Specie e soggetti
|---|---|---|---|---|
| 1 | madre | Suri | 62× | 100% |
| 1 | padre | Huacaya | 42× | 99,9% |
| 1 | figlio/a | Suri | 50× | 99,9% |
| 2 | madre | Huacaya | 37× | 99,9% |
| 2 | padre | Suri | 51× | 99,8% |
| 2 | figlio/a | Huacaya | 43× | 99,8% |
Species and subjects
Three species, and the distinction is the heart of the design.
Alpaca (Vicugna pacos): n = 6. Not just any six animals: two test-cross pairs with their offspring, i.e. two complete families of three individuals.
| Family | Role | Phenotype | Sequencing coverage | Genotyping rate |
|---|---|---|---|---|
| 1 | dam | Suri | 62× | 100% |
| 1 | sire | Huacaya | 42× | 99.9% |
| 1 | offspring | Suri | 50× | 99.9% |
| 2 | dam | Huacaya | 37× | 99.9% |
| 2 | sire | Suri | 51× | 99.8% |
| 2 | offspring | Huacaya | 43× | 99.8% |
Origin of the alpacas: the INIA (Instituto Nacional de Innovación Agraria, Peru) experimental station at Quimsachata. The four parents belong to pure-line populations (suri × suri and huacaya × huacaya) selected for twenty years, with a generation interval of about 4-5 years.
Vicuña (Vicugna vicugna): n = 6. Guanaco (Lama guanicoe): n = 6. Not sequenced for this study: WGS data reused from the public NCBI SRA project PRJNA612032, with coverage 16-21× and genotyping rate 99.5-99.7%.
The role of the twelve wild individuals is an assumption, and it should be said: they are used as controls assuming that the variants linked to the suri phenotype do not exist in their genome. The assumption is plausible — suri does not exist in nature — but it is an assumption, not a datum.
One guanaco is an exception and should be flagged: sample SRR11905252 has coverage 15× and genotyping rate 93.9%, the only one below 99%.
The material. Skin biopsies obtained in 2008 according to the procedure described in an earlier paper by the same group, DNA extracted with a commercial kit, library prepared at Genomix4Life (Salerno) with Illumina DNA Prep, sequencing 150 bp paired-end on Illumina NovaSeq 6000. Sixteen years between collection and publication: it is a sample bank, and it is exactly what chapter 13 of the KT means when it says that Camerino has «a school».
Study type and level of evidence
Discovery genomic study, not comparative in an epidemiological sense: segregation analysis of variants in two informative families, filtered against wild control species, followed by bioinformatic prediction of the protein effect.
Level of evidence: it is a category that clinical hierarchies do not cover well. In the vocabulary of §1.3 of chapter 14 of the KT it sits between an experimental study with ethical approval and a stated sample size and a case series: the sample is minuscule (six alpacas) and the result is a segregation association, i.e. the finding that a variant is present in all three suri and in none of the other fifteen individuals examined.
Consequence for the reader, and it is the most important line of this card: with six alpacas and twelve wild individuals, a variant that segregates perfectly with the phenotype may be the cause, or may be in linkage with the cause, or may be a coincidence. The authors know it and write it: the title says suggests, the conclusion says may arise, and the limitations section explicitly calls for validation on a larger sample. Anyone turning this result into a commercial genetic test for Suri would be selling a hypothesis.
Relevant reporting standard: ARRIVE 2.0 for the in vivo component (bioptic sampling on animals). Declared by the authors: yes — the ethics statement affirms that the protocols were approved by the Animal Ethics Committee of the University of Camerino and are «in accordance with ARRIVE guidelines». This is a statement of compliance, not a deposited checklist: the difference matters, and §3.3 of chapter 14 recalls it.
