Complete list, eight authors, in the order and form of the Europe PMC/PubMed record (PMID 8502296).
⚠️ Two surnames diverge between the two records, and this sheet does not smooth them over. The Crossref record for the same DOI registers the fifth and eighth authors as «Hammers C.» and «Hammers R.», with a double m. The Europe PMC/PubMed record registers «Hamers C» and «Hamers R». The correct form is Hamers: Raymond Hamers is the head of the Vrije Universiteit Brussel group to which the article is attributed throughout the subsequent literature, and Hamers-Casterman is the first author's double surname, which contains the same surname.
This is not a curiosity. In the open-access full text of Evers et al. 2025 (sheet G-02 of this issue), reference 16 of the bibliography reads: «Hamers-Casterman C, Atarhouch T, Muyldermans S, Robinson G, Hammers C, Songa EB, Bendahman N, Hammers R.» The typo from a bibliographic record has made its way, thirty-two years later, into the field's reference review, published in a serious peer-reviewed journal. This is exactly the mechanism described in §9.3 of chapter 14 of this KT — the one that manufactures nonexistent authors — caught in the act. Anyone citing this article should copy it from PubMed, not from Crossref.
Corresponding author's affiliation: the corresponding author is not identified in the accessible records → [TO BE CHECKED AGAINST THE FULL TEXT]. Europe PMC's affiliation field reports the first author's affiliation: Vrije Universiteit Brussel, Instituut voor Moleculaire Biologie, Belgium. This is Hamers-Casterman's affiliation, not necessarily that of whoever handled the correspondence.

Specie e soggetti
Species and subjects
Dromedary — Camelus dromedarius, named in full in the abstract.
Sample size is not stated in the abstract → [TO BE CHECKED AGAINST THE FULL TEXT]. It is not known how many animals, of what age, of what sex, in what husbandry context, nor where the serum came from. Being a 1993 Letter to Nature on serum material, it is possible that the number of animals does not even appear in the full text.
No other camelid was studied in this work. The authors write that heavy-chain antibodies are «a feature of all camelids» — but this, in the abstract, is an extension, not a measurement repeated on llama, alpaca, guanaco or vicuña. This distinction underpins the entire Relevance to the species field of this sheet.
Domanda clinica (PICO)
Clinical question (PICO)
The design does not allow for a PICO, and faking one would be a disguise. The article has no patients, no intervention, no comparison and no clinical outcome. The real question is structural: does the dromedary's serum contain immunoglobulin material that binds antigen while lacking light chains? The PICO format does not apply, and this field says so instead of leaving a blank (chapter 14, §10).
Study type and level of evidence
Experimental protein-biochemistry study on serum. PubMed classifies it as Journal Article · Research Support, Non-U.S. Gov't. The unit of analysis is not the animal, it is the molecule.
Level of evidence: basic, non-clinical research; no comparison group, no outcome in a live animal. Consequence for the reader: it establishes what is present in a dromedary's serum; it does not establish anything about how a camelid gets sick, is treated or is vaccinated, and anyone citing it for a clinical decision is using it out of scope. In the evidence hierarchy it ranks low; in the history of the field it is ground zero. Chapter 14 (§1.3) asks that both sentences be written, and this sheet writes both.
Relevant reporting standard: no EQUATOR checklist covers this design. CONSORT, REFLECT, PRISMA 2020, STROBE and CARE are all out of scope. ARRIVE 2.0 would be the relevant checklist if the serum had been collected for research purposes from live animals — which is probable but not verifiable without the full text → [TO BE CHECKED AGAINST THE FULL TEXT]. It should be said for fairness that ARRIVE dates from 2010 and ARRIVE 2.0 from 2020: demanding it of a 1993 article would be an anachronistic reproach. The checklist serves here to say what a reader today does not find, not to give a moral grade to a thirty-three-year-old article. Declared by the authors: not verifiable.
In breve
L'ultima frase dell'abstract è quella che ha cambiato la vita a un certo numero di alpaca europee: «Heavy-chain IgGs are a feature of all camelids» — e, subito dopo, «These findings open new perspectives in the engineering of antibodies».
In brief
An antibody, in all mammals studied up to 1993, is made of two heavy chains and two light chains; the antigen-binding site arises from the pairing of a variable domain of the heavy chain (VH) with one of the light chain (VL), and the combinatorics between the two repertoires is one of the main sources of antibody diversity. Heavy-chain-only antibodies were known as a pathology — heavy-chain disease — not as a normal occurrence.
