◆ COMMENTO DELL'ASSOCIAZIONE

Camelid Research Studies APS — redazione CRL · 31/08/2026

◆ ASSOCIATION COMMENTARY

Camelid Research Studies APS — CRL editors · 31/08/2026

This is the only work in which a European country's national diagnostic network searched for eleven groups of agents on the same animals — ten flocks, chosen by judgement. Not because it is perfect — it is a sample of ten flocks chosen by judgement, without a sample-size calculation — but because it is the only one in which the national diagnostic network of a European country has searched for eleven groups of agents on the same animals, four times over two years, stating every denominator and every interval. Before this, what were cited were cross-sectional studies on one agent at a time, and numbers were summed that were not summable.

Three numbers to take away, and one not to.

The first: 232 of 716 faecal samples (32.4 %) carry stx genes, i.e. indicate shedding of Shiga toxin-producing E. coli, and per animal it rises to 187 of 442 (42.3 %). There are no published human outbreaks attributed to South American camelids — this datasheet does not claim it and neither does the article. But the route is orofaecal, EFSA considers every STEC strain potentially pathogenic for humans, and the measure that interrupts that route costs as much as a gel dispenser and thirty seconds: wash your hands after touching an alpaca, before eating. At educational farms this is the one number from this article that needs to be said to parents, and it is enough.

The second: 54.6 % of German camelids (245 of 449) have antibodies against Schmallenberg virus, in every flock and at every round. It is not a zoonosis, it causes no clinical signs in this species, and it requires nothing new: the Culicoides control already done for bluetongue covers it. It does, however, close a discussion: camelids are inside the transmission cycle, not alongside it.

The third, and it is the one that changes a decision: 36 of 739 sera (4.9 %), from 30 animals, were non-negative on the ELISA for the M. tuberculosis complex in disease-free flocks — and only 3 of the 19 animals with more than one sample reproduced the result. The authors say it without qualification: a test with that specificity is not usable in a disease-free population, because it would produce an unacceptable number of presumably false positives.

The number not to take away is «Candidatus Mycoplasma haemolamae 31.6 %» (142 of 449 animals, found with a PCR on blood) set next to «Coxiella burnetii 0.7 %» (3 positive sera of 738, found with an ELISA not validated for the species). These are two different measurements of two different things with two different methods, and the authors write explicitly that they are not comparable. Anyone who lines them up on a slide is constructing a ranking that the data do not support.

The piece of theory worth the read on its own. Why do serological tests get it so wrong in camelids? Because these animals produce two types of IgG: the mammalian kind and heavy-chain antibodies (HCAb), structurally smaller, which according to the literature cited by the authors can reach 75 % of total serum IgG. An indirect assay — an ELISA, an immunofluorescence — is built around a conjugate that recognizes the IgG of the host for which the kit was validated. In camelids, three-quarters of the target have a different shape. That is why in this species a ruminant ELISA can be wrong in both directions, and why the same animal can be negative on one assay and positive on another. It is the same reason VHH nanobodies exist and are worth an entire literature (chapter 08): the same immunological peculiarity that makes these animals valuable in the laboratory makes them difficult in diagnostics.

What changes for a veterinarian in Italy, today. (a) A positive serology in a camelid, for any agent, is repeated and confirmed with a different method before telling the owner. Here 16 of the 19 rechecked animals did not reproduce the result. (b) A camelid rectal swab sent for PCR has a real probability of being unusable due to inhibition: 87 of 717 samples, 12 %. Worth knowing before interpreting a "negative" as an absence. (c) A chlamydia positive on PCR in a camelid is not news until the species is typed: here three-quarters of positives were a C. suis-like bacterium, the zoonotic C. psittaci was absent, and the abortifacient C. abortus appeared in eleven animals from a single flock and in a single round, most likely from contamination by cohabiting ruminants. (d) The authors close by noting that in Germany there are no specific hygiene guidelines for camelids in animal-assisted interventions, whereas there are for dogs. In Italy the situation has not been verified in this datasheet and cannot be inferred from a German article: it is, however, exactly the gap that a trade association can fill, and it is why this dossier exists.

