È stata aperta il 31/08/2026: ha risposto HTTP 200 con 137.191 byte di PDF, e la prima pagina di quel PDF porta stampata la dichiarazione «This is an open-access article distributed under the Creative Commons Attribution License, which permits unrestricted use, distribution, and reproduction in any medium, provided the original work is properly cited».

  • Open access of the article: yes. Unpaywall, queried on 03/09/2026: gold, published version on the publisher's site, licence CC BY.

⚠️ Three registries, two different answers to the same question — and how it was decided. Europe PMC reports isOpenAccess: N, inPMC: N, hasPDF: N: according to that record, the article is not freely available. OpenAlex reports oa_status: gold and cc-by license, with a PDF on the publisher's site. DOAJ has a record for the journal (ISSN 2036-6590/1972-2680).

Chapter 14 (§8.2) states what to do when two registries diverge: the publisher's article page prevails. It was opened on 31/08/2026: it responded HTTP 200 with 137,191 bytes of PDF, and the first page of that PDF carries the printed statement «This is an open-access article distributed under the Creative Commons Attribution License, which permits unrestricted use, distribution, and reproduction in any medium, provided the original work is properly cited». The Europe PMC field is wrong. The article is open, has been downloaded, and has been read line by line: this record is, together with G-02, one of only two in this issue that does not speak from the abstract.

Journal status (verified on 31/08/2026): The Journal of Infection in Developing Countries, ISSN 2036-6590 (print) and 1972-2680 (electronic) per NLM and Europe PMC — Crossref registers only one, 1972-2680; NLM ID 101305410. PubMed: yes. DOAJ: yes — record found, with declared APCs. OpenAlex: 3,837 indexed works, is_oa: true, is_in_doaj: true. It is an open-access journal dedicated to infectious diseases in developing countries — a plausible venue for an Argentine work on a camelid clostridiosis, and indexed in the databases that count.

License of the original article: Creative Commons Attribution (CC BY), read on 31/08/2026 on the first page of the publisher's PDF and confirmed by OpenAlex's license: cc-by field. This record may reproduce: text, data and tables, with attribution. Table 1 of the article is therefore reproduced in full further below, with its legend: it is the only complete numerical dataset in this entire issue, and it is the copyright status that allows it to be given to the reader instead of summarized.

How much weight it carries. Crossref registers 10 citations, OpenAlex 14 (read on 31/08/2026). In seventeen years, for the only existing study on the llama's antibody response to epsilon toxoid.

Specie e soggetti

Species and subjects

26 llama crias (Lama glama), 3–5 months of age, «apparently healthy», belonging to an extensive breeding herd in 9 de Julio, Buenos Aires province, Argentina.

The age is not arbitrary and is the design's best choice: the authors chose 3–5 months because «at this age colostral IgG can no longer be detected» — i.e., to measure the vaccine response without confounding from maternal antibodies. This reasoning is sound; the reference used to support it, however, does not support it (see the Limitations added by the editors field).

Assignment to the four groups (from the full text, verbatim «arbitrarily assigned»):

GroupnAdministrations
unvaccinated controls7none
one dose7day 0
two doses6days 0 and 21
three doses6days 0, 21 and 42
total26

Not stated: sex, weight, sex distribution across groups, and — a relevant fact in an extensive herd — whether the four groups were kept together or separately. Weekly health checks for the entire duration of the study: this, at least, is stated.

Other subjects appear in the ELISA/MNT comparison, and they must be counted because they are animals: one llama (a single one) and 30 albino mice of about 22 g each, used in the neutralization test, via the intraperitoneal route, two mice per dilution, with deaths recorded at 48 hours.

Domanda clinica (PICO)

Clinical question (PICO)

P — in llama crias 3–5 months old, after the disappearance of colostral IgG; I — vaccination with a commercial polyvalent vaccine containing epsilon toxoid of C. perfringens type D, in one, two or three doses twenty-one days apart; C — versus unvaccinated crias from the same herd; O — anti-epsilon antibody titer in IU/ml, measured six times over ninety-three days.

Dichiarato dagli autori: no.

Study type and level of evidence

Prospective controlled experimental study with four arms, non-random assignment. PubMed classifies it as Journal Article · Research Support, Non-U.S. Gov't.