This article reports that the dromedary's serum contains a considerable amount («considerable amounts») of IgG-like material of molecular mass 100 kDa, composed of heavy-chain-only dimers, devoid of light chains, which nonetheless has an extensive antigen-binding repertoire. And it adds the structural finding that explains why those molecules hold together: the camel's heavy-chain IgGs lack the CH1 domain, which — the authors write — «in a class of IgG might be structurally replaced by an extended hinge».
The last sentence of the abstract is the one that changed the life of a certain number of European alpacas: «Heavy-chain IgGs are a feature of all camelids» — and, right after, «These findings open new perspectives in the engineering of antibodies».
Why this study matters
Because the isolated variable domain of that antibody — the VHH, today commercially known as nanobody — is the smallest natural antibody fragment that binds antigen on its own, and from it came five approved drugs (sheet G-02), a number of laboratory reagents, and the reason why herds of alpacas are kept for research in Europe.
And because an association concerned with camelids has a direct interest in knowing where all this comes from: not from an industrial project, but from an observation made on a serum, in a laboratory that was studying trypanosomiasis. Anyone wishing to know what it means today, for the animal, to stand upstream of that supply chain will find the numbers in sheet G-02 and the Italian legal framework in chapter 08 §11.2 of this KT.
Obiettivo
Objective
To verify the presence, in the serum of Camelus dromedarius, of immunoglobulin material devoid of light chains, and to characterize its composition and antigen-binding capacity.
Metodi / Protocollo
Methods / Protocol
Not readable. The article is behind a paywall and the abstract does not describe the method. Not verifiable: the number and provenance of the animals, the separation and analysis techniques used, how the antigen-binding repertoire was assayed, the controls, nor any ethical statement → [TO BE CHECKED AGAINST THE FULL TEXT].
This sheet does not assert them, and none of its conclusions rest on them.
Risultati (dati)
Ciò che l'abstract riporta, e nient'altro:
- nel siero del dromedario è presente materiale IgG-simile di massa molecolare 100 kDa, in quantità definita «considerevole». Nessuna percentuale, nessun denominatore, nessun intervallo → da verificare sul testo pieno;
- quelle molecole sono dimeri di catena pesante e sono prive di catene leggere;
- hanno un repertorio di legame antigenico esteso. L'estensione non è quantificata nell'abstract → da verificare sul testo pieno;
Results (data)
What the abstract reports, and nothing else:
- the dromedary's serum contains IgG-like material of molecular mass 100 kDa, in an amount described as «considerable». No percentage, no denominator, no range →
[TO BE CHECKED AGAINST THE FULL TEXT]; - those molecules are heavy-chain dimers and are devoid of light chains;
- they have an extensive antigen-binding repertoire. The extent is not quantified in the abstract →
[TO BE CHECKED AGAINST THE FULL TEXT]; - the camel's heavy-chain IgGs lack CH1.
The quantitative figure everyone attributes to this article is not in this article. The figure that circulates — «about 50 % of a camelid's IgG are heavy-chain antibodies» — comes from a different work, fourteen years later: Maass DR, Sepulveda J, Pernthaner A, Shoemaker CB, «Alpaca (Lama pacos) as a convenient source of recombinant camelid heavy chain antibodies (VHHs)», J Immunol Methods 2007;324(1-2):13–25, DOI 10.1016/j.jim.2007.04.008, PMID 17568607, whose abstract states verbatim: «Alpaca sera IgG consists of about 50 % HCAbs, mostly of the short-hinge variety». Metadata and abstract verified on Crossref and Europe PMC on 31/08/2026. The 50 % is for the alpaca and from 2007, not the dromedary and 1993, and is one of the reasons why the alpaca became the laboratory animal of choice.
Gli autori estendono il fenomeno oltre la specie studiata: «Heavy-chain IgGs are a feature of all camelids».
Gli autori prospettano un'applicazione: «These findings open new perspectives in the engineering of antibodies».
Authors' interpretation (attributed, not merged with the data)
The authors argue that the finding «calls into question the role of light chains in the camel» — a question, not a conclusion.
The authors extend the phenomenon beyond the species studied: «Heavy-chain IgGs are a feature of all camelids». This sentence is an extension by the authors from data obtained on the dromedary, and this sheet reports it as such. Subsequent literature confirmed it, but the confirmation lies in other articles, not this one.
The authors envisage an application: «These findings open new perspectives in the engineering of antibodies». It is a prediction, not a result — and it came true, as documented by sheet G-02. The fact that a prediction came true does not retroactively turn it into a datum of this article.
Limiti dichiarati dagli autori
Limitations declared by the authors
Not verifiable: the abstract does not contain a limitations section and the full text is not accessible → [TO BE CHECKED AGAINST THE FULL TEXT].
