Valutazione redazionale, quattro assi separatiEditorial rating, four separate axes
- AQualità scientifica3/5
- BUtilità pratica4/5
- CValore didattico4/5
- DTrasferibilità ai camelidi europei3/5
Che cosa misurano i quattro assi
- A · Qualità scientifica
- La domanda è ben posta? I metodi sono dichiarati? La statistica è appropriata? La conclusione sta dentro i dati?
- B · Utilità clinica
- Serve a chi ha l’animale davanti, oggi.
- C · Valore didattico
What the four axes measure
- A · Scientific quality
- Is the question well posed? Are the methods stated? Is the statistics appropriate? Does the conclusion stay within the data?
- B · Clinical usefulness
- Useful to whoever has the animal in front of them, today.
- C · Teaching value
- Does it teach something to those studying the subject?
- D · Relevance to the species
- Is it really about camelids, or a result borrowed from another species?
Each axis is worth 1 to 5 stars and carries its written reason. The stars in the index are the rounded average of the four.

1 · Titolo originale
Titolo parallelo in inglese stampato dalla rivista: «Comparative pharmacokinetics of a single dose of ampicillin following intravenous, intramuscular and subcutaneous administration to llamas».)
(Full title, copied from Crossref works/10.4067/s0301-732x2014000200013 and confirmed on the publisher's PDF. Parallel English title printed by the journal: «Comparative pharmacokinetics of a single dose of ampicillin following intravenous, intramuscular and subcutaneous administration to llamas».)
⚠️ Registration divergence, the third of this fascicle — and a genuine trap. The Crossref record declares
language: "en"for this article. That is wrong. The body — Introducción, Material y métodos, Resultados, Discusión, Agradecimientos — is entirely in Spanish; English appears only as the parallel summary. The SciELO ArticleMeta record (S0301-732X2014000200013, collectionchl) correctly registersv40 = "es". Language was verified by reading the article, not by trusting the field. Anyone filteringlanguage:enin Crossref would think they had an English paper; anyone filteringlanguage:eswould not find it at all. The same"en"appears across the Archivos de medicina veterinaria archive on visibly Spanish articles.

The journal no longer exists. Archivos de medicina veterinaria ceased with vol. 48 no. 3 (2016). This changes how the card should be used: no correction, erratum or confirmatory work will ever appear in this same title. The research continued elsewhere — the university now publishes Austral Journal of Veterinary Sciences — but this article's thread, on this journal, is closed.
Access note, for anyone repeating the verification. On 31/08/2026
www.scielo.clreturned 403 Forbidden to every automated read, both for the HTML page and via WebFetch. The full text was obtained by two routes that did work: the publisher's PDF (www.scielo.cl/pdf/amv/v46n2/art13.pdf, 94,255 bytes) and the legacy mirrorscielo.conicyt.cl. A 403 does not mean the article is gone.
8 · Licenza dell'articolo
8 · Article licence
Not declared on the article. The publisher's PDF carries no licence statement, and the Crossref license field is empty. The SciELO mirror page shows no Creative Commons mark for this article.
What is certain: SciELO registers the article as open access (v71 = "oa"). But open access is not a licence: it says you may read it, not what you may reproduce.
What this card therefore reproduces: only numerical data — not copyrightable — and attributed short quotations, under the right of quotation. Cuadro 1, Cuadro 2 and Figura 1 are not reproduced: their values are reported as text, with attribution. Licence status: [TO BE CHECKED WITH THE PUBLISHER].
9 · Specie e soggetti
Specie: lama (Lama glama).
- 6 lama adulti, come dichiarato: «seis llamas adultas»
- Età: da 4 a 7 anni
- Peso: 117,0 ± 15,5 kg (media ± deviazione standard).
9 · Species and subjects
Species: llama (Lama glama). Not alpaca. The authors compare their values with published alpaca figures, but the data are llama data, and this card repeats that at every number.
- 6 adult llamas, as declared: «seis llamas adultas»
- Age: 4 to 7 years
- Weight: 117.0 ± 15.5 kg (mean ± SD). Each llama was weighed on the day of treatment to calculate the dose — not decorative: it means the 20 mg/kg are real mg/kg, not estimated
- Sex: not declared →
[TO BE CHECKED IN THE FULL TEXT]. In a pharmacokinetic study this matters - Health status: clinically healthy on physical and haematological examination, both declared
- Housing: all llamas remained stabled in a shed («estabuladas en un galpón») throughout the experiment, with ad libitum access to green feed and water
No protocol number given → [TO BE CHECKED IN THE FULL TEXT].