⚠️ The article contradicts itself on assignment. The Methods state that animals were assigned «arbitrarily»; the Statistical methods section states that data were analyzed according to «a completely randomized design». These are not the same thing: the latter is an assumption of the statistical model, the former describes what was actually done in the field. This record registers what the Methods say and flags the discrepancy rather than choosing one over the other.

Level of evidence: controlled experimental study on a surrogate outcome, with very small groups (6–7 per arm) and no sample-size justification. Consequence for the reader: it establishes how the antibody titer moves after one, two or three doses in llama crias; it does not establish whether that titer protects against enterotoxemia, because no animal was challenged — and the authors state this explicitly.

Relevant reporting standard: ARRIVE — this is in vivo experimental research with animals assigned to groups (the crias and, separately, the 30 mice). ARRIVE dates from 2010 and ARRIVE 2.0 from 2020: this article is from 2009 and could not comply with them, and chapter 14 (§3.6) is explicit that demanding it would be an anachronistic reproach. The checklist serves here only to state what a reader does not find, and the most glaringly missing item is the first of the Essential 10: no statement of ethical approval appears in the full text, either for the llamas or for the thirty mice. Declared by the authors: no.

In breve

In brief

Enterotoxemia caused by Clostridium perfringens is one of the leading causes of death in crias, and vaccination against epsilon toxoid is, in Italy as elsewhere, the vaccination that is actually done in camelids — with products registered for sheep and cattle, off-label. In 2009 there was no data whatsoever on the llama's antibody response to that vaccine. This study, seventeen years later, is still the only one.

The result has three parts, and the first two are what a veterinarian needs to know. One dose produces nothing. Two doses twenty-one days apart produce high titers, but they are gone within three months. And the third part is the one nobody expects: the third dose was followed by a decrease in mean titers.

Why this study matters

Because it is the only one, and because clostridial vaccination is the single most common vaccination act in camelid practice. An Italian veterinarian vaccinating crias against enterotoxemia is, every time, doing something whose experimental basis in the species is this article: twenty-six animals, in Argentina, in 2009, with an Argentine vaccine, measured with a test the authors themselves say they do not fully trust.

And because it contains, in one sentence of the discussion, the link between the two halves of this issue — the reason it was worth putting Hamers-Casterman and a clostridial vaccine in the same binder.

Obiettivo

Objective

Declared by the authors: to characterize the IgG response of llamas to vaccination with a commercial anti-enterotoxemia vaccine, and to verify whether an indirect ELISA gives reproducible estimates of antitoxin levels correlated with the in vivo mouse neutralization test.

Metodi / Protocollo

Il vaccino

Mancha Gangrena Enterotoxemia, Instituto Rosenbusch Sociedad Anónima, Argentina.

Sede: «over the shoulder», sopra la spalla. Tecnica: asettica.

L'ELISA

ELISA indiretta secondo un metodo già pubblicato. Antigene: tossina epsilon a 50 UI/ml diluita 1:3200 in tampone carbonato pH 9,6, fornita dall'Instituto Rosenbusch SA; adsorbimento in piastra per cinque giorni a 4–8 °C.

A HRP), scelta dagli autori perché «known to detect IgG of llamas». Substrato TMB/H₂O₂, reazione fermata a cinque minuti con H₂SO₄ 0,5 N, lettura a 450 nm, risultati espressi in UI/ml.

Software InfoStat, 2009.

Methods / Protocol

The vaccine

Mancha Gangrena Enterotoxemia, Instituto Rosenbusch Sociedad Anónima, Argentina. It is a combined vaccine against symptomatic anthrax (mancha), gangrene and enterotoxemia. The enterotoxemia component is an inactivated, ultrafiltered C. perfringens type D toxin, adsorbed onto an aluminum salt.

Route, site, species and product are given. The dose volume is not. Route: subcutaneous. Site: «over the shoulder». Technique: aseptic. Species: Lama glama, crias 3–5 months old. Product: named in full, with manufacturer. Dose volume: not reported in the full text → [TO BE CHECKED] — and the full text was read in its entirety, so here this means that the number is not in the article.

This record does not write a number of milliliters and does not infer it from the product's ovine label. Chapter 14 and this collection's rules forbid it, and in this case the prohibition is not merely formal: a clostridial vaccine dosed by eye on a 3–5-month-old cria is exactly the kind of improvisation that harms an animal.

The schedule and the sampling

Administrations on days 0, 21 and 42 according to group. Samples from the jugular vein on days 0, 16, 28, 49, 59 and 93; sera stored at −20 °C.