In breve
In brief
The alpaca fleece exists in two forms: Huacaya, with compact, soft, strongly crimped fibres; Suri, with long, straight, poorly crimped, lustrous fibres with fewer cuticular scales. That the trait was heritable and that Suri was dominant was known from segregation studies; which gene was responsible was not. This work sequenced the whole genome of two suri × huacaya test-cross families and searched for heterozygous variants present in all suri and absent in the huacaya, in the six vicuñas and in the six guanacos. This yielded eight high-impact variants at eight loci and 153 missense variants at 136 loci. Among these, the most plausible on hair-follicle biology grounds is a premature stop codon in the TRPV3 gene, on chromosome 16: substitution c.1423G > T, p.Glu475\*, which truncates the protein, causing it to lose 290 amino acids — i.e. five of the six transmembrane domains and the pore loop needed for ion transport. TRPV3 is a thermosensitive channel expressed in keratinocytes, the inner root sheath and the hair shaft, and in mice regulates entry into catagen. The authors propose that the loss of function delays catagen, prolongs anagen and thereby produces the longer Suri fibre.
Why this study matters
Because it closes — hypothetically — a decades-old question with a mechanism, and because the proposed mechanism explains more than one trait at once: fibre length, the lower number of cuticular scales, and — if the lethal-homozygote hypothesis holds — even the rarity of the phenotype.
And because it hands a breeder a piece of information no manual gives: Suri, in all likelihood, is not a variety. It is a heterozygote for a loss-of-function mutation in a gene that also regulates cold tolerance. If true, the fragility of Suri that Andean breeders talk about is not folklore: it is the same mutation, read from the other side.
Obiettivo
Dichiarato dagli autori: «the aim of this study was to identify the gene variants responsible of the suri and the huacaya phenotypes, through a WGS analysis».
Objective
Declared by the authors: «the aim of this study was to identify the gene variants responsible of the suri and the huacaya phenotypes, through a WGS analysis».
Question in PICO form (editorial formulation; the design only partially allows it): in Suri-phenotype alpaca (Vicugna pacos) [P], is there a high-impact gene variant [I] that segregates with the phenotype relative to Huacaya alpaca and the two ancestral wild species [C], and that is biologically plausible for hair-follicle biology [O]?
Disegno dello studio
- si tengono le varianti eterozigoti presenti in tutti e tre i campioni suri;
Study design
Family segregation analysis on whole genome sequencing data, under a dominant inheritance model assumed a priori (because prior segregation literature suggested it). The filter is two-staged:
- keep heterozygous variants present in all three suri samples;
- discard those present in the huacaya, the vicuñas and the guanacos.
Then, a non-automatic third stage: a literature search on PubMed and Google with the algorithm (gene name) AND (hair) for each remaining locus, to choose the most plausible candidate.
This third stage is the weak point and the interesting point at once. The candidate was not chosen by a statistic: it was chosen by a biological judgment informed by the literature. It is a legitimate and very common way to proceed, and it is also how the most convincing false positives are produced — because the gene with the most hair literature will always be the one that looks most convincing.
Metodi / Protocollo
Methods / Protocol
- Reference and alignment: reads mapped to the alpaca reference genome VicPac3.1 with BWA-MEM; quality control with FastQC, adapter trimming with Trimmomatic.
- Variant calling: GATK v3.4, HaplotypeCaller approach, joint variant calling on the 18 samples together.
- Annotation and filtering: SNPeff for annotation, SNPsift for filtering the heterozygotes in the three suri.
- Manual verification: the BAMs were opened in the Integrative Genomics Viewer and the variants visually inspected to verify their fidelity. This is good practice and should be flagged: many «high-impact» variants from automated filters are alignment artefacts, and looking at them is the only way to notice this.
- Prediction of protein domains: InterProScan.
- Structural modelling: SWISS-MODEL, using as template the human and mouse TRPV3 protein sequences.
