive one: the protective titer in the camelid is not known, so efficacy is not demonstrated. Whether the full text discusses others is not verifiable → to be checked against the full text.

Limitations added by the editorial team

  • The comparison is confounded by construction. Horses received a different schedule from camelids (2 doses at 4 weeks versus 3 at 3 weeks). The difference after the first dose — 7/16 versus 4/28 and 4/29 — is the only clean comparison, because at that point everyone had received a single dose; but even there the interval before sampling was different (4 weeks in the horse, 3 in the camelid), and three weeks versus four favors the horse. The direction of the result is not overturned by this, its magnitude is.
  • No group of unvaccinated camelids. There is no way to exclude that some seropositivity was pre-existing, in a species and an area where the virus was circulating.
  • The subsample of 29 of 56 llamas is stated to be random but the procedure is not described, and the 27 excluded llamas are not characterized.
  • No titer, no statistics, no threshold in the accessible records: the proportions have no confidence intervals and the differences between species have no p-value.
  • No clinical outcome and no challenge trial, which is the limitation the authors declare and which bears repeating because it is the only one that truly matters.
  • Twenty-two years. The equine West Nile vaccine is no longer, in 2026, what it was in 2004: the licensed products have changed, and in Europe they are not the same as in the United States. This study says nothing about the vaccines licensed in Italy today.

Finanziamento

Funding

Not stated in the accessible records → to be checked against the full text. PubMed classifies the work as Research Support, Non-U.S. Gov't: there was some U.S. non-governmental external support, and who provided it is not known. Europe PMC reports no funding entries (empty funding list).

It should be said, as information and not as an accusation, that a study evaluating a commercial vaccine used off-label is precisely the type of study for which the source of support is decisive — and that the accessible record does not allow this to be read. It should also be said that the authors are a public university group and that the conclusion is unfavorable to product promotion, which is the strongest indication available in the absence of the data.

  • identificazione del prodotto in prova (nome, produttore, tipo, lotto);
  • dose e via di somministrazione;
  • descrizione della popolazione: età, sesso, razza, mandria o mandrie, criteri di inclusione ed esclusione, stato vaccinale anteriore;
  • numero di sedi e periodo di svolgimento;
  • metodo di assegnazione e procedura del campionamento casuale dei 29 lama;
  • assenza dichiarata di un gruppo di controllo non vaccinato, e la sua giustificazione;
  • la prova sierologica, la sua soglia di positività e i titoli ottenuti;
  • analisi statistica, valori di p, intervalli di confidenza;
  • durata e metodo dell'osservazione per gli eventi avversi;
  • approvazione etica, finanziamento, conflitti di interesse;

◆ COMMENTO DELL'ASSOCIAZIONE

Camelid Research Studies APS — redazione CRL · 31/08/2026

Valutazione a stelle

AsseVotoMotivazione

What is missing relative to the reporting standard

Relative to REFLECT/CONSORT applied as a declared adaptation to a non-randomized study, the following are missing from the accessible records:

  • identification of the product under trial (name, manufacturer, type, lot);
  • dose and route of administration;
  • description of the population: age, sex, breed, herd or herds, inclusion and exclusion criteria, prior vaccination status;
  • number of sites and study period;
  • assignment method and the random sampling procedure for the 29 llamas;
  • declared absence of an unvaccinated control group, and its justification;
  • the serologic assay, its positivity threshold, and the titers obtained;
  • statistical analysis, p-values, confidence intervals;
  • duration and method of observation for adverse events;
  • ethical approval, funding, conflicts of interest;
  • the outcome of the fourth administration in 55 llamas.

Many of these items are, in all likelihood, in the full text. This card has not seen them and does not assert them.

◆ ASSOCIATION COMMENTARY

Camelid Research Studies APS — CRL editorial team · 08/31/2026

After the first dose the response is almost absent, and the label schedule is the horse's.

Anyone summarizing this article writes «three doses give 96–97 %». It is true and it is not the interesting part. The interesting part comes before: 4 of 28 alpacas and 4 of 29 llamas had antibodies after the first administration, versus 7 of 16 horses. Fourteen percent versus forty-four. The camelid is not a slow horse: after one dose it is nearly stalled.

This, for a veterinarian working in Italy, is not an exotic data point. West Nile circulates, there is no vaccination mandate, camelids have no licensed vaccine, and the question «shall I give it the horse's?» arrives every summer. The answer this study allows is neither yes nor no: it is «if you give it, the label schedule is not its schedule». An alpaca vaccinated once in June is, in all likelihood, an unvaccinated alpaca.

And here we must be strict with ourselves, not with the authors. This article, read as we were able to read it, does not authorize us to write a schedule. We do not know which product was used — and in 2004, in the United States — we do not know how many milliliters, we do not know by which route, we do not know which titer was considered positive. Chapter 14 of this KT has a rule that seems pedantic until it is needed: no clinical-utility score of three stars or higher can be based on doses, routes, or protocols that the article does not report in full. Here the temporal protocol is present (three doses, three weeks) and the dose is not. So axis B stops at two stars, and we write it with the reasoning in plain sight instead of rounding up because we like the message.