What this datasheet cannot tell you. Whether these numbers hold for Italy. Ten German flocks, with their sizes, their climates and their practices, are not twelve flocks in Lombardy and Piedmont — which is precisely what datasheet CRL-I-04 measures, and it is the reason these two sit next to each other in this dossier.

The composition of the group is itself a piece of information. Twenty-two authors distributed across five institutes of the Friedrich-Loeffler-Institut — the German federal institute for animal health — and three university clinics in Giessen: this is not a single research group, it is the national diagnostic network put to work on one species. Each agent was searched for by the reference laboratory responsible for it. The consequence for data quality is in field 22, and it is positive; the consequence for the limitations is that no single agent was studied in depth.

9 · Specie e soggetti

9 · Species and subjects

South American camelids in Germany: 449 animals — 292 alpacas (Vicugna pacos), 156 llamas (Lama glama) and 1 huarizo (llama × alpaca hybrid; the article counts it separately, and this datasheet does the same).

From 449 animals were collected 739 blood samples and 723 faecal samples, because 10 flocks were visited four times at intervals of about six months, with 20 animals sampled per visit: several animals were therefore sampled more than once. The periods of the four rounds are stated: A = 04/2021–09/2021 · B = 11/2021–04/2022 · C = 05/2022–09/2022 · D = 11/2022–02/2023.

Animal inclusion criteria, verbatim: animals had to be at least 2 years old and be selected and classified as "healthy" by the owner prior to the study. Each was given a general clinical examination, with assessment of grooming state, body condition score, mucous membrane colour and recording of any abnormalities (skin lesions, limb deformities).

Flock distribution (Hesse 2, Saarland 1, Bavaria 2, North Rhine-Westphalia 2, Rhineland-Palatinate 1, Saxony 2), with sizes from 20 to 68 sampled camelids per flock, and compositions ranging from alpaca-only (flocks 4, 6, 7, 9, 10) to llama-only (flock 5) to mixed.

10 · Domanda clinica (PICO)

10 · Clinical question (PICO)

  • P — clinically healthy South American camelids, aged ≥ 2 years, in ten German flocks chosen to reflect national variety in size and use.
  • I / E — the exposure is membership in the species and the husbandry system; there is no intervention.
  • C — no internal comparison group; the comparison is with published prevalence data for European livestock species, discussed by the authors.
  • O — presence (serological, cultural, by PCR) of eleven groups of viral and bacterial agents of epizootic and zoonotic relevance, with per-sample and per-animal rates.

As a question: what do healthy South American camelids reared in Europe carry, and how often?

11 · Disegno dello studio

Gli autori lo distinguono espressamente dallo studio precedente sulla stessa popolazione: «This study differs from the precursor study by González–Santamarina et al. by incorporating a strong longitudinal component rather than being purely cross-sectional, and it sampled individual animals instead of taking composite fecal samples».

11 · Study design

Repeated cross-sectional study (with a longitudinal component) on flocks selected for representativeness, with individual sampling repeated over time. The authors expressly distinguish it from the earlier study on the same population: «This study differs from the precursor study by González–Santamarina et al. by incorporating a strong longitudinal component rather than being purely cross-sectional, and it sampled individual animals instead of taking composite fecal samples».

Sampling is not random: flocks were chosen from among participants in a previous survey, according to five stated criteria — primary activity (6 of 10) rather than secondary (4 of 10); high headcount (over 50); inclusion of llama herds alongside the predominant alpacas; coverage of all German regions; diversity of uses.

12 · Level of evidence

Evidence from an observational cross-sectional study with a longitudinal component, on a sample selected by stated criteria (non-random), with denominators and 95 % confidence intervals reported systematically.

Consequence for the reader: it is the best available estimate of the frequency of these agents in European camelids, and its numbers can be used as a benchmark. It does not establish causal links between carriage and disease (the animals were healthy by design), it does not demonstrate transmission to humans, and it is not a national prevalence: ten selected flocks are not a probability sample of the German population, and the authors do not claim it is.

13 · Standard di reporting pertinente

STROBE (studi osservazionali).

