◆ ASSOCIATION COMMENTARY
Camelid Research Studies APS — CRL editorial team · 08/31/2026
«MERS concerns dromedaries, not South American camelids, which are distinct species; no additional measure is warranted»: the first half is true, the second is true for the wrong reason, and the third does not follow. The statement is: «MERS concerns dromedaries, not South American camelids, which are phylogenetically distinct species; no additional measure is warranted». The first half is true. The second is true for the wrong reason. And the third does not follow.
What these nine alpacas demonstrated. Three alpacas infected intranasally shed infectious virus — not RNA, not fragments: live virus, isolated in culture — from day one to day five. Three healthy alpacas placed in the same room two days later became infected: infectious virus was isolated in two out of three, and all three seroconverted. In the three controls euthanized on day five, the virus was in the turbinates, larynx, and trachea, and in no lung lobe. At re-exposure after seventy days, the first three were completely protected.
And now the point no one expects: none of these nine alpacas showed a single symptom. No fever, no nasal discharge, unchanged appetite and temperament, and at necropsy no macroscopic lesions. An alpaca shedding a zoonotic coronavirus through its nose, in a pen, for five days, is an alpaca that, to look at, is perfectly well.
Why this changes a decision, even in Italy, where MERS is not present. Because the biological susceptibility of a species is a permanent fact, while the absence of a virus from a continent is a geographic condition, which changes with an airplane. If one day the importation of camelids from an area where MERS-CoV circulates were to be assessed, or the introduction of the virus into Europe, the statement «it does not concern South American camelids» would produce exactly the wrong decision: no isolation, no observation, no suspicion — on animals that would show nothing to see anyway. The correct formulation, which this card proposes to adopt, is: the confirmed natural reservoir of MERS-CoV is the dromedary, and it is from there that the jump to humans occurs; the alpaca and the llama do not appear to be naturally infected, but are fully susceptible in the laboratory, shed infectious virus, and transmit it to other camelids by contact — so much so that they are used as a model of the reservoir host. In Europe the practical risk is remote because the virus is not there, not because the species is refractory.
The contrast with SARS-CoV-2 must be made, because it is illuminating and goes in the opposite direction. For SARS-CoV-2 the reassurance regarding camelids is well founded, and the reason is also known: a pilot study on the susceptibility of domestic livestock did not detect shedding of infectious virus in alpacas inoculated intranasally (Bosco-Lauth AM, Walker A, Guilbert L, Porter S, Hartwig A, McVicker E, Bielefeldt-Ohmann H, Bowen RA, Emerg Microbes Infect 2021;10(1):2199-2201, DOI 10.1080/22221751.2021.2003724 — details copied from Crossref on 08/31/2026, abstract not read for this card), and the mechanism has been described in camelid nasal organoids, which are highly susceptible to MERS-CoV but not to SARS-CoV-2, in association with the absence of ACE2 expression in the upper airways while DPP4 — the MERS-CoV receptor — is expressed (Breugem TI, Riesebosch S, Schipper D, Mykytyn AZ, van den Doel P, Segalés J, Lamers MM, Haagmans BL, npj Viruses 2024;2(1):42, DOI 10.1038/s44298-024-00054-0 — details copied from Crossref, content not read directly for this card). The moral is one of method, and it holds well beyond coronaviruses: «susceptible» and «not susceptible» are not deduced from kinship between species. They are measured, one virus at a time, and the answer can be opposite for two viruses of the same family in the same animal. Dromedary, llama, and alpaca are «camelids»: for MERS-CoV the word has one meaning, for SARS-CoV-2 it has another.
Not to be confused, and it is the most frequent misunderstanding about these animals. Alpacas and llamas are immunized in the laboratory with the Spike protein to produce single-domain antibodies (VHH, nanobodies) against SARS-CoV-2: it is a vast literature and is the subject of chapter 08. That literature concerns experimental immunization and says absolutely nothing about the natural susceptibility of the species. An alpaca that produces antibodies against a virus is not an alpaca that catches that virus.
What this card cannot tell you. Whether natural infections of alpacas or llamas by MERS-CoV exist: this was not searched for in this card, and the 2016 article writes that as of that date no neutralizing antibodies had been detected in camelids outside Africa and the Middle East. It says nothing about what has been found since. Anyone who needs that updated datum should look for it, and not deduce it from here.

All text from this work may be reprinted freely. Use of these materials should be properly cited». Crossref does not register any license for this DOI, which in this case does not mean «all rights reserved» but the opposite, and must be read from the full text.