Risultati principali

| Famiglia | Ruolo | Fenotipo | Genotipo TRPV3 c.1423G > T |
|---|---|---|---|
| 1 | madre | Suri | G/T |
| 1 | padre | Huacaya | G/G |
| 1 | figlio/a | Suri | G/T |
| 2 | madre | Huacaya | G/G |
| 2 | padre | Suri | G/T |
| 2 | figlio/a | Huacaya | G/G |
- TRPV3 è espresso nei cheratinociti, nella guaina epiteliale interna e nel fusto del pelo, ed è un canale TRP termosensibile;
- agisce da attivatore del catagen, inibendo l'allungamento del fusto e la proliferazione dei cheratinociti della matrice e inducendo una regressione catagen prematura guidata dall'apoptosi;
- nel topo, le mutazioni di TRPV3 alterano la struttura del pelo compromettendo la formazione del canale e del fusto; i mutanti mostrano proliferazione aumentata nella guaina esterna, ciclo accelerato, riduzione delle cellule staminali del follicolo, miniaturizzazione dei follicoli e riduzione del diametro del pelo; i knockout hanno mantello ondulato e vibrisse ricce;
- nell'uomo, mutazioni di TRPV3 sono coinvolte nella sindrome di Olmsted (OMIM #614594);

B · What survived the filter: eight variants of potentially high phenotypic impact at eight loci, plus 153 missense variants distributed across 136 different loci.
The eight high-impact ones, with huacaya/vicuña/guanaco genotype against suri genotype:
| Locus | Chromosome | Control genotype | Suri genotype | Variant | Predicted effect | Type |
|---|---|---|---|---|---|---|
| LOC116280269 (uncharacterized) | 4 | CA/CA | CA/C | c.717delA | p.Lys239fs | frameshift |

| PLIN5 | 22 | G/G | G/GT | c.912dupT | p.Gly305fs | frameshift |
Three of the eight are premature stop codons, not just TRPV3. The choice of TRPV3 among the three is bibliographic, not statistical. The authors state this.
C · Segregation in the two families — this is the table that makes the result non-trivial:
| Family | Role | Phenotype | TRPV3 genotype c.1423G > T |
|---|---|---|---|
| 1 | dam | Suri | G/T |
| 1 | sire | Huacaya | G/G |
| 1 | offspring | Suri | G/T |
| 2 | dam | Huacaya | G/G |
| 2 | sire | Suri | G/T |
| 2 | offspring | Huacaya | G/G |
Perfect segregation, autosomal dominant, across two independent crosses. These are six informative meioses in total: the pattern is the expected one, but the probability that such a pattern would be observed by chance across only two transmissions is not negligible, and this is exactly what a validation cohort would serve to exclude.
D · The predicted effect on the protein. The ion-transport domain of alpaca TRPV3 is predicted starting from amino-acid position 416 and comprises six transmembrane helices, three extracellular and three cytoplasmic regions. The stop codon falls at position 475, i.e. in the second transmembrane helix: the truncated protein loses 290 amino acids, five transmembrane domains and the pore loop essential for ion transport.
Quality of the structural models built with SWISS-MODEL: wild-type GMQE 0.75, sequence identity 89.5%, coverage 1.00 (amino-acid range 118-731); mutant GMQE 0.73, identity 85.7%, coverage 1.00 (range 85-473). These are models predicted on human and mouse templates, not experimentally resolved alpaca structures.
Authors' interpretation (attributed, not merged with the data)
The authors' reasoning, reported as theirs:
- TRPV3 is expressed in keratinocytes, the inner root sheath and the hair shaft, and is a thermosensitive TRP channel;
- it acts as a catagen activator, inhibiting shaft elongation and matrix keratinocyte proliferation and inducing premature apoptosis-driven catagen regression;
- in the mouse, TRPV3 mutations alter hair structure by impairing channel and shaft formation; mutants show increased proliferation in the outer sheath, an accelerated cycle, reduced follicle stem cells, follicle miniaturization and reduced hair diameter; knockouts have a wavy coat and curly whiskers;
- in humans, TRPV3 mutations are involved in Olmsted syndrome (OMIM #614594);
- the same mutation has been reported as positively selected in the woolly mammoth lineage, and mutations of the gene have been found in high-altitude sheep and bighorn sheep, in the American beaver, in the colugo, in the lesser Egyptian jerboa and in the wild Bactrian camel (Camelus ferus) — this last group in a hypothesized association with tolerance to high temperatures.