13 · Relevant reporting standard

STROBE (observational studies). ⚠️ The authors declare no reporting checklist: the full text was read in its entirety and contains neither the word STROBE nor any reference to a reporting guideline. This is a verified absence, not a marker of missing data.

14 · Metodo

Letto nel testo pieno (sezioni 2.1-2.13).

Selezione. Lo studio si appoggia a campioni e dati del progetto «Development and establishment of a multi-stage animal health management system for farms with South American camelids», parte dei progetti modello e dimostrativi (MuD) sul benessere animale, condotto in cooperazione con un'associazione tedesca di allevatori (Verein der Züchter, Halter und Freunde von Neuweltkameliden e.V.). Cinque criteri di selezione dichiarati (campo 11).

Campionamento.

Dichiarazione dei metodi, testuale: «Diagnostic methods applied herein followed WOAH or EU-RL recommendations, whenever available, although not all are validated for use in SAC».

Figure in GraphPad Prism 10.4.0.621.

Etica.

Dato metodologico che condiziona ogni denominatore: «Because of material shortages and logistical constraints not all samples were subjected to all diagnostic tests». Ecco perché i denominatori del campo 15 cambiano da un agente all'altro — 739, 738, 723, 719, 717, 716, 630, 442, 370, 376 — e perché non si possono confrontare fra loro senza guardare quale sia.

Selection. The study draws on samples and data from the project «Development and establishment of a multi-stage animal health management system for farms with South American camelids», part of the model and demonstration projects (MuD) on animal welfare, conducted in cooperation with a German breeders' association (Verein der Züchter, Halter und Freunde von Neuweltkameliden e.V.). Five stated selection criteria (field 11).

Sampling. Four visits per flock about six months apart, 20 animals per visit — a number chosen «for practical and compliance reasons», i.e. for feasibility and adherence, not from a sample-size calculation. Faeces taken directly from the rectum: swabs and about 25 g of faecal material collected with a gloved hand into 100 ml containers, avoiding cross- contamination between samples and animals. Blood from the left jugular: 5 ml in an EDTA tube and 15 ml without anticoagulant; serum obtained by centrifugation the following day (2,808 × g, 5 min), cooled to 4 °C and sent by express courier to the FLI's distributing laboratory in Jena, or hand-delivered to the collaborating laboratories in Giessen.

Tests performed. Serology for ruminant pestiviruses (BVDV-1, BVDV-2, BDV), BoHV-1, Schmallenberg virus (SBV), Borna disease virus 1 (BoDV-1, two-step IFAT: screening on formalin-fixed Vero cells, confirmation on heat-fixed SK6 cells), Brucella (iELISA with confirmation by complement fixation), Coxiella burnetii (ELISA), Chlamydiaceae, Mycobacterium tuberculosis complex (Tuberculosis DR-ELISA). Direct tests: PCR for Candidatus Mycoplasma haemolamae; PCR for Chlamydia spp. with species typing; qPCR IS1111 for C. burnetii; culture for Mycobacterium avium subsp. paratuberculosis (MAP) and non-tuberculous mycobacteria; culture for Clostridioides difficile and Clostridium perfringens with ribotyping and screening for toxin genes; culture for Campylobacter spp. and for Salmonella spp.; PCR for the stx1 and stx2 genes (STEC).

Statement of methods, verbatim: «Diagnostic methods applied herein followed WOAH or EU-RL recommendations, whenever available, although not all are validated for use in SAC». It is a sentence worth reading twice: the methods are the official ones, and they are not validated for this species.

Analysis. Explicit distinction between per-sample positivity rate and per-animal positivity rate (an animal sampled more than once is positive if at least one sample is). Fisher's exact test; 95 % confidence intervals by the Wilson score method, with the formula written out in full in the article. Figures in GraphPad Prism 10.4.0.621.

Ethics. Approval from the Regierungspräsidium Giessen, reference V 54–19 c 20 15 h 02 GI 18/4 Nr. A 3/2021, dated 29/04/2021, recognized by the competent authorities of the Länder concerned. Written informed consent from owners, pursuant to Directive 95/46/EC.