Country: Argentina. The single affiliation and the corresponding author's address are in Buenos Aires.
Altitude: NOT DECLARED. The article does not say where the shed was or at what altitude. This card does not infer it from the university's address: that the corresponding author's postal address is in Buenos Aires does not prove the llamas were there. →
[TO BE CHECKED IN THE FULL TEXT: housing location and altitude].Why altitude weighs more in this card than elsewhere. Ampicillin is renally eliminated and its clearance depends on glomerular filtration; high-altitude adaptation changes haematocrit, blood volume and volumes of distribution. A clearance measured at 4000 m and one measured at sea level are not automatically the same number, and camelid literature on this is thin. Because the article is silent, the card asserts neither lowland nor highland origin: it declares the datum missing. For readers in Italy, though, this is the one card of the five whose animals are not certainly high-altitude, which is one more reason to use it.
10 · Domanda clinica (PICO)
10 · Clinical question (PICO)
In healthy adult llamas [P], does ampicillin sodium 20 mg/kg given subcutaneously [I], compared with the intramuscular and intravenous routes [C], produce a different pharmacokinetic profile and time above the minimum inhibitory concentration (T > MIC) [O]?
(Framed by the editors from the authors' stated objectives: to «describir y comparar la farmacocinética y biodisponibilidad» and «calcular los correspondientes T > CIM … a fin de proveer una base farmacológica para la terapia antimicrobiana racional en esta especie».)
11 · Disegno dello studio
11 · Study design
Experimental in vivo study, randomised crossover, single dose, three routes, with a 2-week washout between treatments.
Note on the closed list in §1.3. None of the listed values describes this well: it is not a clinical RCT, and not a «non-randomised controlled study» either, because randomisation is present — but it is randomisation of the sequence of routes, not of allocation to groups, and there is no control group because each animal is its own control. The card spells this out rather than forcing it into a box, as chapter 14 requires when the taxonomy does not cover the case.
12 · Level of evidence
Experimental within-subject controlled study, non-clinical, in 6 animals, single-centre.
Consequence line for the reader: these numbers describe how ampicillin behaves in six healthy adult llamas. They demonstrate clinical efficacy in no infection: no animal was ill, no infection was treated, no clinical outcome was measured. The dosing intervals the authors propose come from a PK/PD calculation using MICs taken from the literature, not from camelid clinical isolates — and the authors say so themselves.
13 · Applicable reporting standard
ARRIVE 2.0 — this is experimental in vivo research, so the NC3Rs guideline applies. REFLECT is relevant as a frame, because the llama is treated here as a food-producing animal (the authors explicitly discuss meat withdrawal times), but REFLECT is for RCTs and this is not one.
Declared by the authors: neither. The article names no guideline.

Differenze significative dichiarate (P < 0,05), i.m. contro s.c.: Cmax (34,1 contro 18,6 µg/ml), tmax (0,26 contro 0,6 h), t½λ (0,83 contro 1,66 h), TMR (1,3 contro 2,7 h), TMA (0,72 contro 2,1 h).
| CIM₉₀ | e.v. | i.m. | s.c. |
|---|---|---|---|
| 4,0 µg/ml | 1,5 (1,3-1,7) | 3,0 (2,3-3,7) ᵃ | 4,3 (3,2-5,5) ᵇ |
| 0,5 µg/ml | 3,0 (2,3-3,7) | 5,4 (4,2-6,6) ᵃ | 8,4 (6,1-10,7) ᵇ |

The dosage — the only therapy figure in the whole fascicle
Dose: 20 mg/kg. Drug: ampicillin sodium (BioAmpi, 200 mg/ml, aqueous solution). Species: adult llama (Lama glama), 117.0 ± 15.5 kg. Routes: intravenous (jugular), intramuscular (semitendinosus), subcutaneous (thoracic skin fold). Single administration. Source: Kreil et al., Arch Med Vet 2014;46(2):271-276.
Dose, route, species and product are all four present: that is why the number is written. It is the dose administered in the study, not a dose recommended by any regulator.

Significant differences (P < 0.05), i.m. versus s.c.: Cmax (34.1 vs 18.6 µg/ml), tmax (0.26 vs 0.6 h), t½λ (0.83 vs 1.66 h), MRT (1.3 vs 2.7 h), MAT (0.72 vs 2.1 h).