Hence the central hypothesis, which is the authors' and not a datum: loss of function delays entry into catagen, anagen persists, the shaft keeps elongating — and this would give the long Suri fibre, together with the lower number of cuticular scales due to alteration of the inner root sheath.
A second hypothesis, even more speculative and declared as such by the authors: that the mutated TRPV3 homozygote is embryonically lethal, as has been proposed for the classic grey coat colour of alpaca. If true, this would explain why Suri remains rare — the authors cite an estimate according to which the suri phenotype represents about 7% of the South American alpaca population (a figure taken from a third-party source cited in the article, not measured in this study).
A third, concerning welfare: since TRPV3 also regulates thermal adaptation and cold tolerance, that function might be compromised in Suri. The authors explicitly write that there are «casual reports of breeders» — anecdotal reports from breeders — of greater weakness in Suri regarding growth, disease and mortality. These are anecdotes, not data, and the article presents them as such.
Implicazioni pratiche
- Nessuna, oggi, in clinica.
Practical implications
- None, today, in the clinic. There is no test, no recommendation, no dose. Anyone looking in this card for something to do tomorrow morning will not find it.
- One, however, in the examination room, and it is about language. If the hypothesis holds, «Suri» and «Huacaya» are not two breeds: they are two genotypes at one locus, heterozygous and reference homozygous. A breeder who crosses suri × suri is, under this hypothesis, generating an expected proportion of homozygotes — which may not be born. If the lethal-homozygote hypothesis were confirmed, the suri × suri cross would become a breeding choice with an expected cost. At present it is not confirmed, and this card recommends nothing on that basis.
- A third, for those designing research. Validation is within reach and not expensive: a targeted genotyping of c.1423G > T on a few hundred phenotyped alpacas and a genotype count in the offspring of suri × suri crosses would suffice. It is a publishable piece of work, with low risk of a null result, and no one has done it yet.
Limiti
- il disegno ha identificato le varianti che segregano col fenotipo suri, ma «i risultati devono essere validati su un campione più ampio»;
- il compromesso funzionale della proteina TRPV3 nel suri è suggerito dai risultati e andrebbe confermato con approcci trascrittomici e proteomici;
- n = 6 alpaca, e sei sole trasmissioni meiotiche informative.
Conflitti di interesse: dichiarati assenti — «The authors declare no competing interests.»
Limitations
Limitations declared by the authors — the article has an explicit «Limitations» section, and it is quoted here, not summarized:
- the design identified the variants that segregate with the suri phenotype, but «the results need to be validated in a larger sample size»;
- the functional compromise of the TRPV3 protein in suri is suggested by the results and should be confirmed with transcriptomic and proteomic approaches;
- the estimate of the age of the mutation — which would indicate when Suri was selected starting from Huacaya — requires recombination maps that do not yet exist for these species.
Limitations added by the editors:
- n = 6 alpacas, and only six informative meiotic transmissions. It is a discovery-study sample, and this should be said every time the result is cited.
- The dominant model is assumed a priori, not tested. If Suri were not dominant, the filter would have discarded the true variant.
- The choice of the candidate among the eight high-impact variants is bibliographic. Three of the eight are stop codons; ARPC4 and HELZ2 were not excluded by a datum, they were discarded because they have less literature on hair. The authors declare this; the consequence is that the result is not a statistical test but an informed judgment.
- The twelve wild individuals are a control by assumption. Assuming that the suri variant does not exist in vicuña and guanaco is reasonable but not independently verified.
- The protein model is predicted, on human and mouse templates, with GMQE 0.73-0.75. It is not an experimental alpaca structure.
- No fibre measurement was taken on these six animals. The phenotype is categorical — suri or huacaya — and not a single micron is measured in the article. The link between the variant and shaft length therefore remains purely inferential: no one measured the staple length of the three suri carriers.
- The biopsies are from 2008, sequenced and published in 2024. This is not a flaw, but it means the animals are no longer available for finer phenotyping.
- The reports on Suri fragility are breeders' anecdotes reported by the authors as such. They are not a study datum and should not be cited as if they were.
