Specie e soggetti

Species and subjects

Two different objects in a single article, and they must be kept separate.

  1. A narrative review of the literature on copper intoxication in South American camelids: four published case series, all in llamas and alpacas (Junge & Thornburg 1989, four zoo llamas; Weaver 1999, a 9-year-old llama; Carmalt 2001, a 10-year-old alpaca; Fowler 2010, a llama).
  2. A single case report: a 3-year-old female vicuña (Vicugna vicugna), 36 kg, from a zoological garden in northern Germany, admitted on September 1, 2018 to the Clinic for Swine and Small Ruminants in Hannover. In the enclosure had lived since November 2016 another three vicuñas and two alpacas, all sampled on September 6.

It is the first documented case of copper intoxication in a vicuña, according to the authors' statement.

Dichiarati dagli autori: no, né l'uno né l'altra.

Study type and level of evidence

Narrative review combined with a single case report. PubMed classifies the article as a journal article + case report + review; Europe PMC adds research article.

Level of evidence: single case report, framed within a narrative review. Consequence for the reader: the case describes; it does not establish prevalence, does not demonstrate a causal link between the ration and death — and the authors themselves write that the cause of death «remains elusive». The review portion has no declared or reproducible search strategy: it gathers the works the authors were aware of, which is why the threshold values in §Results are all second-hand, as indicated line by line.

Relevant reporting standard: CARE for the case report portion; for the review portion no EQUATOR guideline exists, so SANRA applies (§4.2 of chap. 14 of the KT). Declared by the authors: no, neither one nor the other.

In breve

In brief

A 2024 German article does two things at once: it lines up everything that has been published on chronic copper poisoning in South American camelids, and describes the first case ever documented in a vicuña. The review portion reconstructs the ration numbers — requirement, ceiling, poisoning threshold, copper:molybdenum ratio — and shows that the margin between what is needed and what kills is about threefold, not the wide margin that popular literature attributes to the species. The case portion describes a vicuña that died within 48 hours with hepatic copper 747.4 mg/kg on wet weight against an adequate range of 25-100, serum copper 33.4 µmol/L against 9.9-15.1 µmol/L of the other animals in the same enclosure, icterus of the sclerae, hepatic and renal failure — and without signs of hemolysis. The mineral concentrate in the enclosure contained copper; molybdenum was not listed in either of the two feeds.

Why this study matters

Because it dismantles two statements that circulate in every European camelid farm.

The first is «camelids tolerate copper much better than sheep». The review places South American camelids in an intermediate position between sheep and other ruminants, and its figures give a margin between requirement (9-12 mg/kg DM) and presumed poisoning (> 35 mg/kg DM) of about threefold. Threefold, for a mineral that accumulates in the liver for months without giving signs, is not a wide margin.

The second is «in camelids there is no hemolytic crisis». The review writes «not necessarily», which is not «never» — and reports the counterexample of Fowler 2010, a llama with anorexia, icterus, hemoglobinuria and hemolysis, «more similar to ovine presentations». In the vicuña's case hemolysis was absent, but icterus was present, with total bilirubin at 104.4 µmol/L: the authors interpret it as pre- and intrahepatic icterus, and note that clinical icterus had never before been reported in other South American camelids.

Obiettivo

Dichiarato dagli autori: «This study aims to provide an overview of the existing reports on this species, focusing on clinical, laboratory and pathological findings in a vicuña, alongside laboratory findings from other SACs kept in the same enclosure.»

Objective

Declared by the authors: «This study aims to provide an overview of the existing reports on this species, focusing on clinical, laboratory and pathological findings in a vicuña, alongside laboratory findings from other SACs kept in the same enclosure.»

Clinical question in PICO form (editorial formulation; the design allows it only in part): in a South American camelid with nonspecific signs of hepatic failure [P], which laboratory and anatomopathological findings [I], compared to the species' reference intervals and to enclosure companions [C], allow recognition of chronic copper poisoning [O]?