Methodological fact that conditions every denominator: «Because of material shortages and logistical constraints not all samples were subjected to all diagnostic tests». This is why the denominators in field 15 change from one agent to another — 739, 738, 723, 719, 717, 716, 630, 442, 370, 376 — and why they cannot be compared with one another without checking which one it is.

16 · Interpretazione degli autori

Attribuita esplicitamente e tenuta separata dal campo 15.

  • Sul virus di Schmallenberg: i risultati «imply that SAC have become an integral part of the natural SBV transmission cycle» fra i Culicoides vettori e gli ospiti mammiferi del Paese. Gli autori richiamano che una precedente infezione sperimentale non produsse segni clinici né lesioni macroscopiche nei lama e negli alpaca, e che uno studio di campo tedesco non riportò segni clinici né aumento di aborti o malformazioni congenite: «it is assumed that SBV causes only subclinical infection in SAC».
  • Su BoHV-1 e BVDV: i camelidi sudamericani «have no decisive relevance for the BoHV-1 control program in Germany».
This particularly applies to sera from camels and SAC as they produce two types of immunoglobulin γ (IgG), i.e., the mammalian IgG and structurally smaller so-called heavy chain IgG antibodies (HCAb). The latter reportedly can represent 75 % of the total IgG content in serum, which may influence the functionality of test systems used and lead to erroneous results in indirect tests».
  • Sulle clamidie: «a positive chlamydia test should be considered only a minor risk as long as abortifacient C. abortus or zoonotic C. psittaci are not detected, emphasizing the need for differential diagnosis at the chlamydial species level».
  • Sugli STEC: i tassi entro allevamento sono simili a quelli pubblicati per le mandrie bovine (Michigan 8,2–53,7 %; Irlanda 52 % e 76 %; Germania 0–100 %); l'EFSA classifica tutti i ceppi STEC come potenziali patogeni per l'uomo, ma «the information on the presence of stx1 and/or stx2 alleles in the individual animal samples generated in this study so far is clearly limited in allowing a critical assessment of the public health risk emanating from SAC».
  • Conclusione generale, testuale: «SAC do not appear to pose a greater direct risk than other livestock species. However, since SAC are often kept together with other susceptible animal species, such as cattle or sheep, they are of particular epidemiological importance as a potential source of potential pathogens». E, sull'uso con le persone: «the detection of foodborne and abortifacient zoonotic agents underscores the importance of maintaining strict hygienic standards. This particularly applies when people interact with these animals, e.g., when SAC are used in animal-supported therapy or tourist activities. Unfortunately, while hygienic guidelines exist for dogs or, more general, for "animals" being deployed in animal-assisted therapy, no specific guidelines have been established or made publicly available for SAC in Germany thus far.»

16 · Authors' interpretation

Explicitly attributed and kept separate from field 15.

  • On Schmallenberg virus: the results «imply that SAC have become an integral part of the natural SBV transmission cycle» between the Culicoides vectors and the country's mammalian hosts. The authors note that a previous experimental infection produced no clinical signs or gross lesions in llamas and alpacas, and that a German field study reported no clinical signs or increase in abortions or congenital malformations: «it is assumed that SBV causes only subclinical infection in SAC».
  • On BoHV-1 and BVDV: South American camelids «have no decisive relevance for the BoHV-1 control program in Germany».
  • On BoDV-1: the virus is strictly neurotropic in spillover hosts, which act as dead-end hosts; the natural reservoir is the bicoloured white-toothed shrew (Crocidura leucodon), in a restricted endemic area of Germany, Austria, Switzerland and Liechtenstein.
  • On serology in camelids — the mechanistic point of the paper: serological tests «are prone to false-positive and -negative results.
  • On tuberculosis — the article's sharpest operational conclusion: all flocks can be considered free of infection on the basis of history, clinical status and epidemiological investigation; the proportion of non-negative results is consistent with the published specificity for that ELISA; «a test with this specificity value is not applicable in an aTB-free population as it would lead to an inacceptable high number of (presumptive false) positive results». The authors add that the animals had not been stimulated with an intradermal test 15-30 days before sampling, as recommended by the European reference laboratory, and that the test's specificity should be increased for field use — with higher dilutions, with a different cut-off value, or by combining other serological assays.
  • On chlamydiae: «a positive chlamydia test should be considered only a minor risk as long as abortifacient C. abortus or zoonotic C. psittaci are not detected, emphasizing the need for differential diagnosis at the chlamydial species level». The sporadic detection of C. abortus in flock 1 alone and in round D alone is interpreted as environmental contamination from ruminants shedding on the same premises, rather than as establishment in the camelid herd — a hypothesis supported by the serological negativity for C. abortus in those animals.
  • On STEC: within-flock rates are similar to those published for cattle herds (Michigan 8.2–53.7 %; Ireland 52 % and 76 %; Germany 0–100 %); EFSA classifies all STEC strains as potential human pathogens, but «the information on the presence of stx1 and/or stx2 alleles in the individual animal samples generated in this study so far is clearly limited in allowing a critical assessment of the public health risk emanating from SAC».
  • General conclusion, verbatim: «SAC do not appear to pose a greater direct risk than other livestock species. However, since SAC are often kept together with other susceptible animal species, such as cattle or sheep, they are of particular epidemiological importance as a potential source of potential pathogens». And, on interaction with people: «the detection of foodborne and abortifacient zoonotic agents underscores the importance of maintaining strict hygienic standards.