This card may reproduce: everything, text and figures, with proper citation. It is the most permissive regime of the five cards in this issue, and it is why field 15 can cite the antibody titers animal by animal.
9 · Specie e soggetti
I tre gruppi:
| Gruppo | Animali | Ruolo |
|---|---|---|
Il testo pieno è stato letto per intero e dice soltanto «Nine locally bred alpacas were obtained by private sale for use in this study».
9 · Species and subjects
Nine alpacas (Vicugna pacos), locally bred, obtained by private sale for this study, in a biocontainment facility. One week of acclimation before infection, hay ad libitum. The day before infection each animal received subcutaneously an identification and temperature-measuring transponder (Lifechip, Destron Fearing), and body temperature was monitored throughout the study.
The three groups:
| Group | Animals | Role |
|---|---|---|
| A1–A3 | 3 alpacas | experimentally infected intranasally |
| A4–A6 | 3 alpacas | infected by contact: introduced into the same room two days later and cohoused for the entire duration |
| A7–A9 | 3 alpacas | infection controls, immunologically naïve, infected at the time of re-exposure and euthanized at 5 days |
⚠️ Sex, age, and weight of the alpacas are not reported in the article. The full text was read in its entirety and states only «Nine locally bred alpacas were obtained by private sale for use in this study». This is a verified absence, not a marker for unread data, and it recurs in field 21 as a gap against ARRIVE 2.0.
10 · Domanda clinica (PICO)
10 · Clinical question (PICO)
- P — immunologically naïve alpacas.
- I — experimental intranasal infection with MERS-CoV, strain HCoV-EMC/2012, and re-exposure at 70 days.
- C — internal comparison among experimentally infected animals, contact-infected animals, and naïve controls; comparison with published dromedary data, discussed by the authors.
- O — shedding of infectious virus, animal-to-animal transmission, seroconversion, protection against re-exposure, distribution of virus across organs, histopathological and immunohistochemical picture.
As a question: does the alpaca become infected with MERS-CoV, shed it, transmit it to another camelid, and for how long does it remain protected afterward?
11 · Disegno dello studio
Gli autori ne dichiarano lo scopo dichiaratamente strumentale: «Because of logistic difficulties in working with dromedaries in containment, a more manageable animal model would be desirable».
11 · Study design
In vivo experimental study, in biocontainment, with three non-randomized groups of three animals, longitudinal observation over 70 days and a re-exposure trial, with scheduled necropsy at 5 days for the infection control group.
The authors explicitly state its purpose as instrumental: «Because of logistic difficulties in working with dromedaries in containment, a more manageable animal model would be desirable». It is not a study on the alpaca: it is a study that uses the alpaca as a model of the dromedary. The difference governs the reading of the entire card, and field 22 explains why it matters.
12 · Level of evidence
In vivo experimental evidence with stated ethical approval and stated sample size (3 + 3 + 3), without described randomization, without stated blinding, and without justification of sample size.
Consequence for the reader: it solidly establishes a biological possibility — the alpaca is susceptible, sheds infectious virus, and transmits it — and establishes it in the hardest way to contest, by isolating infectious virus and not merely viral RNA. It does not establish any frequency, any quantitative risk, and nothing about what happens in nature: nine animals in a room under biocontainment are not a population.
13 · Relevant reporting standard
ARRIVE 2.0 (in vivo experimental research), from the closed list of chapter 14 §3.3.
⚠️ The authors do not declare any reporting checklist: the full text was read in its entirety and contains no mention of ARRIVE. Nor could it: the article is from June 2016, ARRIVE 2.0 is from 2020. Under the rule of chapter 14 §3, the checklist is used retroactively to state what is missing for today's reader, not to reproach an article for failing to comply with a checklist published four years later. Field 21 is written in this spirit.
14 · Metodo
Letto nel testo pieno.
Etica. «Animal experiments were approved by the Animal Care and Use Committee of Colorado State University. Every effort was made to minimize stress and pain of the animals.»
Virus.

Ethics. «Animal experiments were approved by the Animal Care and Use Committee of Colorado State University. Every effort was made to minimize stress and pain of the animals.» No protocol number is reported → this is a verified absence.
Virus. Low-passage human isolate of MERS-CoV, strain HCoV-EMC/2012, propagated on Vero E6 cells in DMEM medium.