Disegno dello studio

Study design

Narrative review with no declared search strategy, combined with the clinical, laboratory and anatomopathological description of a single animal, with control sampling of the five cohabitants (three vicuñas, two alpacas) five days later, and analysis of hepatic copper on an alpaca from the same enclosure that died two years later of another cause and on a sheep from the same zoo slaughtered in 2020.

Metodi / Protocollo

Methods / Protocol

  • Hematology with standard manual methods; clinical chemistry and trace elements with atomic absorption spectroscopy (AA Spectrometer M Series, Thermo Electron).
  • Reference intervals: none exist for the vicuña, so those of adult female alpacas from Hengrave Burri 2005 were used — an extrapolation the authors declare.
  • Necropsy with a representative organ spectrum, fixation in 10% neutral buffered formalin, sections of ~3 µm, hematoxylin-eosin staining; additional histochemical stains: rhodanine (copper), Turnbull blue (iron), Fouchet (bile pigments), Heidenhain's azan (collagen fibers).
  • Microbiological examination of the intestine and screening for major toxin genes by mass spectrometry.

The treatment received, with dose, route and species, from the full text. On site, before admission, the vicuña received: continuous intravenous infusion of electrolytes and glucose 1,000 mL (Sterofundin®); metamizole 40 mg/kg IV; dexamethasone, total dose 8 mg IV (2 mL); penicillin-dihydrostreptomycin IM, 5 mL — the dose in mg/kg of the latter is reported in the article as «5 mL = mg/kg BW», i.e. with the value missing in the published text → to be checked against the full text. For the diagnostic laparotomy: ketamine IV for intubation (dose not reported → to be checked against the full text) and maintenance with isoflurane 1-3% in inspired air.

B · The case: the vicuña, and its five cohabitants

Clinical signs: refusal of food for one day, no defecation in the preceding 24 hours, sternal recumbency, apathetic behavior, bruxism, tacky mucous membranes, atony of the gastric compartments and intestine on auscultation, icteric sclerae. Abdominal radiography, ultrasound and fecal parasitological examination: without pathological findings. Diagnostic laparotomy: no macroscopic abnormality, suspicion of ileus excluded; rectum with large, firm but deformable feces, remainder of the intestine nearly empty, motility reduced everywhere, C3 full with deformable content, liver with sharp margins and physiological consistency. The animal was found dead the following day.

Interpretazione degli autori

Attribuita esplicitamente, e distinta dai dati.

Authors' interpretation

Explicitly attributed, and distinguished from the data.

  • The authors do not close the diagnosis: they write that the precise cause of death «remains elusive» due to the nonspecific nature of the signs, and conclude that they are «inclined to interpret the cause of death as copper poisoning, or a combination of factors including copper poisoning, as the most likely cause».
  • The authors hypothesize that the vicuña had an individual predisposition to accumulate copper, or that it had an additional source of intake: they note that it is unusual to find such high values in a single animal if the poisoning depended solely on the ration — and recall that the same happened in Carmalt's 2001 case.
  • The authors hypothesize that the absence of molybdenum from the mineral concentrate unbalanced the ration by increasing copper absorption. It is a hypothesis, not a datum: the vicuña's hepatic molybdenum was 0.06 mg/kg WW, and no reference values for hepatic molybdenum exist for South American camelids.
  • The authors recommend liver biopsy as a more reliable test than blood, and plasma rather than serum for copper determination — «copper may be lost through clots».
  • The authors observe that the vicuña's icterus resembles the ovine hemolytic crisis more than previous cases in South American camelids, and that this supports Fowler's statement that signs in camelids differ from the classic ovine presentation.

Limiti

  • «Unfortunately, due to the poor state of preservation, corresponding necrosis could not be verified» — l'autolisi ha impedito di documentare la necrosi epatocitaria, che è il reperto atteso.
  • «it is not entirely clear how feeding ration data from other camelids can be transferred to vicuñas» — i numeri della razione vengono da alpaca e lama, l'animale era una vigogna.
  • L'eziologia dell'iperbilirubinemia «resta insufficientemente chiarita».

