Camelid Research Studies APS — redazione CRL · 20/09/2026

◆ ASSOCIATION'S COMMENTARY
Camelid Research Studies APS — CRL editorial team · 20/09/2026
For once the «camelid» data was measured on an alpaca, not on a dromedary. It is the exception worth flagging in an issue built on the opposite rule.
What has actually been shown. That the elliptical shape and rigidity of the camelid red blood cell are not a morphological curiosity: they are the consequence of a different structural protein. The alpaca's 4.1R weighs 90 kDa instead of 80, because it includes a piece — the e14 cassette — that other mammals with biconcave red cells leave out. And the authors did not stop at description: they put that piece inside the human protein and measured what happens. What happens is that the affinity for β-spectrin rises five-fold (121 nM versus 633 nM) and the binding forms eleven times faster. The resulting skeleton is hyperstable: it holds its shape even when the membrane wrapping it is dissolved.
The detail that makes the article memorable, and that is worth teaching. The same effect is obtained with a human mutation that causes hereditary elliptocytosis. That is: what in humans is a red blood cell disease, in the camelid is the norm, written into the splicing program. This is not a metaphor — it is the same biophysical mechanism, reached by two different routes. Anyone who has to explain to a student why «normal» is a word that depends on the species has here the best example this collection has encountered so far.
What changes for a veterinarian in Italy, today. One thing only, and it is not a procedure: it is a well-founded caution. An automated haematology analyzer that has not been validated for camelids counts and sizes cells with algorithms calibrated on biconcave, deformable cells. This study establishes, on a molecular basis and with reconstruction experiments, that the alpaca erythrocyte is the opposite of both: ellipsoidal and rigid by construction. One cannot deduce from this how much the analyzer errs, and this card will not write that: the study does not measure it. What can be deduced is that the caution has a physical basis, and that reading the smear by eye is not a leftover from the past but a sensible control. The measurement of that error lies elsewhere, in CRL-A-01, and that is where it should be sought.
A limitation that must be stated with the same force as the result. The title says «camelid red cells». The bench saw adult male alpacas, and nothing else. No llama, no vicuña, no dromedary. The extension to the whole family is plausible and the authors argue for it, but this issue makes no concessions in either direction: just as it does not accept a dromedary finding passed off as camelid, it does not accept an alpaca finding passed off as valid for all — not even when the generalization would be convenient.
What this card cannot tell you. How many alpacas were used; whether the proportion of isoforms changes with age or in the cria; what happens to this skeleton in the course of haemolytic anaemia or Candidatus Mycoplasma haemolamae infection; and whether the llama has the same isoform. None of these questions was asked by the authors, and none is answered by the published data.

Full title, not truncated, in the form given by the Crossref record (queried on 20/09/2026).
2 · Titolo in italiano
⚠️ Attribution note, and it belongs in the same sentence as the title: the title says «camelid red cells», in the plural and for the whole family; the erythrocytes measured are from alpaca. The direction of the extrapolation, for once, is the opposite of the one this issue denounces elsewhere: here a New World finding is extended to all camelids. The extension is plausible and the authors argue for it (field 16), but it remains an extension, and the card flags it with the same rigor it would apply to the reverse case.

The abbreviated form of the Europe PMC record is: Chen Y, Miyazono K, Otsuka Y, Kanamori M, Yamashita A, Arashiki N, Matsumoto T, Takada K, Sato K, Mohandas N, Inaba M. — same order, same eleven people. Three ORCIDs out of eleven are registered; for the other eight no ORCID appears either in Crossref or in PubMed.
A name worth recognizing. The tenth author, Narla Mohandas (Red Cell Physiology Laboratory, New York Blood Center), is one of the internationally leading names in the biophysics of the human erythrocyte membrane. His presence indicates that this is not a veterinary work that happens to look at camelids: it is a basic hematology work that uses the alpaca as a model to understand the red blood cell membrane in general — and field 22 explains why the difference matters.

9 · Specie e soggetti
Specie dell'esperimento animale: alpaca (Vicugna pacos) — camelide sudamericano, Nuovo Mondo.
«Adult male alpacas and dogs were housed at the animal experimentation facility of the Graduate School of Veterinary Medicine, Hokkaido University.»
Dal testo pieno: «All experimental procedures were reviewed and approved by the Laboratory Animal Experimentation Committee, Graduate School of Veterinary Medicine, Hokkaido University with an approval number 16-0085.»
9 · Species and subjects
Species of the animal experiment: alpaca (Vicugna pacos) — South American, New World camelid. No Old World camelid was used. The MeSH term assigned by the National Library of Medicine is «Camelids, New World», which independently confirms the attribution.