Not significant: AUC₀₋ₙ, AUC₀₋∞ and bioavailability (f 1.5 ± 0.2 vs 1.6 ± 0.9) between i.m. and s.c. — that is, the amount absorbed is the same; the rate is what changes.
Editors' note on bioavailability. Both values can be recomputed from the AUC₀₋∞ given above: 49.1 / 33.0 = 1.49 for i.m. and 52.4 / 33.0 = 1.59 for s.c., that is 1.5 and 1.6 as in the article. These are values above 1, whereas an absolute bioavailability should not exceed 1 (more cannot be absorbed than was given). The full text is not readable from here and, as far as this card records, the article does not comment on it: the values are reported as published, without an explanation, and the comparison between the two routes (same amount absorbed) rests on the non-significant difference, not on the absolute value of f.
T > MIC₉₀ — mean (95 % confidence interval), hours, 6 llamas, 20 mg/kg:
| MIC₉₀ | i.v. | i.m. | s.c. |
|---|---|---|---|
| 4.0 µg/ml | 1.5 (1.3-1.7) | 3.0 (2.3-3.7) ᵃ | 4.3 (3.2-5.5) ᵇ |
| 0.5 µg/ml | 3.0 (2.3-3.7) | 5.4 (4.2-6.6) ᵃ | 8.4 (6.1-10.7) ᵇ |
ᵃ ᵇ different superscripts within a row = significant difference (P < 0.05). The i.m. and s.c. routes differ significantly for both MICs.
Internal inconsistency, found by the editors. In the discussion the authors write that concentrations stay above 4 µg/ml «durante al menos 1,0, 2,0 y 3,0 h» and above 0.5 µg/ml «por lo menos 2,0, 4,0 y 6,0 h» after i.v., i.m. and s.c. These six numbers do not match Cuadro 2, which gives 1.5 · 3.0 · 4.3 and 3.0 · 5.4 · 8.4. The phrase «al menos» makes them compatible as conservative roundings, but they are two different sets of numbers in one article, and citing one while believing you cite the other is an error. The card uses the Cuadro values — the measured ones, with their confidence intervals — and flags the other set as a discussion rounding.
Comparisons with the literature, reported by the authors as others' data (not measured here): in the alpaca, t½λ 0.6 ± 0.2 h and Clt 16 ± 3 ml/min·kg (Kreil et al.); in the llama, Clt 0.88 ± 0.16 ml/min·kg and t½λ 3.3 ± 0.5 h (Christensen et al. 1996); MRT 0.7 ± 0.1 h in the alpaca and 5.0 ± 0.6 h in the llama; Vss 0.6 ± 0.05 l/kg in the alpaca and 0.277 ± 0.08 l/kg in the llama.
16 · Authors' interpretation
Attributed, not merged with the data.
The authors conclude that the pharmacokinetic and therapeutic profile differs significantly between i.m. and s.c. and that, verbatim, «estas inyecciones no deben ser consideradas intercambiables».
The authors attribute the longer t½λ by the s.c. route to a flip-flop phenomenon: slower absorption becomes rate-limiting and dictates the apparent elimination rate. They note that absolute bioavailability was similar, «indicando que el grado de absorción de la ampicilina fue similar luego de las dos administraciones».
The authors propose — and this is a proposal, not a result.
This dosing scheme is not a study result. It is a PK/PD extrapolation built on T > MIC measured in 6 healthy llamas and on MICs taken from other species' literature. No sick llama was treated at these intervals. It sits here, in the interpretation field, with attribution, as KT chapter 14 §5.1 requires.
The authors add, as their own recommendation, that for marginally susceptible organisms — MICs near the ampicillin breakpoint — «se debería utilizar una dosificación mayor a la empleada en este estudio»; and that the i.v. route «provee una buena profilaxis» against surgical site infection in dental and abdominal surgery.
The authors recommend the s.c. route for two distinct reasons: the more favourable profile (concentrations above MIC for longer, hence wider dosing intervals) and, verbatim, «debido a la pequeña masa muscular de los CS, las inyecciones s.c. son más recomendables que las i.m.».