Limitations added by the editors
- A single species. The data are on Camelus dromedarius. The blanket statement about camelids is not, in this article, a measurement repeated on llama and alpaca.
- No readable quantification. «Considerable» and «extensive» are not measurements. A reader wanting to know what fraction of a dromedary's IgG is HCAb will not find it here.
- No sample size. It is not known how many animals.
- A 1993 article read with 2026 eyes cannot be assessed for reporting, but it can be assessed for accessibility: thirty-three years later, the founding text of an entire field is still closed access, and anyone without a Nature subscription cannot read it. This is not a fault of the authors; it is a fact that affects anyone who needs to verify, and it is the reason why this sheet is shorter than it would like to be.
Finanziamento
Funding
Not stated in the accessible records → [TO BE CHECKED AGAINST THE FULL TEXT]. PubMed classifies the work as Research Support, Non-U.S. Gov't, which says that non-U.S.-government external support existed, not who provided it. Europe PMC reports no funding entry (grantsList: null).
Conflicts of interest
Not declared in the accessible records → [TO BE CHECKED AGAINST THE FULL TEXT]. It should be said as information, not as an accusation, that in 1993 conflict-of-interest disclosure was not standard editorial practice; and that the priority date of the founding patent on this subject matter is 21 August 1992, about ten months before publication (fact verified in chapter 08 §10.1 of this KT). A reader today has the right to know that the filing preceded publication; they have, from this record, no way of knowing whether and how this was disclosed.
- numero, età, sesso e provenienza degli animali da cui viene il siero;
- come è stato ottenuto il siero (prelievo a fini di ricerca? materiale residuo? macello?) e con quale autorizzazione;
- metodi analitici e controlli;
- quantificazione della frazione a catena pesante, con denominatore;
- come è stato saggiato il repertorio di legame;
- finanziamento e conflitti di interesse;
◆ COMMENTO DELL'ASSOCIAZIONE
Camelid Research Studies APS — redazione CRL · 31/08/2026
Valutazione a stelle
What is missing relative to the reporting standard
Since no EQUATOR checklist exists for this design, the list is built on the items that a reader today looks for in an experimental study on animals and that are not present in the accessible records:
- number, age, sex and provenance of the animals the serum comes from;
- how the serum was obtained (drawn for research purposes? leftover material? slaughterhouse?) and under what authorization;
- analytical methods and controls;
- quantification of the heavy-chain fraction, with a denominator;
- how the binding repertoire was assayed;
- funding and conflicts of interest;
- relationship with the August 1992 patent filing.
Almost all of these items are, in all likelihood, present in the full text. This sheet has not seen them and does not assert them.
◆ ASSOCIATION COMMENTARY
Camelid Research Studies APS — CRL editorial team · 31/08/2026
It is the article everything comes from: a vet does not need to know what a VHH domain is to work well, but does need to know why their animals matter so much to researchers.
The first is obvious and should be stated without emphasis: this is the article from which everything comes. A veterinarian caring for alpacas does not need to know what a VHH domain is to do their job well, but they need to know why, when they tell a client that their animals are of interest to biomedical research, they are not repeating a legend. They are repeating a two-page Letter from June 1993 in which eight people from Brussels write that a dromedary's serum contains a considerable amount of antibodies without light chains.
And it is worth saying what that article does NOT say, because popular science writing has added three layers on top of it. It does not say 50 % (that is from 2007, and is about the alpaca). It does not say 12–15 kDa (that is the mass of the isolated recombinant domain, not of the molecule described here, which weighs 100 kDa). And above all it does not contain the anecdote: the students, the human serum discarded over HIV risk, the band that was missing from the gel. That story is alive and perhaps even true, but it lives in secondary sources and in the group's oral retellings, and chapter 08 of this KT already had to strip out two incompatible dates that someone had written as if they were history. Here, it is not given a date.
The second reason is that this sheet stumbled into a demonstration. Chapter 14 of this KT has a section, §9.3, that explains why every sheet must carry the corresponding author's ORCID and why names are copied from the record instead of being reconstructed: because compound and foreign surnames get broken up in indexes, and every break creates a person who does not exist. The example that section uses is «Percie du Sert», broken into «du Sert NP».
While filling in the Authors field of this sheet we found a second one, and a more instructive one, because it is not a break but a doubled consonant: Crossref registers «Hammers C.» and «Hammers R.» where PubMed registers «Hamers C» and «Hamers R». One letter. And that letter had already arrived in 2025, inside the bibliography of the field's reference review published in mAbs — which is the next sheet in this same issue, and which we read in full because it is open access. We did not have to look for it: it was right there, at reference 16.