The ELISA

Indirect ELISA following a previously published method. Antigen: epsilon toxin at 50 IU/ml diluted 1:3200 in carbonate buffer pH 9.6, supplied by Instituto Rosenbusch SA; plate adsorption for five days at 4–8 °C. Test and control sera diluted 1:100, 60 minutes at 37 °C. Positive control: rabbit anti-C. perfringens D antitoxin, USDA lot IRP249, at 10 IU/ml; negative control: normal rabbit serum.

⭐ The detector, and this is the point that links this record to G-01. The detection conjugate is not an anti-llama IgG: it is Protein A conjugated to horseradish peroxidase (Protein A HRP), chosen by the authors because «known to detect IgG of llamas». TMB/H₂O₂ substrate, reaction stopped at five minutes with 0.5 N H₂SO₄, read at 450 nm, results expressed in IU/ml.

Why this matters is set out in the field Authors' interpretation, and it alone is worth the whole record.

The threshold

A llama was classified as carrying a high titer if its antibody level exceeded 0.25 IU/ml. The threshold was not determined in the llama: it was imported from the goat, declaredly («As in the case of goats»), citing a 1998 study on the variability of kids' response to a commercial epsilon toxoid.

The mouse neutralization test (MNT)

⚠️ This is the part the abstract presents as a validation, and that the full text reveals to be something far more fragile.

The ELISA/MNT comparison was done on the sera of ONE SINGLE llama, vaccinated on days 0 and 16 and sampled on days 0, 16 and 47. One animal, three time points. Serial dilutions from 1:2 to 1:256 incubated with pre-titrated epsilon toxin at 37 °C for 60 minutes, then inoculated intraperitoneally into 30 albino mice of about 22 g, two mice per dilution, following the British Pharmacopoeia 2007 method; deaths recorded at 48 hours.

The statistics

Logarithmic transformation of optical densities, repeated measures over time, design declared as «completely randomized» (see the contradiction flagged above), four treatment levels and six time levels, multiple comparisons with the Scheffé test. Software InfoStat, 2009.

Sicurezza

«Vaccinated animals remained healthy for the duration of the study and all recorded weight gains.» Nessun effetto collaterale apparente.

Values in IU/ml, mean ± standard deviation, in crias of Lama glama 3–5 months old. «High titer» threshold set by the authors at 0.25 IU/ml, imported from the goat.

To be read together with the table: the maximum value reached in the entire study is 0.31 ± 0.19 IU/ml, on day 49, in the two-dose group. The standard deviation is 0.19 on a mean of 0.31: the variability among the six animals is enormous, and this is visible only because the authors published the dispersion instead of the mean alone.

The proportions, each with its own denominator

  • Day 0: titers were substantially equal across all groups (p > 0.05) and no animal had a high titer.
  • Day 16 (16 days after the first dose): similar levels across groups; 3 of 19 vaccinated animals had developed high titers.
  • Day 28 (one week after the second dose): means rising in both groups that had received it; 7 of 12 twice-vaccinated animals had high titers.
  • Day 49 (one week after the third dose for those who received it): 6 of 6 in the two-dose group had high titers; 1 of 6 in the three-dose group. Three weeks earlier, in the three-dose group, 4 of 6 had had high titers.
  • Day 93 (end of the study): 1 of 20 vaccinated animals still had a high titer — and it was an animal from the two-dose group.

The statistical differences

  • The group vaccinated twice differs significantly both from the unvaccinated group and from the group vaccinated once (p < 0.05).
  • No other difference among the pairs compared was significant. The three-dose group does not differ significantly from any other group, including controls.
  • In the multiple comparison across the 24 dose × day values (Scheffé test), the highest response is observed at day 49 in the two-dose group (p < 0.05).

The ELISA/MNT comparison

Significant linear relationship (r = 0.9977, P 0.95) between the LD50 values of the mouse test — antitoxin at 0 / 8.3 / 5.53 IU/ml on days 0, 16 and 47 — and the ELISA optical densities — 0.11 / 2.16 / 0.93 on the same days. On a single llama and three points.

Safety

«Vaccinated animals remained healthy for the duration of the study and all recorded weight gains.» No apparent side effects. Weight gain is not quantified, nor is the weighing method described.