Gli autori scrivono che «this approach may have led to underestimating prevalence data, at least for pathogens known to be shed in more abundant numbers under stress-full conditions, e.g., when animals are handled by persons they are not used to on the occasion of trekking events or animal-assisted therapy, or if the animals are undernourished».
  8. Sul ruolo patogeno di BoDV-1, C. burnetii, clamidie, C. perfringens e C. difficile nei camelidi: «awaits to be assessed by analyzing samples from diseased animals».

17 · Limitations stated by the authors

Quoted, not summarized as «the authors acknowledge some limitations».

  1. Only ten flocks, albeit distributed across the territory and chosen to reflect national variety.
  2. Only animals without overt clinical disease were sampled, in the environment they are accustomed to. The authors write that «this approach may have led to underestimating prevalence data, at least for pathogens known to be shed in more abundant numbers under stress-full conditions, e.g., when animals are handled by persons they are not used to on the occasion of trekking events or animal-assisted therapy, or if the animals are undernourished». This is the most important limitation of the work for anyone working with animal-assisted interventions, and it is stated by the authors themselves.
  3. Sampling only non-diseased animals also limits conclusions about the pathogenicity of some agents for camelids, for which a correlation with clinical disease is not yet established (explicit example: C. difficile).
  4. Not all applied diagnostic methods are validated for this species, even though they follow WOAH or European reference laboratory recommendations.
  5. The peculiar antibody composition of camelids (HCAb up to 75 % of serum IgG) can distort indirect assays, both upward and downward (field 16).
  6. Data obtained by direct and indirect methods are not comparable from one agent to another. The authors write this explicitly, and this datasheet repeats it because it is the easiest trap: reading «31.6 % M. haemolamae» next to «0.7 % Coxiella» as if they were the same measurement.
  7. On STEC, assessing the public-health risk requires subtyping of the stx genes and characterization of other virulence factors, which this study did not do: strains would need to be isolated from positive samples.
  8. On the pathogenic role of BoDV-1, C. burnetii, chlamydiae, C. perfringens and C. difficile in camelids: «awaits to be assessed by analyzing samples from diseased animals».

18 · Limiti aggiunti dalla redazione

Campo distinto dal precedente.

  1. ⚠️ Un'incoerenza aritmetica interna, sulla tubercolosi. L'abstract e la sezione 3.6 danno 36 campioni non-negativi su 739 da 30 animali; la discussione scrive «the ELISA … identified 4.9 % of samples and 6.0 % of SAC as non-negative». Il 4,9 % torna (36/739 = 4,87 %). Il 6,0 % no: 30 su 449 fa 6,7 %, e 30 su 448 (escluso l'huarizo) fa 6,70 %.
  2. «Sano» lo ha deciso il proprietario. Il testo dice che gli animali erano «selected and classified as "healthy" by the animal owner prior to the study», e poi sottoposti a esame clinico.