The authors warn about withdrawal times: llamas now provide meat as well as wool; since withdrawal time is calculated from the elimination rate, «la diferente permanencia de la ampicilina luego de la administración i.m. o s.c. debe ser considerada cuando se calcula el tiempo de retirada del preparado comercial». The article calculates no withdrawal time and proposes none.
The authors list as future work: multiple-dose pharmacokinetics, possible adverse effects, and ampicillin susceptibility of South American camelid clinical isolates — which, they say, they could not find.
17 · Limiti dichiarati dagli autori
- sulle CIM: «hemos utilizado CIM reportadas previamente para microorganismos aislados en especies veterinarias para el cálculo del T > CIM, ya que no hemos encontrado datos de aislamientos clínicos de CS»;
17 · Limitations declared by the authors
No «Limitations» section. Two explicit acknowledgements appear in the discussion and are quoted verbatim:
- on MICs: «hemos utilizado CIM reportadas previamente para microorganismos aislados en especies veterinarias para el cálculo del T > CIM, ya que no hemos encontrado datos de aislamientos clínicos de CS»;
- on differences from earlier literature: «pueden ser debidas a factores relacionados a los animales, a la preparación farmacológica y al diferente análisis farmacocinético aplicado por los distintos autores».
The first is the article's most important limitation, and the authors deserve credit for writing it.
- Animali sani, esito surrogato.
2011. Pharmacokinetics of ampicillin in Alpacas (Lama pacos).
18 · Limitations added by the editors
- Six animals, no sample-size justification. No power calculation, no rationale for n = 6. With n = 6, an SD of 0.9 h on a MAT of 2.1 h (s.c.) is large, and some T > MIC confidence intervals are wide: 3.2-5.5 h for 4 µg/ml s.c., 6.1-10.7 h for 0.5 µg/ml s.c. A dosing scheme built on these is more uncertain than the means suggest.
- No blinding declared, not even for reading the microbiological assay plates, where it would have been easy.
- Sex of the animals not reported (§9).
- No numbered ethics protocol (§9): the committee is named, approval declared, the number not.
- No conflict-of-interest statement (§20). Funding, by contrast, is declared and public (§19).
- Healthy animals, surrogate endpoint. T > MIC is a predictor, not a clinical outcome. The link between T > MIC and cure is well founded for beta-lactams, but it remains a model: no infection was cured in this study.
- Two different sets of T > MIC numbers in the same article (§15).
- A bibliographic error inside the article, verified by the editors. The text repeatedly cites «Kreil y col 2001»; the reference list prints the same item as «Kreil V, C Lüders, R Hallu, M Rebuelto, L Betancourt. 2011. Pharmacokinetics of ampicillin in Alpacas (Lama pacos). Arch Med Vet 33, 241-246» — year 2011 but volume 33. The editors checked which year is right, querying Crossref: the work exists with DOI
10.4067/s0301-732x2001000200013, title «Farmacocinética de la ampicilina en Alpacas (Lama pacos)», authors KREIL V, LUDERS C, HALLU R, REBUELTO M, BETANCOURT L, Archivos de medicina veterinaria 2001;33(2). The correct year is 2001, as the body text says, and the reference list prints the wrong year. The card does not «fix» someone else's reference: it flags the error and publishes the verified record. - Something this card cannot resolve and the Italian reader must know. This is an Argentine study using an Argentine product. It says nothing about the conditions of use of ampicillin in camelids in Europe — authorisation, the veterinary medicine cascade, applicable withdrawal times. Those are regulatory matters, they live in KT chapter 05, and they cannot be deduced from a pharmacokinetic figure.

- Protocollo etico numerato (voce 9)
23 · Valutazione a stelle

21 · What is missing against the reporting standard (ARRIVE 2.0)
Applied retrospectively as a lens, not a reproach: ARRIVE 2.0 is from 2020, the article from 2014.
Of the Essential 10 — what is there: experimental design described; complete experimental procedures (dose, route, site, formulation, timings); animal details (species, age, weight, health status); results with a measure of dispersion; ethical approval named.
What is missing: sample size (item 2) declared but not justified; randomised allocation (item 4) — the sequence is called randomised but the generation method is not described; blinding (item 5) not reported for any phase; inclusion/exclusion criteria (item 3) — «healthy on physical and haematological examination», with no thresholds and no statement of exclusions; sex (item 8); numbered ethics protocol (item 9); conflicts of interest (Recommended Set).