  1. cali di anticorpi dopo vaccinazione con anatossine in soggetti già immuni sono stati riportati in precedenza (citano un lavoro sull'anatossina tetanica nell'uomo);
A binds with only 75 % of IgG of llamas, the third vaccination stimulated an increase in an undetected isotype of IgG while decreasing levels of detectable isotypes».

Authors' interpretation (attributed, not merged with the data)

The authors conclude that llamas develop antibodies against epsilon toxoid after two vaccinations 21 days apart, and that further studies are needed to determine whether those inoculations protect against enterotoxemia and what the most appropriate schedule would be.

The authors state that ELISA is comparable to MNT for detecting anti-epsilon antibodies in llama serum, and they claim its advantage: it is «ethically, economically and technically more acceptable». The first adjective is pertinent — the alternative is killing mice — and this record reports it as the authors' argument, noting that it rests on a comparison made with one llama and thirty mice.

The authors offer two explanations for the decline after the third dose, and present them as such («There is no obvious explanation for this finding»):

  1. declines in antibodies after toxoid vaccination in already-immune subjects have been reported previously (they cite a work on tetanus toxoid in humans);
  2. ⭐ and this is the sentence that links the whole issue together: «since protein A binds with only 75 % of IgG of llamas, the third vaccination stimulated an increase in an undetected isotype of IgG while decreasing levels of detectable isotypes». That is: the decline may not be a decline in antibodies, but a decline in antibodies visible to the test.

The authors acknowledge that the observed response was short-lived, and that the vaccine should therefore be administered «frequently or strategically»; and they propose evaluating different adjuvants and, above all, conducting challenge trials to establish whether the vaccine protects.

Limiti dichiarati dagli autori

Citati, non riassunti:

  • non si sa se i titoli osservati indichino un'immunità efficace alla sfida, né quanto persista;
  • lo schema vaccinale definitivo per il lama resta da determinare;
  • il calo dopo la terza dose non ha una spiegazione ovvia;
  • la Proteina A lega solo il 75 % delle IgG di lama — limite dichiarato dagli autori sull'oggetto stesso della loro misura;

Limitations declared by the authors

Quoted, not summarized:

  • it is not known whether the observed titers indicate effective immunity to challenge, nor how long it persists;
  • the definitive vaccination schedule for the llama remains to be determined;
  • the decline after the third dose has no obvious explanation;
  • Protein A binds only 75 % of llama IgG — a limitation declared by the authors about the very object of their measurement;
  • challenge trials are needed, as well as evaluation of other commercial vaccines and other intervals.

    • il riferimento 11 è «Hamers-Casterman C, Atarhouch T, Muyldermans S, Robinson G (1993) Naturally occurring antibodies devoid of light chains. Letters to Nature 363: 446-448» — cioè l'articolo della scheda G-01 di questo fascicolo — ed è chiamato a sostenere che «at this age colostral IgG can no longer be detected». Quell'articolo non parla di IgG colostrali né di cria: descrive anticorpi a catena pesante nel siero di dromedario. Lo stesso riferimento elenca inoltre quattro autori su otto senza «et al.», che è precisamente l'elenco troncato che il capitolo 14 (§10, campo 3) definisce «un elenco falso»;
    • il riferimento 14, chiamato per il test di Scheffé, è «Pugh DG, Wright J, Rowe S (1995) Serologic response of llamas to a commercially prepared leptospirosis vaccine». Un metodo statistico attribuito a uno studio sulla leptospirosi. Non è malafede: è quasi certamente uno slittamento di numerazione in una lista compilata a mano. Ma chi usa quella bibliografia come fonte di riferimenti sta copiando attribuzioni sbagliate, ed è esattamente il meccanismo che ha reso necessaria la riscrittura della versione 2 di questa KT.