From the full text, Experimental procedures · Animals section, verbatim:
«Adult male alpacas and dogs were housed at the animal experimentation facility of the Graduate School of Veterinary Medicine, Hokkaido University.»
Hence: adult male alpacas, housed in a Japanese university facility, together with dogs — which in the study serve as a comparison species and as a source of spectrin for the in vitro experiments.
⚠️ How many alpacas? The number of animals is not stated, either in the abstract or in the Animals section of the full text, which we read in full → to be checked against the full text (in the supplementary material, which was not retrieved). What is stated instead are the experimental numerosities, which are a different matter and which the card reports in field 15: n = 20 for the ellipticity measurements (cells, not animals), n = 3-4 for the viscosity assays, n = 3 for surface plasmon resonance.
Ethical approval: present, and with the number. From the full text: «All experimental procedures were reviewed and approved by the Laboratory Animal Experimentation Committee, Graduate School of Veterinary Medicine, Hokkaido University with an approval number 16-0085.» — this is an element that carries weight in field 23, and one that many of the articles catalogued in this collection lack.
Comparison materials, all declared: human, canine and bovine erythrocytes (for the electrophoretic comparison of the 4.1R protein); recombinant human proteins expressed in Escherichia coli, including human 4.1R carrying the e14 cassette and a human 4.1R with a duplication of the spectrin-actin binding domain, which is one of the known mutations of human hereditary elliptocytosis.
10 · Domanda clinica (PICO)
10 · Clinical question (PICO)
The PICO format does not apply: there is no patient, no clinical intervention, no clinical outcome. This is molecular biology of the cell membrane.
The article's question, in the authors' words (abstract): camelid red blood cells are flat ellipsoids with decreased deformability, but «the mechanisms responsible for their ellipsocytic shape and decreased deformability have not been determined». Restated by the editorial team as a comparison:
- Object: the alpaca erythrocyte membrane skeleton.
- Comparison: erythrocytes of mammals with a deformable biconcave disc shape (human, dog, bovine) and normal human 4.1R protein.
- Outcomes measured: presence and size of the 4.1R isoform; capacity to form the spectrin-actin-4.1R ternary complex; binding affinity for β-spectrin; cellular ellipticity along erythroid maturation.
11 · Disegno dello studio
11 · Study design
Experimental laboratory study, with three legs: (a) morphological and biochemical characterization of alpaca erythrocytes; (b) identification and sequencing of the protein isoform; (c) functional in vitro reconstruction with mutagenized recombinant human proteins.
⚠️ As with CRL-K-01, no entry in the closed list of chapter 14 §1.3 describes this design: the list is built for clinical literature. The most honest definition is «molecular biology experimental study, with an animal component limited to blood sampling».
12 · Level of evidence
Experimental laboratory study — outside the clinical hierarchy.
Bottom-line for the reader: this work establishes a molecular mechanism, with reconstruction experiments that are the strongest form of evidence available in cell biology. It establishes nothing clinical: it does not say what happens to the alpaca red blood cell in the course of disease, does not measure erythrocyte survival, does not produce reference values and does not indicate any treatment. The bridge between this mechanism and the clinic is editorial reasoning, and it is in field 22, marked as such.
13 · Relevant reporting standard
ARRIVE 2.0 for the animal component (housing and blood sampling from alpacas and dogs). The authors do not declare having followed ARRIVE, nor any other checklist.
It should however be recorded that the article in fact satisfies some essential ARRIVE items that many works in the camelid literature do not satisfy: ethical approval with protocol number, species and sex of the animals (adult male alpacas), sample size stated for each experiment, statistical methods named for each comparison. The item that remains uncovered is the most basic one: how many animals (the other missing or partial items are in field 21).
14 · Metodo
Letto nel testo pieno (PMC9926112, recuperato il 20/09/2026). In sintesi, e per gamba sperimentale:
(a) Morfologia. L'ellitticità è definita dagli autori come (a − b) / a.
14 · Method
Read in the full text (PMC9926112, retrieved on 20/09/2026). In summary, by experimental leg:
(a) Morphology. Alpaca peripheral blood smears stained with new methylene blue. Cells were classified into three stages — aggregate reticulocytes (youngest), punctate reticulocytes (maturing) and mature erythrocytes — and for each the major diameter (a) and minor diameter (b) were measured. Ellipticity is defined by the authors as (a − b) / a. In parallel, erythrocytes were treated with 1% Triton X-100 in PBS for 30 minutes on ice and the shape of the residual skeleton («Triton shell») was observed under the light microscope.