The message we can give, and that needs no such number, is another one: the booster is not optional, and the camelid that has received a single dose of any vaccine must be considered unprotected until there is reason to think otherwise. This is stated jointly by this card, G-04 and G-05, across three diseases and two different species, and it is the only generalization this issue allows itself.

Where we ask for more — and we ask for little, because the little already exists. Fifty-five llamas received a fourth dose in this study and the result is not in the abstract. We are not asking for a new study: we are asking that someone open the full text and tell us. It is the kind of work this series can do in an afternoon, once it has access to the journal, and it is marked among the things to do.

A note on access, that concerns everyone. The indexes classify this article as freely available on the publisher's site. On August 31, 2026 that address returned HTTP 500, and a second address returned 1,444 bytes of a block page. It is not an isolated incident: the same thing happened, with a different error, for card G-04. An article classified as open and unreachable is, for whoever must verify it, a closed article — and the cards in this series must say so, because the reader who takes us at our word deserves to know how much we were able to check.

Star rating

AxisScoreRationale
A · Scientific quality★★☆☆☆The prospective design is clear, the denominators are explicit for every species and every timepoint — including the honest correction from 56 to 29 llamas for serology — and the species comparison group is declared. But there is no randomization, no blinding, no group of unvaccinated camelids, the species factor and the schedule factor are confounded by construction, and the accessible record contains no titers, no positivity threshold, no statistical analysis, no ethical statement, no funding, and no conflicts. Two stars: chronology and denominators are present, controlled comparison and verifiable reporting are not.
B · Clinical utility★★☆☆☆The result would change a widespread practice — giving the camelid the equine label schedule — and this is what keeps it above a single star. It does not reach three for a rule-based reason, not a matter of taste: chapter 14 (§11, constraint on axis B) forbids a score ≥ 3 based on protocols that the article does not report in full, and here product, dose, and route are missing. It suggests an operational hypothesis to verify against the product one has on hand, not a protocol to apply.
C · Educational value★★★★☆It teaches two things, and the second is the rarer one. The first: the kinetics of the camelid's primary response differ from the equine's, and a schedule does not transfer with the vial. The second: how to write an honest conclusion — «three doses give titers similar to two in the horse; since it is not known which titer protects, we cannot conclude that the vaccine is efficacious». That sentence is coursework material, and it is worth more than the data accompanying it.
D · Species relevance★★★☆☆A study on camelids, on both species of interest to the European reader, with 84 animals: well above the norm in this field. It does not rise to four because the checklist requires, for the fourth star, a population described well enough to assess its transferability — and of these 84 animals the accessible record states no age, sex, herd, origin, or prior vaccination status. Add that the product is American and from 2004, and that nothing read here concerns the vaccines licensed in Italy today.

No overall score, by construction. B = 2 does not mean «unimportant»: it means that what is important here must be used as a warning, not as a recipe.

Bottom line

It can be concluded that, in this study, after a single administration of an equine vaccine against the West Nile virus, 4 of 28 alpacas and 4 of 29 llamas were seropositive for neutralizing antibodies, versus 7 of 16 horses; that after the second administration, 27 of 28 alpacas, 26 of 29 llamas, and 15 of 16 horses were seropositive; that after the third — given to camelids only — 28 of 28 alpacas and 28 of 29 llamas were seropositive; and that none of the 100 animals showed local or systemic adverse reactions.

It cannot be concluded that the vaccine protects: no animal was exposed to the virus and — as the authors state — it is not known which antibody titer is protective in alpacas and llamas. It cannot be concluded by how much each animal responded: titers, positivity threshold, statistical analysis, and confidence intervals are not in the accessible records → to be checked against the full text. A schedule cannot be written from this card, because product, dose, and route of administration are not reported → to be checked against the full text. It is not known what happened after the fourth dose received by 55 llamas: the data is not in the abstract. And none of this can be transferred to the vaccines licensed in Europe in 2026: the product is American and the study is twenty-two years old.

Messaggi da portare a casa

Take-home messages

  1. After the first dose, the camelid is nearly stalled: 4 of 28 alpacas and 4 of 29 llamas, versus 7 of 16 horses. The booster is not a reinforcement, it is the vaccination.
  2. The equine label schedule is not the camelid schedule. In this study three doses three weeks apart were needed to reach where the horse arrives with two four weeks apart.
  3. Seroconversion is not protection. The authors explicitly refuse to conclude that the vaccine is efficacious, because the protective titer in the camelid is not known. Anyone summarizing this article without that sentence betrays it.
  4. No adverse reaction in 84 camelids — with the caveat that it is not stated how and for how long they were observed.
  5. This study does not speak to the products licensed in Italy today, and anyone citing it to justify off-label use must say so.