What we take from this, operationally. That the advice «copy from the record» is incomplete: there are two records and they do not agree. The correct advice is copy from PubMed and, if the names diverge, say so in the sheet. Crossref is excellent for volume, issue, pages and licence — these are the data the publisher deposits and that PubMed often lacks; it is less reliable on proper names, which the publisher types in. In this sheet the two fields come from two different sources for this reason, and it is stated which comes from where.
Where we ask for more. Thirty-three years later, the text that founds a multi-billion-euro field is still behind a paywall, and anyone wanting to verify how many dromedaries were used and how their blood was taken cannot do so. An association concerned with welfare has more reason than most to find this irritating: the welfare question for this line of research begins right there, and the answer is unreadable.
Star rating
Axis Score Rationale A · Scientific quality ★★☆☆☆ The result is clear-cut, never refuted in thirty-three years, and the question is posed with clarity. But the reporting cannot be assessed: only the abstract of the article can be read, and it contains no sample size, methods, controls, ethical statement, funding or conflicts; and the two quantitative terms it carries — «considerable» and «extensive» — are directions, not measurements. The CRL grid does not allow a third star for a work whose method cannot be verified at all. The score is on the accessibility of the reporting, not on the value of the result, and the two diverge more in this sheet than in any other in the issue. B · Clinical utility ★☆☆☆☆ Purely descriptive. No diagnostic, therapeutic or management implication for anyone treating a camelid: there is no sick animal, no intervention, no outcome. We say this without embarrassment — an article can be foundational and clinically useless, and the two judgments sit on different axes precisely so that they can be written together. C · Educational value ★★★★☆ It teaches, in one page, that a structural dogma of immunology (the paratope arises from VH–VL pairing) has a physiological exception in one order of mammals, and that the exception has a precise structural cause (the absence of CH1). The fifth star is denied to the text, not to the content: anyone who today needs to learn what an HCAb is cannot start from here, because they cannot open it — they start from sheet G-02, which is open access, illustrated and up to date. D · Relevance to the species ★★☆☆☆ The data are on the dromedary. The Italian reader of this series treats, in almost all cases, alpacas and llamas. Chapter 14 (§11, note on axis D) is explicit: when the article studies one species and the sheet serves those who treat another, D does not exceed two stars, and the sheet says so. We say it: the sentence «it is a feature of all camelids» is, in this article, an extension by the authors, and the measurement on the alpaca came in 2007 with a different work. No overall score, by design. A = 2 and C = 4 are not a contradiction: an article of which we cannot verify a single line of method remains the place where a true thing was said for the first time.
Bottom line
It can be concluded that in the serum of the dromedary (Camelus dromedarius) there exists a considerable proportion of IgG-like material of 100 kDa, consisting of heavy-chain-only dimers, devoid of light chains, endowed with an extensive antigen-binding repertoire, and that such IgGs lack the CH1 domain, apparently replaced in one class by an extended hinge; and that the authors extend the phenomenon to all camelids and envisage its use in antibody engineering.
It cannot be concluded, from what this sheet has read, what fraction of a camelid's IgG is heavy-chain: the abstract carries no percentages nor denominators, and the «about 50 %» that circulates comes from Maass et al. 2007 and is measured on the alpaca, not here →
[TO BE CHECKED AGAINST THE FULL TEXT]. Nothing can be concluded about how many dromedaries were studied, with what method and under what authorization: sample size, methods, controls, ethical statement, funding and conflicts are not in the accessible records. It cannot be concluded that the finding was measured on llama or alpaca: in this article it was not, and the blanket statement about camelids is an extension by the authors. And nothing clinical can be concluded: there is no sick animal in this work, no intervention and no outcome.
Take-home messages
- Camelid heavy-chain-only antibodies were described in June 1993 in Nature, and they were described in the dromedary. The measurement on the alpaca — about 50 % of IgG, mostly short-hinge — is from 2007 and from a different group.
- The structural reason they hold together is the absence of CH1, the domain that in the conventional antibody holds the heavy chain and light chain together.
- The molecule described here weighs 100 kDa; the «12–15 kDa» read everywhere is the mass of the isolated VHH domain produced recombinantly, which is a different thing and comes later.
- The discovery anecdote is not in the article, and has no documented date. Whoever tells it should tell it as a story.
- Between Crossref and PubMed, on proper names PubMed is to be trusted. Here Crossref gets two surnames out of eight wrong, and the error has already been copied into a 2025 review.