Limitations added by the editors

  • Groups of six and seven animals, with no sample-size justification. No power calculation. With six animals per arm, «6 of 6» and «1 of 6» are differences of four or five subjects.
  • No statement of ethical approval, either for the crias or for the thirty mice that were inoculated intraperitoneally with epsilon toxin and observed until death. This is the most serious reporting gap.
  • The «high titer» threshold is the goat's. 0.25 IU/ml was not validated in the llama, and all the article's proportions rest on that threshold — including «6 of 6» and «1 of 20». Shift the threshold and the results change.
  • The ELISA validation rests on a single animal. An r = 0.9977 calculated on three points from one llama is not a method validation; it is an encouraging clue. The abstract writes «The ELISA gave comparable results to the MNT» without that denominator, and a reader of the abstract alone believes something far more solid than what was actually done.
  • Group assignment is contradicted within the article itself («arbitrarily» in the Methods, «completely randomized» in the statistics).
  • No blinding in plate reading.
  • The dose volume is not reported, and without a volume the protocol is not reproducible.
  • Discrepancy between abstract and full text on a number. The abstract states that the high titers of 7/12 on day 28 «were present in only one animal at the end of the study (day 93)»; the full text shows that on day 49 the two-dose group was 6 of 6, i.e., that between days 28 and 49 titers had actually kept rising. The two statements are reconcilable, but the abstract gives the impression of a monotonic decline that the data do not show.
  • Two bibliographic references are called upon to support things they do not say, and this is a verifiable defect once their list is opened:
    • reference 11 is «Hamers-Casterman C, Atarhouch T, Muyldermans S, Robinson G (1993) Naturally occurring antibodies devoid of light chains. Letters to Nature 363: 446-448» — i.e. the article of record G-01 in this issue — and it is called upon to support that «at this age colostral IgG can no longer be detected». That article does not discuss colostral IgG or crias: it describes heavy-chain antibodies in dromedary serum. The same reference furthermore lists four of eight authors without «et al.», which is precisely the truncated listing that chapter 14 (§10, item 3) defines as «a false listing»;
    • reference 14, cited for the Scheffé test, is «Pugh DG, Wright J, Rowe S (1995) Serologic response of llamas to a commercially prepared leptospirosis vaccine». A statistical method attributed to a leptospirosis study. This is not bad faith: it is almost certainly a numbering slip in a manually compiled list. But anyone using that bibliography as a source of references is copying wrong attributions, and it is exactly the mechanism that made rewriting version 2 of this KT necessary.
  • Seventeen years, another continent, another product. Mancha Gangrena Enterotoxemia by Rosenbusch is not registered in Europe. The observed kinetics are those of that product's toxoid, with that adjuvant.

Finanziamento

Dichiarato. «This work was supported by a research grant from the International Foundation for Science (IFS B3069/1).»

Ma i ringraziamenti dichiarano anche altro, e va letto insieme: «The authors wish to thank the Rosembusch Institute for technical support and for donating the vaccine» — supporto tecnico e donazione del vaccino dal produttore.

Funding

Declared. «This work was supported by a research grant from the International Foundation for Science (IFS B3069/1).» IFS is an international foundation that funds researchers in developing countries: it is not the vaccine manufacturer, and this should be stated.

But the acknowledgments also declare something else, and it must be read together: «The authors wish to thank the Rosembusch Institute for technical support and for donating the vaccine» — technical support and donation of the vaccine from the manufacturer. And the Methods state that the epsilon toxin antigen used in the ELISA was supplied by the same Instituto Rosenbusch SA de Biologia Experimental Agropecuaria.

Conflitti di interesse

Dichiarati dagli autori: assenti. Il testo pieno chiude con la riga: «Conflict of Interest: No conflict of interest is declared.»

  1. il quarto autore, Iribarren F., porta come affiliazione numero 5 l'Instituto Rosenbusch SA, che è il produttore del vaccino provato;
  2. l'Istituto ha donato il vaccino e fornito supporto tecnico (ringraziamenti);
  3. l'Istituto ha fornito anche l'antigene con cui è stata costruita la prova di misura;

Conflicts of interest

Declared by the authors: none. The full text closes with the line: «Conflict of Interest: No conflict of interest is declared.»

⚠️ This record registers the facts that stand next to that line, as information and not as an accusation (chapter 14, §5.4):

  1. the fourth author, Iribarren F., carries as affiliation number 5 the Instituto Rosenbusch SA, which is the manufacturer of the vaccine tested;
  2. the Institute donated the vaccine and provided technical support (acknowledgments);
  3. the Institute also supplied the antigen with which the measurement test was built;
  4. the study concludes that the product induces antibody development.

There is nothing irregular in a company collaborating on an independent study of its own product, and transparency is present: the three facts are all printed in the article, on the first page and at the end. What does not add up is the final line, because that line tells the hurried reader — and the automated systems that index conflict-of-interest statements — that there is nothing to know, when there is. The correct formulation would have been to declare it and explain why it did not influence the outcome.