What is not missing, and should be said plainly: the part that makes the article usable. Dose, product, concentration, route, anatomical site, sampling times, and the analytical method with its CVs and limit of quantification are all reported. That is more than many pharmacokinetic studies in indexed journals contain.
22 · Association's editorial comment
This is the card that on its own justifies the fascicle, and the reason is one phrase: «no deben ser consideradas intercambiables».
In daily practice i.m. and s.c. are treated as variants of the same gesture — chosen by how cooperative the animal is, not by pharmacology. This work measures that in the llama, at equal dose and equal amount absorbed, the subcutaneous route holds concentrations above the MIC about one and a half times as long as the intramuscular (4.3 / 3.0 = 1.4 and 8.4 / 5.4 = 1.6): 4.3 h versus 3.0 h at MIC 4 µg/ml, 8.4 h versus 5.4 h at MIC 0.5 µg/ml. For a time-dependent antibiotic, where what counts is how long you stay above the threshold rather than how high you go, that is the difference between one interval and another.
There is also an anatomical argument the authors put at the end, and it is worth double in Europe where camelids are often kept as companion animals: South American camelids have small muscle mass. The s.c. route is not only pharmacologically more convenient here: it is more practicable on a lean animal, with less risk of muscle damage and injection-site reaction. The two reasons converge, and they converge on the route that is also the easiest to perform in the field.
What this card does not authorise. It does not authorise giving ampicillin to a camelid in Italy on the strength of these numbers. Drug choice, off-label use, the authorised product and the withdrawal time are regulatory and clinical decisions this article does not touch, and on which the authors themselves stop: they say withdrawal time must account for the route, and they do not calculate it. The card helps you frame a better question, not skip the prescription.
One last note, of method rather than clinic. This article is from 2014, the journal closed in 2016, it is not in PubMed, and Crossref classifies it as English when it is Spanish. For an Italian series to find it, four things must coincide: searching in Spanish, knowing SciELO exists, noticing that
www.scielo.clreturns 403 and finding the mirror, and not trusting thelanguagefield. Remove any one and the article does not exist. That is how a useful, verified datum stays invisible for a decade.Editor's initials: CRS-M · Date: 31/08/2026

A · Scientific quality: ★★★☆☆ — Experimental study with declared ethical approval and declared sample size, a validated assay with CVs and LOQ, crossover design with washout: a full step 3. It does not rise to 4 because step 4 requires consistency with the ARRIVE 2.0 Essential 10 with randomisation described in the methods, discussed limitations and declared conflicts: here the sequence generation is not described, there is no limitations section and conflicts are not declared.
B · Practical usefulness: ★★★★☆ — A result applicable with reasonable confidence in similar cases, with clear limits and explicit conditions of use: dose, product, concentration, route and site are all present, and the operational conclusion — do not treat i.m. and s.c. as equivalent — is robust to the sample-size uncertainty, because it is a between-route difference measured within the same animals. It does not reach 5 because it does not change a recommended practice: it calls one into question. The cross-constraint (B ≤ A + 2) holds.
C · Teaching value: ★★★★☆ — Cleanly and verifiably teaches three things that are hard to explain in words: why time above MIC rather than peak matters for a beta-lactam; what flip-flop is and how to recognise it (longer t½ by the slower route, with equal bioavailability); and why two routes with the same AUC can need different intervals.
D · Transferability to European camelids: ★★★☆☆ — A study on one camelid species (llama) extended to others in the discussion: step 3 by definition. Supported by the animals being stabled rather than on high pasture, and by altitude not being declared — which makes these the least distant values from European conditions in the whole fascicle. It does not rise because these are six animals from one centre, and the population is not described well enough (sex missing) to judge transferability fully.
24 · Bottom line
You may conclude that: in six healthy adult llamas, ampicillin sodium 20 mg/kg given subcutaneously keeps plasma concentrations above the MIC significantly longer than intramuscularly (4.3 h vs 3.0 h at MIC 4 µg/ml; 8.4 h vs 5.4 h at MIC 0.5 µg/ml), with the same bioavailability — and that the two routes are therefore not interchangeable at the same dosing interval.
You may not conclude that: the intervals the authors propose (i.m. every 6-8 h or 12 h; s.c. every 8-12 h or 12-24 h) are effective. No sick animal was treated, no clinical outcome was measured, and the MICs used do not come from camelid isolates — the authors state they found none. Nor may you derive any withdrawal time from this article: the authors say it must account for the route, and they do not calculate it.