(b) Membrane biochemistry. 8% SDS-PAGE electrophoresis of membrane proteins from human, canine, bovine and alpaca erythrocytes, staining with Coomassie brilliant blue (7 µg per lane) and immunoblot with anti-canine 4.1R antibody (3 µg per lane). Fractionation of membrane ghosts in 1% Triton X-100 with centrifugation at 100,000 g for 30 minutes to separate supernatant and pellet. Sequence identification by tandem mass spectrometry (MS/MS). A dedicated antibody — anti-e14 — was raised in rabbit against the synthetic peptide NH2-419VEKTHIEVTVPTSDGDQTQ437-COOH.
(c) Functional reconstruction. Recombinant human 4.1R proteins, fused to GST, expressed in E. coli BL21(DE3)pLysS and purified on glutathione-Sepharose resin; spectrin extracted from canine erythrocyte membranes and separated into dimers and tetramers by gel permeation chromatography, with conversion of dimers to tetramers at 30 °C for 2 hours; commercially sourced rabbit muscle actin. Formation of the spectrin-actin-4.1R ternary complex was measured with viscosity assays; the affinity for β-spectrin with surface plasmon resonance.
Statistics declared: one-way ANOVA with Tukey's multiple comparison test for ellipticity; one-way ANOVA with Dunnett's test for the viscosity assays; unpaired t-test for the surface plasmon resonance comparisons. The statistical methods are named comparison by comparison — which is by no means the norm in this collection.
⚠️ What the method does not say → to be checked against the full text (supplementary material, not retrieved): how many alpacas provided blood, their age, the volume drawn, the frequency of sampling and the animals' fate.
15 · Risultati (dati)
Solo numeri, con la loro provenienza. I valori sono copiati dal testo pieno.
A · La cellula
|---|---|---|---|---|
15 · Results (data)
Numbers only, with their source. The values are copied from the full text.
A · The cell
- The ellipticity of alpaca erythroid cells increases during terminal erythroid differentiation, as immature reticulocytes mature into erythrocytes. Comparison across the three stages (aggregate reticulocytes R1, punctate reticulocytes R2, erythrocytes E), n = 20, p < 0.0001 (one-way ANOVA with Tukey's test). ⚠️ The numerical ellipticity values for the three stages are in Figure 1A, not in the text, and the figure cannot be converted into numbers by this card → to be checked against the full text. What the text states, and what is reported here, is the direction and significance, not the three figures.
- Alpaca erythrocytes retain their ellipsoidal shape even after solubilization of the plasma membrane in 1% Triton X-100: the residual skeleton is ellipsoidal. Qualitative data, not numerical.
B · The protein
- In alpaca erythrocyte membranes the 4.1R protein contains a cassette derived from the alternative exon 14 (e14), absent from the highly conserved 80 kDa 4.1R of other mammals with deformable biconcave erythrocytes.
- Together with the disordered proline- and glutamic-acid-rich peptide (PE) that precedes it, this results in a 90 kDa camelid 4.1R, migrating more slowly on electrophoresis than the 80 kDa 4.1R of human, dog and bovine analysed on the same gel.
- Peptide sequence against which the anti-e14 antibody was raised:
419VEKTHIEVTVPTSDGDQTQ437. The authors state that the amino acid sequence encoded by alternative exon 14 is identical across several species, including alpaca, dog and human: what changes in the camelid is not the exon's sequence, it is the fact that it is included.
C · The reconstructed function — the strongest numbers in the article
| Measure | Human 4.1R with e14 + PE | Human 4.1R normal (WT) | n | p |
|---|---|---|---|---|
| Equilibrium dissociation constant (K_D) for the β-spectrin N-terminal fragment | 121 ± 15 nM | 633 ± 112 nM | 3 per group | < 0.01 |
| Association rate constant (k_ass) | (55.07 ± 8.21) × 10³ M⁻¹ s⁻¹ | (5.04 ± 2.16) × 10³ M⁻¹ s⁻¹ | 3 per group | < 0.001 |
That is: the variant carrying the camelid cassette has a K_D five times lower — meaning affinity five times higher — and associates with β-spectrin roughly eleven times faster. (The ratios 633 / 121 ≈ 5.2 and 55.07 / 5.04 ≈ 10.9 are editorial divisions performed on the published values, not statements by the authors: the authors write «five-fold lower» for the first and do not give a ratio for the second.)