Conflitti di interesse: «The authors declare no competing interests.»

Totale: 9/12.

◆ COMMENTO DELL'ASSOCIAZIONE

Camelid Research Studies APS — redazione CRL · 31/08/2026

Valutazione a stelle

AsseVotoMotivazione

Limitations

Declared by the authors (there is no "limitations" section; these are statements scattered throughout the text, quoted here verbatim or nearly so):

  • «Unfortunately, due to the poor state of preservation, corresponding necrosis could not be verified» — autolysis prevented documentation of hepatocellular necrosis, which is the expected finding.
  • «it is not entirely clear how feeding ration data from other camelids can be transferred to vicuñas» — the ration numbers come from alpacas and llamas, the animal was a vicuña.
  • The role of Cl. perfringens, the petechiae and the renal changes «remains unclear»; the anterior chamber urea may have prerenal causes.
  • The reference intervals used are those of adult female alpacas, because for the vicuña none exist.
  • The etiology of the hyperbilirubinemia «remains insufficiently clarified».

Added by the editorial staff.

  • The review portion has no search strategy: it does not declare databases, strings, dates or languages. The threshold values the clinician will use — 15, 35, 10:1, 16:1, 25-100, 250-400 — are all second-hand, some from manuals (Fowler, Puls) and not from original studies on camelids. This card reports them with their source precisely so that they do not become, through repetition, "the camelids' values."
  • The case does not demonstrate the link. The vicuña had hepatic copper seven times the upper limit of the adequate range, and this is a fact; that it died of it is an inference of the authors, which they themselves qualify.
  • The most disturbing datum is not the patient, it's the cohabitants. The alpaca from the same enclosure, which died two years later of another cause, had hepatic copper 235.3 mg/kg WW — more than double the upper limit of the adequate range, and close to the lower end of Puls's «toxic» range. The sheep from the same zoo was at 123.7. The enclosure was accumulating copper in all the animals, and the only way this was learned is that one died.
  • A single case allows no conclusion about the vicuña as a species. That it is the first case described does not make it representative: it makes it unique.

Funding: «Open Access funding enabled and organized by Projekt DEAL», plus the open access publication fund of the University of Veterinary Medicine Hannover. No research funding is declared, and no industrial sponsor appears.

Conflicts of interest: «The authors declare no competing interests.»

What is missing relative to the reporting standard.

  • CARE, for the case portion: missing a complete dated clinical timeline (the admission day is known and that the animal died «the following day», but not the date signs began nor the duration of exposure to the feed); missing an explicit and reasoned differential diagnosis; missing owner consent (the zoo) and missing any ethical statement. The "follow-up" field does not apply: the animal died.
  • SANRA, for the review portion — compiled by the editorial staff, item by item, from 0 to 2 (§4.2 of chap. 14): importance 2 · aims and questions 1 (the objective is declared but generic) · description of the literature search 0 (absent) · referencing 2 · scientific reasoning 2 · presentation of outcome data 2. Total: 9/12.
  • The quantity of VitaMiral® mineral feed actually administered is not reported, and the authors say so («Fed quantities are not documented»). Without that quantity the ration's copper content cannot be calculated, which is why this card does not write a mg Cu/kg DM value for the vicuña's enclosure: to be checked against the full text — and in the full text it is not there.

◆ ASSOCIATION COMMENTARY

Camelid Research Studies APS — CRL editorial staff · 31/08/2026

«The ration contains 20 mg of copper per kg of dry matter» is not enough to say whether that ration is deficient or toxic: it depends on molybdenum and sulfur.