Funding: «Not applicable» — verbatim, in the article's Funding section. That is: no dedicated funding declared. This is not a source of conflict; but it is information about the state of the field, and it should be read together with another fact: this research was done on samples preserved for sixteen years, without dedicated funds, by a group that has carried it forward for twenty years.
Conflicts of interest: declared absent — «The authors declare no competing interests.»
Ethical approval: declared — skin biopsies obtained in 2008 according to the guidelines of the Animal Ethics Committee of the University of Camerino, protocols approved by the same committee, «in accordance with ARRIVE guidelines».
Data availability: declared and verifiable — VCF files in the EVA repository (project PRJEB61878), FASTQ files in NCBI SRA (project PRJNA1020284), wild samples in SRA (PRJNA612032). This is exemplary data-transparency reporting and should be acknowledged.
◆ COMMENTO DELL'ASSOCIAZIONE
Camelid Research Studies APS — redazione CRL · 31/08/2026
La conclusione dice may arise.
(nome del gene) AND (hair).
Valutazione a stelle
What is missing relative to the reporting standard (ARRIVE 2.0, applied retroactively)
- There is no completed ARRIVE checklist: there is a statement of compliance, which is a different thing.
- Sample-size justification is missing (Essential 10, item 2): six animals are the number available, not the number calculated. It should be noted that for a test-cross design the justification is logical rather than statistical, but it should be written down.
- Description of animal characteristics is missing beyond phenotype and family: age, sex, weight, condition at the time of biopsy are not reported.
- Any exclusion criteria are missing, not declared.
- A quantitative measure of the phenotype is missing (staple length, diameter, cuticular scale count) on the sequenced animals.
- Present and well done: ethics statement, conflict-of-interest statement, availability of raw data with project identifiers, manual inspection of variants, explicit and honest limitations section.
◆ ASSOCIATION COMMENT
Camelid Research Studies APS — CRL editorial team · 31/08/2026
A group at the University of Camerino, with ENEA in Rome, has published the first serious mechanistic hypothesis of what Suri is — on samples from 2008 and without dedicated funding.
The first. A group at the University of Camerino, with ENEA in Rome, has published in BMC Genomics the first serious mechanistic hypothesis of what Suri is. It did so on samples collected in 2008, without dedicated funding — «Funding: Not applicable», written in black and white — and with six alpacas. Chapter 13 of the KT says it plainly: Camerino is the only Italian group working on fibre, i.e. on the sector's only real economic value chain, and it has a continuity of over twenty years. An Italian veterinarian following camelids has, two hours' drive from almost anywhere, the group that found this gene. Almost no one knows it, and that is why this card exists.
The second, and it should be said with equal force. The article's title says suggests. The conclusion says may arise. The limitations section calls for validation on a larger sample, calls for transcriptomic and proteomic confirmation, and admits that the age of the mutation cannot be estimated because recombination maps are lacking. Anyone who, at a fair or in a catalogue, presented «the Suri gene» as an established fact, or offered a genetic test on this basis, would be going beyond what the authors wrote — and the authors are, in this respect, far more cautious than how they will be cited.
The technical point we want to leave with the reader, because it holds in general. The automated filter left standing eight high-impact variants, of which three premature stop codons: TRPV3, ARPC4 and HELZ2. Statistics did not choose among the three. A literature search with the algorithm
(gene name) AND (hair)did. It is a legitimate procedure, declared by the authors, and used everywhere in genomics. And it is also how the most persuasive false positive there is gets built, because the gene with the most hair literature will always seem the most convincing, whether it is the right one or not. This is not a criticism of the authors: it is the reason why the validation they themselves call for is not a formality.What would change if the hypothesis were confirmed. Three things, and none of them small. (a) «Suri» would stop being a breed and become a heterozygous genotype at one locus. (b) If the lethal-homozygote hypothesis also held, the suri × suri cross would have an expected and calculable reproductive cost, and this changes a real breeding decision. (c) The greater cold fragility of Suri — today an anecdote from Andean breeders — would have a mechanism: TRPV3 is also the cold-tolerance channel, and the same mutations have been found in the woolly mammoth and in the wild Bactrian camel. For a breeder in northern Italy, who keeps animals outdoors in winter, this would not be an academic detail.