The one dosing/administration figure in this issue, with all its elements on the same line: 10⁷ PFU of MERS-CoV (strain HCoV-EMC/2012) per animal, by intranasal instillation, 3 mL per nostril in sterile buffered saline, in alpacas (Vicugna pacos) — Adney DR et al., Emerg Infect Dis 2016;22(6):1031-1037, full text read on 08/31/2026. The same dose was used for re-exposure at day 70.
⛔ This is not a therapy and not a clinical dosage. It is a viral inoculum dose under high-level biocontainment, in a species and a context that have no counterpart in practice. It appears in this card because our house method requires writing out in full every dosing/ administration figure that is reported, not because it is of use to anyone in clinical practice.
Experimental sequence.
- A1–A3 housed together and infected intranasally.
- Two days later, A4–A6 introduced into the same room and cohoused for the entire duration.
- Nasal swabs in viral transport medium, immediately frozen; weekly blood draws into serum tubes.
- A1–A6 maintained for 70 days after infection, then all six re-exposed intranasally at the same dose.
- Three additional alpacas (A7–A9), immunologically naïve, infected together with them as infection controls and to assess tissue distribution of replication.
- Daily nasal swabs from all animals for 5 days; A7–A9 humanely euthanized on day five.
- Swabs and sera from A1–A6 collected for two weeks after the second infection, then humane euthanasia.
Organs collected at necropsy of A7–A9 for infectious virus screening: nasal turbinates, trachea, larynx, and all four lung lobes. For histopathology and immunohistochemistry, brain, kidney, liver, skeletal muscle, heart, spleen, bladder, mesenteric lymph node, submandibular lymph node, and mediastinal lymph node were added, fixed in formalin.
Histopathology and immunohistochemistry. Fixation in 10% neutral buffered formalin for over 7 days, paraffin embedding, hematoxylin-eosin staining, evaluation by a veterinary pathologist (H.B.-O.). Immunohistochemistry for MERS-CoV antigen with rabbit polyclonal antiserum against HCoV-EMC/2012 diluted 1:1,000 as primary antibody.
Virus isolation and serology. Virus titration from swabs and tissue homogenates by plaque assay; 1 mL of transport medium considered a 10⁻¹ dilution, with serial tenfold dilutions in BA1 medium. Neutralizing antibodies measured with a plaque reduction neutralization test (PRNT), with a seropositivity threshold of 90% neutralization.

Clinica: nulla

Virus negli organi (A7–A9, giorno 5)
Anatomia patologica e immunoistochimica (A7–A9)
- Immunoistochimica: rare cellule sparse positive all'antigene virale nell'epitelio respiratorio dei turbinati e in rari leucociti intraepiteliali.

Clinical: nothing
- No appreciable increase in body temperature in any of the nine alpacas, either at first infection or at re-exposure.
- No observable nasal discharge in any alpaca, throughout the course — unlike the dromedary, in the authors' own words.
- All alpacas maintained constant activity, temperament, and feed intake.
Shedding of infectious virus
| Group | Outcome |
|---|---|
| A1–A3 (infected) | infectious virus detectable in all 3 on days 1-5 post-infection; shedding ceased by day 10 |
| A4–A6 (contact) | infectious virus isolated from A6 on days 7-14 and from A4 only on day 14; no infectious virus was isolated from A5 → 2 of 3 animals |
The authors hypothesize that A4 became infected through contact with A6 after A1–A3 had already cleared the infection, «which suggested that transmission is linked to intimate animal contact, rather than to aerosol transmission».
Seroconversion — PRNT neutralizing titers, 90% threshold
All six animals seroconverted, including A5, from which no infectious virus had been isolated.

(Values reproduced from the article's Table; A1–A3 experimentally infected, A4–A6 cohoused. Titers determined at a 90% neutralization threshold. Reproduction permitted under the public domain regime — field 8.)
First appearance of antibodies: day 14 for A1–A3; day 21 for A5 and A6; day 28 for A4.
Re-exposure at day 70
- A1–A3 (experimentally infected): completely protected, no detectable amount of virus shed.
- A4–A6 (contact-infected): minimal shedding on days 1-2, nothing on days 3-5 → partial protection.
- A7–A9 (naïve): infected, with shedding on days 1-5, then euthanized.
Virus in organs (A7–A9, day 5)
- Infectious virus detected in nasal turbinates, larynx, and trachea in all 3 animals.
- Not detected in any of the four lung lobes, in any of the three.
Pathological anatomy and immunohistochemistry (A7–A9)
- No macroscopic lesions at necropsy, in any animal.