Whoever reads «the ration contains 20 mg of copper per kg of dry matter» does not yet know anything. The same ration can be deficient if molybdenum and sulfur are high, and toxic if they are low, because molybdenum is what prevents copper from being absorbed. In the vicuña's enclosure molybdenum was not listed in either of the two feeds, and the animal's hepatic molybdenum was 0.06 mg/kg WW — against 1.88 in the sheep from the same zoo. The parameter to ask the feed supplier about is not copper: it is the copper:molybdenum ratio, target < 10:1, alarm > 16:1.

The second thing that must be said aloud is the factor of 3. Requirement 9-12 mg/kg DM, ceiling 15, presumed poisoning above 35. Between the requirement and the poison there is a factor of three. For a mineral that accumulates for months in a silent organ, threefold is not a margin: it is a tightrope. Whoever has written elsewhere that camelids have «a much wider margin» than sheep was comforting, not informing. In the face of a factor of 3, the adverb "much" is not to be used.

The third thing is the most practical, and it is this article's real gift. It is not the patient: it is the enclosure companions. The surviving alpaca, which died two years later from an entirely different cause, had 235.3 mg of copper per kg of liver (wet weight) — more than double the upper limit of the adequate range. No one had suspected it, because it was clinically healthy. This is exactly the long subclinical phase the review describes for sheep. Operational consequence: when a camelid in a group dies with icterus or hepatic failure, the next question is not "what did it die of" — it is "what do the others have in their liver." And a way of answering exists, and is not theoretical: the review recalls that in vivo liver biopsy is performed without significant clinical effects. Blood copper, on the other hand, does not answer that question: Pechová 2018, on 299 alpacas, concludes that neither plasma nor whole blood reflect intake.

Where we do not agree with the authors — or rather, where we ask for more. The work is honest in not closing the diagnosis, and we appreciate its caution. But the review portion, which is the one that will be cited a thousand times, does not declare how it was built, and its threshold values come from two manuals from the 1990s and 2000s. In a year's time someone will write «according to Marahrens 2024 the toxicity threshold in the camelid ration is 35 mg/kg», and it will not be true: according to Marahrens 2024, Puls 1994 says so. The chain of second-hand citations is the mechanism by which a datum without foundation becomes a fact, and in a small and highly self-citing field like this one it is the main risk to our own collection.

What is missing, and what an association can do. Missing is an elementary study within reach of anyone with access to a slaughterhouse or a necropsy service: the hepatic copper of apparently healthy European South American camelids, with the ration documented and molybdenum measured alongside. Today the only interval we use — 25-100 mg/kg WW — comes from a 1994 manual and not from a European population. As long as this remains so, every hepatic copper report is being compared against a number none of us has verified.

Star rating

AxisScoreRationale
A · Scientific quality★★★☆☆The case report is well documented for the laboratory and histochemical portion (rhodanine, azan, negative controls for iron and bile) and includes a real comparison group — the five sampled cohabitants. But it lacks the dated timeline, lacks the explicit differential, lacks any ethical statement, and necrosis is not verifiable due to autolysis. For the review portion, SANRA 9/12 with the "description of the literature search" item at 0: the CRL conversion places 9-12 at ★★★★, but the single case pulls it down, and the declared ceiling for narrative reviews remains ★★★★. Three stars is the honest average of a work strong on findings and weak on the method of selecting the literature.
B · Clinical utility★★★★☆Delivers, applicable tomorrow morning: the ration threshold values with their source, the Cu:Mo ratio as the parameter to ask about, the WW/DM warning that makes any report legible, liver biopsy as a practicable tool, and sampling of the cohabitants as the conduct to follow in the face of an index case. It does not reach five because it does not change a recommendation: it gives them, for the first time, a traceable foundation, and none of the thresholds is measured here.
C · Educational value★★★★★It is the first-choice reference on the subject in the species, and it teaches two things that go beyond copper: that a trace-element value without its unit and its matrix is not comparable, and that a mineral is read in relation to its antagonists, not alone. Anyone who has to learn about copper in camelids starts here.
D · Species relevance★★★☆☆The case is in a vicuña, the species almost none of our readers keep; the review is on llamas and alpacas, which are ours. European context (German zoo), therefore transferable in terms of climate and management. It does not rise to four because the original part — the only datum produced by this work — concerns a single animal of a species different from the reader's, and the CRL grid is explicit: work on one species is not automatically work on another.