- In the viscosity assays, human 4.1R containing e14 and PE — but not PE alone — shows a markedly increased capacity to form the spectrin-actin-4.1R ternary complex. Reactions run at 10, 15 and 20 µg/ml of recombinant protein, n = 3-4, ANOVA with Dunnett's test. ⚠️ The viscosity values are in a figure → to be checked against the full text.
- A similar effect is obtained with a human 4.1R carrying a duplication of the spectrin-actin binding domain, which is one of the known mutations causing human hereditary elliptocytosis.
16 · Authors' interpretation
(explicitly attributed, not merged with field 15)
From the abstract and discussion, verbatim in summary:
- «These results indicate that 4.1R protein with the e14 cassette leads to the formation and maintenance of a hyperstable membrane skeleton, resulting in rigid ellipsoidal red cells in camelid species».
- «…and suggest that membrane architecture is evolutionarily regulated by alternative splicing of exons of the 4.1R gene».
- In the discussion the authors write that camelid 4.1R is unique among erythrocyte 4.1R proteins in containing the e14 cassette, which is absent from 4.1R80, the major isoform in the membranes of other mammals.
- The authors also note, and the card reports this because it is the only mention of biological significance for the animal, that reduced deformability «might be disadvantageous for the animals»: this is a hypothesis, stated conditionally, and not a measurement made in the study.
⚠️ The step from «alpaca» to «camelids» takes place here, in the authors' conclusion, and is not supported by measurements on other camelid species within this work. It is a reasonable inference — the ellipsoidal erythrocyte morphology is described across the whole family — but it remains an inference.
17 · Limiti dichiarati dagli autori
- il significato fisiologico della ridotta deformabilità per l'animale non è stato studiato, ed è lasciato come ipotesi al condizionale (campo 16);
17 · Limitations declared by the authors
No limitations are declared in a dedicated section, which is not a mandatory item in JBC's editorial practice for research articles.
Elements that function as limitations, stated by the authors in the body of the text:
- the physiological significance of the reduced deformability for the animal was not studied, and is left as a conditional hypothesis (field 16);
- the functional reconstruction was carried out with mutagenized human proteins and canine spectrin, not with alpaca proteins: it is a model of the mechanism, not the alpaca's own mechanism reproduced with its own components.
18 · Limitations added by the editorial team
- The number of alpacas is not stated. It is the most serious of the basic ARRIVE items that are missing (field 21), and it is missing from the full text, not just the abstract. All the published
nvalues are numbers of cells or of technical replicates. - All the alpacas were adult males. No data on females, young animals or crias. If the proportion of 4.1R isoforms were to change with age — and the study itself shows that ellipticity changes with cell maturation — the picture in the first months of life might not be the same.
- No data on sick animals. All measurements come from presumably healthy facility animals. What happens to this hyperstable skeleton in the course of haemolytic anaemia, Candidatus Mycoplasma haemolamae infection, or severe dehydration is not addressed by the study.
- The bridge to the clinic is not built by the authors. The work explains why the cell is rigid; it does not measure whether and how much this affects the reading of an automated haematology analyzer, which is the question of interest to a veterinarian. Whoever takes that step — as field 22 of this card does — is doing so on their own account.
- The generalization to «camelids» is not measured. No llama, vicuña, guanaco, dromedary or Bactrian erythrocyte appears among the materials. The title says camelids, the bench says alpaca.
- The comparison animals are dog, human and bovine, not a small ruminant. For a veterinarian working with camelids the everyday term of comparison is the sheep, which is absent here.

20 · Conflitti di interesse
Dichiarati, e negati. Dal record PubMed, campo CoiStatement, verbatim:
«Conflict of interest — The authors declare that they have no conflicts of interest with the contents of this article.»
20 · Conflicts of interest
Declared, and denied. From the PubMed record, CoiStatement field, verbatim:

- Animali sperimentali — presenti in parte: specie e sesso sì (alpaca maschi adulti), età, peso e provenienza no.
21 · What is missing relative to the reporting standard (ARRIVE 2.0, for the animal component)
Of the ten essential ARRIVE 2.0 items listed below, five are fully present (design, outcome measures, statistical methods, procedures, results) and one is not applicable (randomization). The other four are not satisfied: two are absent (inclusion and exclusion criteria, blinding of assessment) and two are only partly present (sample size, absent for the animals; experimental animals, without age, weight or source). The most important is the number of animals:
- Study design — present.
- Sample size — ⚠️ present for the experiments (n = 20 cells; n = 3-4 viscosity assays; n = 3 surface plasmon resonance), absent for the animals: how many alpacas provided the blood is not stated. This is the principal gap.