- A8: mild squamous metaplasia of the turbinate epithelium; rare foci of mucosal erosion with minimal to mild subepithelial infiltration of neutrophils and macrophages, and fewer lymphocytes.
- All three: hypertrophy and follicular hyperplasia of the draining lymph nodes, indicative of immune activation.
- Immunohistochemistry: rare scattered cells positive for viral antigen in the respiratory epithelium of the turbinates and in rare intraepithelial leukocytes. Viral antigen not detected in any other tissue examined.
- A7 and A9: mild encephalitis with perivascular infiltrates of lymphocytes and monocytes and mild gliosis. Brain tissue tested negative on immunohistochemistry, and was not tested for virus either by isolation or PCR, because of the high risk of contamination from the nasal cavity during collection. The authors write that the etiology of that encephalitis remains unknown and might not be related to MERS-CoV infection.
16 · Interpretazione degli autori
Attribuita esplicitamente e tenuta separata dal campo 15.
- Il risultato principale, nelle loro parole: «These data demonstrate efficient virus replication and animal-to-animal transmission and indicate that alpacas might be useful surrogates for camels in laboratory studies».
- Sul confronto col dromedario: gli alpaca infetti hanno escreto quantità considerevoli di virus infettante, ma a concentrazioni inferiori a quelle riportate per il dromedario; e nessuno ha avuto scolo nasale, cosa nettamente diversa da quanto osservato nei cammelli, «which might explain the relatively low efficiency of contact transmission we observed with alpacas».
- Sull'immunità: gli animali infettati sperimentalmente erano completamente protetti alla ri-esposizione a 70 giorni, «which suggests that sterilizing immunity can be achieved»; quelli infettati per contatto hanno escreto virus infettante alla reinfezione, «albeit at much lower levels than infected control animals». Ne traggono che «widespread vaccination of dromedary camels could result in a major decrease in virus transmission to humans».
«Our data suggest that these handling practices could promote animal-to-animal virus transmission and that at the time of slaughter virus could potentially be transmitted to slaughterhouse workers.»
To date, neutralizing antibodies against MERS-CoV have not been detected in camelids outside Africa or the Middle East. However, if virus were to be introduced into immunologically naive camelid populations, it probably would be readily transmitted among animals. Many New World camelids are valued for their fiber, and such transmission might devastate fiber-related industries.»
16 · Authors' interpretation
Explicitly attributed and kept separate from field 15.
- The main finding, in their words: «These data demonstrate efficient virus replication and animal-to-animal transmission and indicate that alpacas might be useful surrogates for camels in laboratory studies».
- On the comparison with the dromedary: infected alpacas shed considerable amounts of infectious virus, but at lower concentrations than those reported for the dromedary; and none had nasal discharge, markedly different from what is observed in camels, «which might explain the relatively low efficiency of contact transmission we observed with alpacas».
- On immunity: the experimentally infected animals were completely protected at re-exposure at 70 days, «which suggests that sterilizing immunity can be achieved»; the contact-infected ones shed infectious virus upon reinfection, «albeit at much lower levels than infected control animals». They hypothesize that a second infection would also have conferred sterilizing immunity on them, and that this supports field data according to which young animals become infected and receive boosters, while most adults have acquired immunity. They conclude that «widespread vaccination of dromedary camels could result in a major decrease in virus transmission to humans».
- On the practical risk for workers: camels are moved frequently in the Middle East for grazing, exhibitions, and races; animals destined for slaughter are held for days in markets and up to 24 hours at the slaughterhouse. «Our data suggest that these handling practices could promote animal-to-animal virus transmission and that at the time of slaughter virus could potentially be transmitted to slaughterhouse workers.»
- On the significance for New World camelids — the passage that concerns Europe: «To date, neutralizing antibodies against MERS-CoV have not been detected in camelids outside Africa or the Middle East. However, if virus were to be introduced into immunologically naive camelid populations, it probably would be readily transmitted among animals. Many New World camelids are valued for their fiber, and such transmission might devastate fiber-related industries.»
- Note added in proof: the authors note that Crameri et al. independently report, in the same issue of the journal, an experimental infection and a response to re-exposure in alpacas (verified: Crameri G, Durr PA, Klein R, Foord A, Yu M, Riddell S, Haining J, Johnson D, Hemida MG, Barr J, Peiris M, Middleton D, Wang LF, Emerg Infect Dis 2016;22(6):1071-1074, DOI 10.3201/eid2206.160007 — details copied from Crossref on 08/31/2026). Two independent groups, in the same issue, with concordant outcomes: this is the circumstance that gives this finding a robustness that the nine animals alone would not have.