No overall score, by design. A = 3 and C = 5 are not a contradiction: an article can be the best available teacher on a topic and remain, as evidence, a single case with a non-reproducible review around it.

Bottom line

It can be concluded that in this vicuña (a single one, 3-year-old female, 36 kg) the hepatic copper was 747.4 mg/kg on wet weight against an adequate range of 25-100, with serum copper 33.4 µmol/L against 9.9-15.1 µmol/L of the five cohabitants, icterus (total bilirubin 104.4 µmol/L) and rhodanine-positive pigment in hepatocytes without other evidence of hemolysis; and that the accumulation did not concern the patient alone, because the alpaca from the same enclosure, which died two years later of another cause and was clinically healthy until then, had 235.3 mg/kg WW of hepatic copper.

It cannot be concluded that copper poisoning is the cause of death: it is the authors themselves who write that it «remains elusive», and autolysis prevented documentation of the expected hepatocellular necrosis. It cannot be concluded that the thresholds were measured here: requirement 9-12 mg/kg DM, ceiling < 15, presumed poisoning > 35, CMR < 10:1 and > 16:1, hepatic copper 25-100 and 250-400 are all second-hand citations from manuals (Fowler, Puls 1994), and the review portion declares no search strategy (SANRA 9/12, with the "literature search" item at 0). The copper content of that enclosure's ration cannot be calculated: the quantity of VitaMiral® administered is not documented, and the card writes to be checked against the full text instead of a mg Cu/kg DM value. The case cannot be read as a measure of the vicuña as a species — the comparison values are of adult female alpacas, because for the vicuña none exist — nor can a therapeutic regimen be derived from it: metamizole 40 mg/kg IV and dexamethasone 8 mg total IV are what one animal that subsequently died received, penicillin-dihydrostreptomycin IM is reported as «5 mL = mg/kg BW» with the number missing, and the ketamine dose does not appear at all.

Messaggi da portare a casa

Take-home messages

  1. The margin between requirement and copper poisoning in South American camelids is about 3× (9-12 mg/kg DM against > 35 mg/kg DM), not the wide margin one hears about. And all these numbers are second-hand, not measured.
  2. A copper value in the ration, alone, is not decidable. The Cu:Mo ratio is needed — target < 10:1, alarm > 16:1. In the vicuña's case molybdenum was not listed in either of the two mineral feeds.
  3. Hepatic copper must always be read with its unit. 747.4 mg/kg wet weight and 640.5 mg/kg dry matter are not comparable: the authors estimate a factor of 4-5 between the two scales.
  4. The absence of hemolysis does not exclude copper poisoning. The vicuña had icterus (total bilirubin 104.4 µmol/L) without other evidence of hemolysis; and in camelids the dominant presentation is hepatic failure, not hemolytic crisis — «not necessarily», however, does not mean «never».
  5. Blood copper does not measure intake; the liver does. And in the face of an index case, the enclosure companions must be examined: the surviving alpaca had 235.3 mg Cu/kg WW in its liver and was clinically healthy.

Card CRL-B-01 · version 1.0 · 31/08/2026 · summarizes a single source, not a body of literature. It has not been searched, and therefore it is not excluded, whether after February 2024 there are new cases of copper intoxication in South American camelids, or original studies on hepatic copper intervals in the species in Europe. The vicuña's hepatic copper value is reported as 747.4 mg/kg WW, which is the value in Table 4 of the article; in the Discussion of the same article 747.3 appears, and in chapter 04 of the Camelidi KT 747.3 is reported: this card adopts the value from the data table.