What changes, practically? On the raw data, nothing: the table is published with its dispersions and the results are not flattering for the product (one dose does nothing, titers vanish within three months, the third dose lowers them). If there had been pressure, it does not show. But the point of a conflict-of-interest statement is not that the article is wrong: it is that the reader can decide for themselves.

  • approvazione etica e numero di protocollo, per i cria e per i 30 topi;
  • giustificazione della numerosità (nessun calcolo di potenza; bracci da 6–7);
  • metodo di assegnazione ai gruppi, descritto in modo coerente fra Metodi e statistica;
  • cecità di chi ha letto le piastre;
  • volume della dose vaccinale;
  • sesso e peso degli animali, e loro distribuzione fra i gruppi;
  • validazione della soglia di 0,25 UI/ml nella specie in cui è usata;
  • numerosità del confronto ELISA/MNT dichiarata nell'abstract accanto al coefficiente;
  • dichiarazione del conflitto derivante dall'affiliazione del quarto autore e dalla fornitura di vaccino e antigene;

◆ COMMENTO DELL'ASSOCIAZIONE

Camelid Research Studies APS — redazione CRL · 31/08/2026

Valutazione a stelle

AsseVotoMotivazione

What is missing relative to the reporting standard

Relative to ARRIVE applied retroactively (the article is from 2009, ARRIVE from 2010: the list states what a reader does not find, not that the authors erred):

  • ethical approval and protocol number, for the crias and for the 30 mice;
  • sample-size justification (no power calculation; arms of 6–7);
  • method of assignment to groups, described consistently between Methods and statistics;
  • blinding of those who read the plates;
  • vaccine dose volume;
  • sex and weight of the animals, and their distribution across groups;
  • validation of the threshold of 0.25 IU/ml in the species in which it is used;
  • sample size of the ELISA/MNT comparison stated in the abstract alongside the coefficient;
  • statement of the conflict arising from the fourth author's affiliation and the supply of vaccine and antigen;
  • fate of the 30 mice and humane endpoint criteria.

◆ ASSOCIATION COMMENTARY

Camelid Research Studies APS — CRL editorial staff · 31/08/2026

The experimental basis for clostridial vaccination in this species is this article: twenty-six animals, in Argentina, in 2009, with an Argentine vaccine.

An Italian alpaca breeder vaccinates their animals against enterotoxemia. They do so with a product registered for sheep or cattle, off-label, on a schedule taken from small ruminants. It is a reasonable and widespread practice, and its entire experimental basis in the species — the entire basis, not part of it — is this article: twenty-six llama crias, in Argentina, in 2009. Seventeen years, ten citations.

The three things it says, and that must be said in this order.

One dose does nothing. On day 16, after the first administration, the one-dose group is at 0.13 ± 0.02 IU/ml versus 0.12 ± 0.01 for the unvaccinated, and by the end of the study that group does not differ statistically from controls. A cria vaccinated once is, as far as we know, an unvaccinated cria. This is the third time in this issue that we find this, across three different diseases.

Two doses work, and then the titer disappears. On day 49 all six twice-vaccinated animals have high titers; by day 93 only one of twenty vaccinated animals in total still has one. From 0.31 to 0.14 IU/ml in forty-four days. If anyone thinks a clostridial vaccination done in spring covers the autumn, this is the datum that says no — with all due caution.

The third dose was followed by a decline. 4 of 6 animals with a high titer on day 28, one of six on day 49, one week after the booster. The authors do not pretend to understand it and write «there is no obvious explanation», which is the right way not to explain something.

And here is the reason this article belongs in an issue about nanobodies.

One of the two explanations the authors put on the table is that the decline is not a decline in antibodies, but a decline in antibodies the test manages to see. Their ELISA does not use an anti-llama IgG: it uses Protein A, and Protein A — they write, citing the literature — binds only 75 % of this species' IgG. If the third dose had shifted the response toward an isotype that Protein A does not bind, the measured titer would fall while antibodies rise.

Why don't a llama's IgG all bind the same way? Because they are not all the same type of molecule. About half of a camelid's IgG are heavy-chain-only antibodies, lacking light chains and lacking the CH1 domain — the thing Hamers-Casterman and colleagues described in 1993 in dromedary serum, and it is record G-01 of this issue; the proportion of roughly 50 % was measured in the alpaca by Maass and colleagues in 2007. The reagent that a collection like this encounters when discussing biotechnology — Protein A, which is used to purify VHHs precisely because it binds them — is the same reagent that in a tube of Argentine clinical serology acts as the detector, and does so unevenly across that animal's IgG classes.