- Inclusion and exclusion criteria — absent. It is not stated how the animals were selected among those available in the facility, nor whether any sample was excluded.
- Randomization — not applicable to the design.
- Blinding of assessment — absent. The diameter measurements on stained smears are operator-dependent and observer blinding is not stated.
- Outcome measures — present and operationally defined: ellipticity has its formula, (a − b) / a.
- Statistical methods — present and named for each comparison (Tukey, Dunnett, unpaired t-test).
- Experimental animals — present in part: species and sex yes (adult male alpacas), age, weight and source no.
- Experimental procedures — present and detailed (concentrations, times, temperatures, centrifugation speeds).
- Results — present with mean, standard deviation, n and p-value for the numerical comparisons; the ellipticity and viscosity values, however, are only in the figures, and a number that exists only in a graph is not a reusable number.
23 · Valutazione a stelle
23 · Star rating
Four independent axes, criteria from chapter 14 §11. The full text was read: this rating does not carry the access caveat that applies to K-01, K-04 and K-05.
| Axis | Score | Rationale, with one fact inside |
|---|---|---|
| A — Scientific quality | ★★★★☆ | Experimental study with ethical approval and protocol number (16-0085), sample size stated for each experiment, statistical methods named comparison by comparison (Tukey, Dunnett, t-test), experimental conditions reported down to concentrations and times, funding with project numbers and conflicts declared. It does not reach five because the number of animals is stated nowhere, inclusion criteria are missing, observer blinding is not declared for an operator-dependent measure, and some values exist only in the figures. |
| B — Clinical usefulness | ★★☆☆☆ | Provides a hypothesis to be tested — that an unvalidated analyzer is a source of error in this species — not an applicable procedure: the study measures no clinical parameter, does not quantify instrumental error and proposes no threshold. The ★★★ criterion requires a procedure replicable in the clinic, which does not exist here. (Chapter 14 §11 constraint respected: B does not exceed A by more than two stars.) |
| C — Educational value | ★★★★★ | It is the first-choice reference on why the camelid erythrocyte has that shape: no other work reaches the molecular mechanism, and the reconstruction with the human hereditary elliptocytosis mutation makes the concept teachable in ten minutes. Anyone who has to learn this starts here. |
| D — Species relevance | ★★★★☆ | The mechanism was identified directly in alpaca erythrocytes, not extrapolated: for the European reader who treats alpacas this is the maximum species relevance this field offers. It does not reach five because the population studied — facility adult male alpacas, in an undeclared number — is not described well enough to assess its representativeness, and no other camelid was measured. |
24 · Bottom line

It can be concluded that: in alpaca erythrocytes the membrane skeleton protein 4.1R contains an alternative exon 14 cassette absent from the 80 kDa 4.1R of other mammals with deformable biconcave erythrocytes, and consequently weighs 90 kDa; that the experimental insertion of that cassette (with the preceding PE peptide) into a human 4.1R increases its affinity for β-spectrin roughly five-fold (K_D 121 ± 15 nM versus 633 ± 112 nM, p < 0.01, n = 3) and its association rate roughly eleven-fold (p < 0.001); and that the same facilitation of the ternary complex is obtained with a human mutation causing hereditary elliptocytosis. The ellipticity of alpaca cells increases with erythroid maturation (p < 0.0001, n = 20).
It cannot be concluded that: the same mechanism has been demonstrated in llama, vicuña, guanaco, dromedary or Bactrian camel — none of these animals appears among the materials, and the extension to «camelids» is an inference by the authors; that erythrocyte rigidity has measured clinical consequences, because the study measures none; that a haematology analyzer errs by a known amount in this species, because the study does not touch analyzers at all; and nothing about the cria, about females, or about sick animals.

- CRL-K-01 (this issue) — Jürgens et al. 1988: the same cell, from the functional side. K-01 measures that the camelid red blood cell is small (21-30 µm³) and that this compensates for the low haematocrit; K-02 explains why it is elliptical and rigid. The two works are thirty-five years apart and do not contradict each other: they describe two different properties of the same cell, volume and deformability.
- CRL-A-01 — the card in issue A on automated blood counts in camelids and on the parameters where the analyzer lies. It is the missing link between K-02 and the clinic: K-02 gives the physical reason, A-01 gives the measurement of the error. Whoever cites K-02 to say that «the analyzer is wrong» is skipping a step that already exists in this collection.
- CRL-K-03 (this issue) — the methodological reverse of this card: there a dromedary finding is read as camelid, here an alpaca finding is extended to all. The editorial team flags both extensions by the same yardstick, and field 22 of K-02 says so.