What we do not conclude. We do not conclude that TRPV3 causes the Suri phenotype: segregation across six alpacas and two transmissions does not demonstrate it. We do not conclude that the homozygote is lethal: no one has counted genotypes in offspring. We do not conclude that Suri is more fragile: breeders' anecdotes remain anecdotes until someone measures mortality and growth by genotype. We write nothing about the length or diameter of the fibre of the three sequenced suri, because in this article not a single micron was measured.
Star rating
Axis Score Rationale A · Scientific quality ★★★☆☆ Experimental study with declared ethical approval and a stated sample size, raw data deposited with verifiable project identifiers, manual inspection of variants in IGV, explicit and honest limitations section, declared conflicts. The fourth star would require consistency with ARRIVE 2.0's Essential 10: missing are sample-size justification, animal characteristics, exclusion criteria and a quantitative measure of the phenotype. B · Clinical utility ★★☆☆☆ Suggests a genetic hypothesis to be verified, not to be applied: no test available, no sustainable breeding recommendation today. The cross-constraint (B ≤ A + 2) is met with margin. No dose, route or protocol is involved. C · Educational value ★★★★☆ Course material on three fronts: the hair-follicle cycle (anagen/catagen/telogen) explained through a loss of function; the difference between a candidate gene and a demonstrated cause, with the article's own title making the point; and pleiotropy — the same channel regulates the hair shaft, thermosensation and cold tolerance, which links fibre genetics and welfare in a single sentence. D · Species relevance ★★★☆☆ A study on Vicugna pacos and its two wild progenitors, on a trait present in every European herd, conducted by an Italian group. It does not reach four stars because it is neither large nor multicentric — six alpacas from a single Peruvian experimental station — and the population is not described enough to assess the transferability of the allele frequency to European herds. No overall score, by design: A = 3 and C = 4 are not a contradiction. A study with six animals can teach more than one with six hundred, and this one does.
Bottom line. It can be concluded that in two suri × huacaya test-cross families a nonsense variant of TRPV3 (c.1423G > T, p.Glu475*, chromosome 16, loss of 290 amino acids) segregates in an autosomal dominant manner with the Suri phenotype, is absent in six vicuñas and six guanacos, and is biologically plausible because TRPV3 regulates entry into catagen of the hair follicle. It cannot be concluded that TRPV3 causes the Suri phenotype, nor that the homozygote is lethal, nor that Suri is more fragile to cold: the first remains an association across six animals, the other two are hypotheses explicitly presented as such by the authors.
Sei trasmissioni informative in tutto.
Take-home messages
- Suri has a candidate gene: TRPV3, mutation c.1423G > T, p.Glu475\*, chromosome 16. Candidate, not cause. The article's title says suggests.
- The sample is six alpacas, two test-cross families, plus six vicuñas and six guanacos as wild controls. Six informative transmissions in total.
- The proposed mechanism is a delay in catagen: if TRPV3 does not work, anagen persists and the shaft keeps elongating. This is the hypothesis that explains both longer fibre and fewer cuticular scales at once.
- TRPV3 also regulates cold tolerance. If the hypothesis holds, the winter fragility that Andean breeders attribute to Suri would have a mechanism. Today it is an anecdote reported as such.
- This was done by an Italian group, in Camerino, without dedicated funding, on biopsies from 2008. It is the only Italian group working on camelid fibre.

Card CRL-E-02 · version 1.0 · 31/08/2026 · summarizes a single source, not a body of literature. It has not been searched, and therefore not excluded, whether validation studies subsequent to February 2024 on the TRPV3 c.1423G > T variant in alpaca exist; the Crossref and Europe PMC query of 31/08/2026 on this article's metadata returned none, but a systematic search of forward citations has not been conducted — which is precisely the search to carry out before updating this card.