This is the point of the whole issue, and it is worth saying in full. The peculiarity that made camelids famous in biotechnology is not a laboratory curiosity: it is a property of the serum the veterinarian draws. Every time an alpaca or llama serum is sent to a serological test developed for another species, one is assuming that the detector sees all of its immunoglobulins. No one has verified this for most of the tests we use. This 2009 article is, as far as we have read, the only place where the problem is spelled out in plain words — and it arrived by the back door, as a possible explanation for a strange result.

Where we dissent. This article's abstract states that ELISA gives results comparable to the mouse test. The full text shows that comparison was made on one llama and three sampling points. A correlation coefficient on three points from one animal does not validate a method, and the abstract sentence — the one that will be cited — says more than was actually done. We say this with some discomfort, because we appreciate that the authors tried to replace a test that kills thirty mice with one that kills none; but the way to defend that goal is to validate it well, not to announce it.

And we record, without dancing around it, the conflict the article declares it does not have. The fourth author works for the company that manufactures the vaccine tested; the company donated the vaccine and supplied the antigen with which the test was built; the final line says there are no conflicts to declare. The data are published with their dispersions and are not flattering for the product, which is the best indication that no one put a thumb on the scale. But the line remains wrong, and a collection that teaches how to read articles cannot let it pass just because it likes the article.

Our operational position, for members. The clostridial three-week booster is not optional, and the first dose alone should be considered ineffective. Duration is short: if this article's reasoning holds in the species being kept, a single annual booster is probably the minimum, and its timing should be chosen based on when the crias are at risk, not on the breeder's calendar. And the choice of product, volume and dates is made with one's own veterinarian, because this record has no number of milliliters to give you: it is not in the article, and we do not invent it.

Star rating

AxisScoreRationale
A · Scientific quality★★☆☆☆It has things almost nothing else in this issue has: an unvaccinated control group, four arms, a schedule of six samplings over 93 days, a statistical analysis with multiple comparisons, and the table published with standard deviations instead of means alone. But: no ethical statement (for the crias and for thirty mice inoculated with toxin), no sample-size justification with arms of 6–7, assignment described in two incompatible ways within the same article, a positivity threshold imported from the goat and never validated in the species, and a method validation conducted on a single animal yet presented in the abstract without the denominator. Two stars: the design is there, the reporting fails at the points that decide the outcome.
B · Clinical utility★★☆☆☆It concerns the most common vaccination given to these animals and states three things that change behavior — one dose is not enough, the titer lasts a short time, the third dose adds nothing. This is the maximum the grid allows: chapter 14's constraint forbids a score ≥ 3 based on protocols the article does not report in full, and here the dose volume is missing; and upstream, the product is not on the market in Europe and no one has shown that those titers protect. A strong operational hypothesis to be verified, not a protocol to apply.
C · Educational value★★★★☆It teaches four things in four pages: that a single dose produces no response; that duration is the question that comes after efficacy; that a threshold imported from another species decides the results without anyone noticing; and — the best one — that a serological test's detector may not see all of the immunoglobulins of the animal being tested. The fifth star is withheld because read alone it also teaches something false: that ELISA was validated against the mouse test. It teaches extremely well, but only if read together with its limitations — which is, after all, this record's job.
D · Relevance to the species★★☆☆☆Data on real camelids, in a real herd, with an unvaccinated control. But the context is an extensive breeding herd in Argentina, with an Argentine vaccine not registered in Europe, and the species is the llama while the Italian reader of this collection mostly keeps alpacas. Chapter 14 (§11, note on axis D) asks not to exceed two stars when the article studies one species and the record serves someone who keeps another: we respect that, and we say so.

No overall score, by design. A = 2 and C = 4 are not a contradiction: it is a fragile article that contains, by chance and in one sentence of the discussion, the most important thing in this whole issue.