18 · Limitations added by the editors

Field distinct from the previous one.

  1. ⚠️ An internal arithmetic inconsistency, on tuberculosis. The abstract and section 3.6 give 36 non-negative samples out of 739 from 30 animals; the discussion states «the ELISA … identified 4.9 % of samples and 6.0 % of SAC as non-negative». The 4.9 % checks out (36/739 = 4.87 %). The 6.0 % does not: 30 of 449 is 6.7 %, and 30 of 448 (excluding the huarizo) is 6.70 %. The discrepancy is small and does not change the authors' conclusion, but a reader citing "6.0 %" is citing a number that the other two sections of the article do not produce. (Editors' calculation: 30/449 with the Wilson method, z = 1.96, gives 6.7 %, 95%CI 4.7–9.4 — the same method the authors state they use elsewhere.)
  2. "Healthy" was decided by the owner. The text states that the animals were «selected and classified as "healthy" by the animal owner prior to the study», and then subjected to clinical examination. Upstream selection is thus entrusted to a non-veterinary judgement, and it operates in the same direction as the authors' limitation 2: underestimation.
  3. Crias are absent. The "at least 2 years" criterion excludes by design the young age class. For orofaecally transmitted agents this is, in many species, the class that sheds the most; this study says nothing about it, in either direction, and it should not be cited as if it did.
  4. Denominators move, and must be checked one by one. Between material shortages and logistical constraints, denominators range from 370 (clostridia) to 739 (serology), passing through 630 (Coxiella by PCR, after exclusion of 87 inhibited samples) and 442 (STEC per animal). The rate of 87 inhibited samples out of 717 — 12 % — is itself a laboratory finding worth remembering for anyone sending camelid faeces for PCR.
  5. One flock outweighs the others. Most of the Campylobacter comes from flock 4, and STEC variability among flocks ranges from 0 % to 95 % of samples. The national average, here, poorly describes any single farm: the figure to use in a discussion with a breeder is not 32.4 %, it is "anywhere from zero to almost all, and you don't know which until you check".
  6. No reporting checklist declared (field 13), no sample-size calculation: the "20 animals per visit" is justified by the authors on practical and compliance grounds, which is honest and worth stating.

19 · Finanziamento

19 · Funding

From the Funding Statement block of the full text. The study drew on samples and data from the project «Development and establishment of a multi-stage animal health management system for farms with South American camelids», part of the model and demonstration projects (MuD) on animal welfare protection. Financial support from the German Federal Ministry of Agriculture, Food and Regional Identity (BMLEH), by decision of the Parliament of the Federal Republic of Germany, disbursed by the Federal Office for Agriculture and Food (BLE), grant FKZ 2819MDT180. A.Z. Moe was supported by a scholarship from the German Academic Exchange Service (DAAD). Consistent with the funders registered in Crossref (Bundesanstalt für Landwirtschaft und Ernährung; Deutscher Akademischer Austauschdienst).

Sponsor's role: not explicitly stated for the public funder. It is instead stated for the breeders' association — see field 20.

E poi la parte che conta: «The Verein der Züchter, Halter und Freunde von Neuweltkameliden e.V. provided data and members of the society allowed sampling of their animals. Neither had a role in drafting of the study design, the sampling and sampling processing and data interpretation. Sources of the samples were pseudonymized before being transported to the laboratories».

20 · Conflicts of interest

Declared by the authors, and the declaration is more informative than average. Verbatim: the work was conducted in the absence of any commercial or financial relationships that could be construed as a potential conflict of interest; no author has proprietary, professional or personal interests in products, services or companies that could have influenced the content or opinions.

And then the part that matters: «The Verein der Züchter, Halter und Freunde von Neuweltkameliden e.V. provided data and members of the society allowed sampling of their animals. Neither had a role in drafting of the study design, the sampling and sampling processing and data interpretation. Sources of the samples were pseudonymized before being transported to the laboratories».

Statement on generative artificial intelligence: the authors state they made no use of it in drafting the manuscript. The journal separately states that alt text for the figures was generated with the support of artificial intelligence and reviewed by the authors where possible.