17 · Limiti dichiarati dagli autori
Citati, non riassunti. Sono tre, in una sezione esplicita.
- «Each of the 3 experimental groups had only 3 animals, which limited our ability to perform statistical analyses.»
- «We evaluated protective immunity 10 weeks after the original infection, which is a relatively short period and does not fully recapitulate seasonal exposures.»
- «Thus, further studies are necessary to better understand duration of immunity in camels and alpacas.»
17 · Limitations declared by the authors
Quoted, not summarized. There are three, in an explicit section.
To these must be added the caution stated in the results regarding the encephalitis of A7 and A9, whose etiology the authors state is unknown and possibly unrelated to the infection.
18 · Limitations added by the editors
- Nine animals, three per group, no described randomization. How the alpacas were assigned to the three groups is not stated. With n = 3, the randomness of assignment matters more than elsewhere, not less.
- No blinding declared. The histopathology was read by a veterinary pathologist named by initials, and it is not stated whether they knew the group of origin of the slides. On outcomes assessed by eye — «mild squamous metaplasia», «mild encephalitis» — this is the variable that weighs most.
- Sex, age, and weight of the animals are not reported (field 9). In a species where immune response and respiratory physiology change with age, this is a gap that prevents any assessment of transferability.
- No justification of sample size, and the authors correctly turn this into a stated limitation rather than concealing it.
- No ethical protocol number, although approval is stated.
- ⚠️ No conflict of interest statement appears in the full text read. The document contains Acknowledgments, Biography, Footnotes, and a bibliography, and no conflicts section. It is a verified absence, not a marker for unread data — and it reflects 2016 editorial practice more than a choice by the authors. See field 20.
- The encephalitis was not investigated to the end. The choice not to test the brain, because of contamination from the nasal cavity, is reasoned and reasonable, but it leaves the question open in two animals out of three — and in a disease in which the neurotropism of other coronaviruses is a live subject, this is the most interesting gap in the work.
- The model is explicitly instrumental. The study was created to replace the dromedary in the laboratory, not to describe the alpaca. Every conclusion transferred to the alpaca as a species — including the one in this card — must be marked as secondary use of data built for another purpose.

Stated in the Acknowledgments of the full text: «This study was supported by the Animal Models Core at Colorado State University. D.R.A. was supported by the Infectious Disease: Translational Research Training Program at Colorado State University».
Neither PubMed nor Europe PMC nor Crossref registers any grant for this article (grant list fields, funder list, and funder are all empty): the datum exists only in the full text, and it is an example of why chapter 14 §5.4 asks for it to be read.
The authors also thank the Erasmus Medical Center in Rotterdam for providing the hCoV-EMC/2012 viral isolate, and Vincent Munster and the Rocky Mountain Laboratories for the rabbit anti-MERS-CoV antiserum.
No commercial sponsor is stated.
20 · Conflicts of interest
⚠️ Not declared: no conflict of interest section is present in the full text read.
This is an absence verified on the complete document, not a retrieval gap, and must be distinguished from that of card CRL-I-01 — where the full text was not read and the statement might exist. Here the text was read in its entirety and the section is not there.
It should be added, for fairness: the stated funding is entirely academic and public (field 19), no commercial product appears, and the journal is published by a federal health agency. The absence of the statement is a reporting defect, not a clue.
| 4 · Randomizzazione | assente — nessuna assegnazione descritta |
| 9 · Procedure sperimentali | presenti e riproducibili — ceppo, dose, via, volume, terreni, tempistiche, organi prelevati |
21 · What is missing against the reporting standard (ARRIVE 2.0, Essential 10)
Assessment conducted on the full text read, and retroactive: the article is from June 2016, ARRIVE 2.0 is from 2020. The list states what is missing for today's reader, not what the authors should have known (chapter 14 §3, rule 3).