Bottom line

It can be concluded that, in 26 crias of Lama glama 3–5 months old in an extensive Argentine herd, vaccinated subcutaneously over the shoulder with the combined vaccine Mancha Gangrena Enterotoxemia (Instituto Rosenbusch SA, Argentina): a single dose produced no titers (one-dose group indistinguishable from controls); two doses 21 days apart produced a significant increase in mean titers (p < 0.05) with 6 of 6 animals above the 0.25 IU/ml threshold on day 49, and the two-dose group is the only one to differ significantly both from unvaccinated animals and from the one-dose group; that by day 93, 1 of 20 vaccinated animals remained above threshold; and that a third dose on day 42 was followed by a decrease in mean titers one week later (1 of 6 above threshold on day 49 versus 4 of 6 three weeks earlier). No apparent side effects, weight gains in all animals.

It cannot be concluded that the crias were protected against enterotoxemia: no challenge trial was performed, and the authors state so. The 0.25 IU/ml threshold cannot be used as a protective value in the llama: it was imported from the goat and has never been validated in this species — and all this study's proportions depend on it. It cannot be concluded that ELISA is validated against the mouse neutralization test: the comparison rests on a single llama and three sampling points, and the abstract reports the coefficient without the denominator. It cannot be concluded that the decline after the third dose is a decline in antibodies: the authors themselves raise the alternative hypothesis that it is a decline in detectable antibodies, because the Protein A used as detector does not bind all of the llama's IgG — and the exact fraction, its distribution across IgG classes, and the primary source of the «75 %» are not verified in this record → [TO BE CHECKED]. No dose volume can be given: it is not in the full text. And nothing quantitative can be transferred to vaccines registered in Italy, nor to the alpaca: the product is Argentine and the species studied is the llama.

Messaggi da portare a casa

Key takeaways

  1. A single dose of epsilon toxoid produces no titers in the llama cria. The one-dose group does not differ statistically from unvaccinated animals.
  2. Two doses twenty-one days apart work: 6 of 6 animals above threshold on day 49, and it is the only group that differs from controls.
  3. Duration is short: by day 93 only 1 of 20 vaccinated animals remained above threshold.
  4. The third dose was followed by a decrease in mean titers, and the authors state they have no obvious explanation.
  5. The «high titer» threshold comes from the goat, not the llama. All this study's proportions rest on a borrowed number.
  6. ⭐ Protein A does not see all of a llama's IgG, because about half of a camelid's IgG are heavy-chain-only antibodies. A serological test developed for another species may measure less than it thinks it does — and it is the same biology that made these animals famous in biotechnology.
  7. The claim that ELISA is comparable to the mouse test rests on one llama and three sampling points. Anyone citing it should also cite the denominator.

Issue coda — what this group of records leaves open

Three questions this issue does not answer, and that no one has yet studied.

  1. What fraction of llama and alpaca immunoglobulins is bound by the detection reagents we use every day? The «75 % for Protein A» in record G-05 is reported by the authors citing a third-party work, and this editorial staff has not verified it at the source. But the question is broader than the single number: every serological test developed for another species and applied to a camelid is assuming something no one has measured. In a species where about half of the IgG are heavy-chain-only, the assumption is not innocuous.
  2. How long does vaccine immunity last in camelids? The day 156 datum from record G-04 was collected in Switzerland and is not in the abstract; the datum from record G-05 states that at ninety-three days only one animal in twenty remained above threshold. It is the question every breeder asks right after «does it work?», and it is the one on which the literature is poorest.
  3. What species, what weight, and what percentage of blood volume? Record G-02 was the first in this KT to bring a sampling volume with an open primary source — 100 mL — and showed that this number, without species and without weight, cannot yet be judged against the 10 % threshold of Annex VIII of Directive 2010/63/EU. It is a three-line answer and anyone managing an immunization herd knows it.

One thing this issue has instead closed. Chapter 08 §11.3 of this KT stated that it had removed the figure «100–250 mL» for the final bleed of an immunized camelid, for lack of a source. Record G-02 now brings a figure with its primary source, open and citable: 100 mL, seven days after the last of four subcutaneous administrations of 300 µg, in the standard protocol of Evers et al. 2025. Chapter 08 must be updated accordingly, and with it the note declaring the gap.

Final note on method. The twenty-five bibliographic claims in this issue were copied from the responses of the Crossref and Europe PMC APIs, queried on 31/08/2026; OpenAlex and DOAJ were used for journal and article status. WebSearch was not used. Where the two registries diverge — on author names (G-01), on publisher and ISSNs (G-04), on open-access status (G-05) — the divergence is written into the record rather than smoothed over, because it is itself information: it tells the reader how solid the ground is that this collection walks on.