21 · What is missing against the reporting standard (STROBE)

Assessment carried out on the full text as read, so the absences below are real absences.

  • The design is not named in the title or abstract (STROBE 1a): a reader only understands that it is longitudinal on reaching the discussion.
  • No reporting checklist declared (field 13).
  • No sample-size calculation (STROBE 10): the "20 per visit" is justified on practical grounds.
  • No analysis of missing data (STROBE 12c): samples not tested for lack of material are stated as fact, not treated as missing data.
  • No analysis accounting for the clustered structure of the data (STROBE 12a-12d): animals are nested within flocks and samples within animals, but comparisons between species and between seasons are made with Fisher's exact test on marginal tables. With one flock of llamas only and five of alpacas only, the "llama versus alpaca" difference and the "flock versus flock" difference are partly the same thing, and the article does not separate them.
  • No sensitivity analysis (STROBE 12e).
  • Not missing, and it should be said: confidence intervals are given throughout, the method is named (Wilson) and the formula is written out in full; ethical approval has a number and a date; funding and conflicts are declared in full; the most uncomfortable limitation (healthy animals only) is stated by the authors first. On these points the article is above the average of the camelid literature.

23 · Valutazione a stelle

23 · Star rating

AxisScoreRationale, with a fact inside
A — Scientific quality★★★★☆Observational study with sample size stated at every level (449 animals, 739 sera, 723 faeces), flock selection criteria explicit and justified one by one, 95 % confidence intervals on every value with the named method and the formula written out, ethical approval with number and date, limitations stated first by the authors themselves, funding and conflicts declared in full. It does not reach five because flock selection is not probabilistic, there is no sample-size calculation, no reporting checklist is declared and the analysis ignores the clustered structure of the data.
B — Clinical utility★★★★☆Result applicable with reasonable confidence in similar situations and with explicit conditions of use: it sets the European benchmarks for eleven agents, and produces at least three immediate decisions — repeat every positive serology, type every chlamydia, consider faecal PCR inhibition. It does not reach five because it does not change a recommended practice: it confirms and reinforces it. (§11 constraint respected: B does not exceed A.)
C — Educational value★★★★☆Explains a verifiable mechanism — HCAb as a structural cause of the unreliability of indirect assays in camelids — and shows how to read a panel of eleven agents without confusing direct and indirect methods. It does not reach five because the educational piece is scattered through the discussion and is not the article's job.
D — Relevance to the species★★★★☆Study on South American camelids, in Europe, multicentre, with the population described enough (species, minimum age, size and use of every flock, region) to assess its transferability — and with llama and alpaca kept distinct even in the results, which is rare. It does not reach five because it is German and very recent (March 2026, 1 citation registered in Europe PMC): it is not yet a reference taken up by subsequent literature.

24 · Bottom line

24 · Bottom line

One can conclude that: in ten representative German flocks, clinically healthy South American camelids carry, at measurable frequency, agents of zoonotic and epizootic relevance — stx genes in 232 of 716 samples, chlamydiae in 131 of 442 animals, Campylobacter in 21 of 439 (4.8 %: 286 alpacas + 152 llamas + 1 huarizo, as in the Results of the full text; 21 of 449 would be 4.7 %) — and are widely seropositive for Schmallenberg virus (245 of 449); that Salmonella Typhimurium (1 of

  1. and Coxiella burnetii (3 sera of 738) are rare there; that the zoonotic C. psittaci was not found; and that the ELISA recommended by the European reference laboratory for bovine tuberculosis produces, in a disease-free population, a number of presumably false positives that the authors judge unacceptable for field use (36 sera of 739, with only 3 confirmations out of 19 rechecked animals).

One cannot conclude that: these values are German national prevalences, nor that they hold for Italy — ten flocks chosen by judgement are not a probability sample. Nothing can be concluded about crias, excluded by design (minimum age 2 years), nor about the pathogenic role of these agents in camelids, because only healthy animals were sampled. Nor can it be concluded that camelids transmit anything to humans: this study measures carriage, not transmission.