| Essential 10 item | Status |
|---|---|
| 1 · Study design | present — groups, roles, and timeline are described precisely |
| 2 · Sample size | present — 3 + 3 + 3, stated; justification is missing, which the authors correctly turn into a stated limitation |
| 3 · Inclusion and exclusion criteria | absent — «nine locally bred alpacas obtained by private sale» is all there is |
| 4 · Randomization | absent — no assignment described |
| 5 · Blinding | absent — not stated for any measure, including histopathology |
| 6 · Outcome measures | present and well defined — infectious virus by plaque assay, PRNT with a 90% threshold, histopathology and immunohistochemistry |
| 7 · Statistical methods | absent, and the authors explain why: with three animals per group they were not possible |
| 8 · Animal details | seriously incomplete — neither sex, nor age, nor weight; species and source, yes |
| 9 · Experimental procedures | present and reproducible — strain, dose, route, volume, media, timing, organs collected |
| 10 · Results | present with data per individual animal, which is more than many studies with larger n provide; without measures of dispersion, because with n = 3 none were calculated |
Items 8 and 5 are the two that weigh most. The rest of the reporting is, for a four-page 2016 journal article, remarkably complete.
23 · Valutazione a stelle
| Asse | Voto | Motivazione, con un fatto dentro |
|---|---|---|
23 · Star rating
| Axis | Score | Rationale, with one fact inside |
|---|---|---|
| A — Scientific quality | ★★★☆☆ | Experimental study with stated ethical approval and stated sample size (3 + 3 + 3), outcomes measured with solid methods — infectious virus isolated in culture, not only RNA — data reported animal by animal, limitations stated precisely and without mitigation. It does not rise to four because the ARRIVE 2.0 Essential 10 is not satisfied: randomization, blinding, inclusion criteria are missing, and above all sex, age, and weight of the animals; and no conflict of interest statement appears in the text. |
| B — Clinical utility | ★★★☆☆ | Offers a management procedure that is replicable and fully described — what to expect from an exposed camelid: early and brief nasal shedding, no clinical signs, seroconversion within 14-28 days, solid immunity at 70 days — and from this follow concrete decisions on isolation, shared airspace, and duration of observation, to be used with the caution imposed by the level of evidence. It does not rise higher because in Europe the virus does not circulate and the decision remains hypothetical. (Constraint §11 respected: B does not exceed A. And the score ≥ 3 rests on a protocol that the article reports in full — dose, route, volume, strain, timing — as the rule requires.) |
| C — Educational value | ★★★★★ | First-choice educational reference on two concepts that are constantly confused in practice: the difference between a susceptible species and a reservoir species, and the fact that an infectious animal can be clinically indistinguishable from a healthy one. Nine alpacas, no symptoms, live virus in the nose for five days, and transmission to the neighbor: anyone who needs to learn this starts here. |
| D — Species relevance | ★★☆☆☆ | Data on alpacas, so the species is the right one — but the context is very distant from the European one: experimental infection under high biocontainment, with a virus that does not circulate in Europe, in a study explicitly built to replace the dromedary in the laboratory. Two stars, and the reason must be stated in full: it is low because the virus is not there, not because the species is irrelevant. If MERS-CoV were to enter Europe, this would be the first article to reread and its D would rise at once. |
24 · Bottom line
24 · Bottom line
One can conclude that: the alpaca is fully susceptible to MERS-CoV. Inoculated intranasally, three alpacas out of three shed infectious virus on days 1-5; two alpacas out of three kept in the same room became infected by contact and all three seroconverted; the virus was found in turbinates, larynx, and trachea and in no lung lobe; and at seventy days the experimentally infected animals were completely protected against re-exposure. All of this without fever, without nasal discharge, without any clinical sign, and without macroscopic lesions.
One cannot conclude that: the alpaca is a natural reservoir of MERS-CoV, or that it represents a risk to humans in Europe. No natural infections in this species have been found; the confirmed reservoir is the dromedary; and the practical risk in Europe is remote because the virus is not there, not because the alpaca is refractory. And nothing quantitative can be concluded: nine animals, no statistical analysis, as the authors themselves state.

Closing of Issue I
What was not searched for
Chapter 14 requires every card to declare its own gaps. This issue declares its own.
- No systematic search was performed. The five articles were chosen at editorial discretion starting from chapter 09 of the KT (version 2), verified one by one against the APIs, and checked against the twenty-five cards already published to avoid repetition. There is no reproducible search strategy, and therefore no claim of having found the five best articles: they are five verified articles, on five different subjects of the issue.
- No web search was used. Only Crossref, Europe PMC, PubMed, OpenAlex (for journal status only), and DOAJ. It is a methodological choice that makes every bibliographic detail traceable to an archived API response, and that at the same time limits what could be read: two articles out of five remained closed.
- Two full texts were not read — CRL-I-01 (subscription) and CRL-I-03 (open access under a CC BY license, but not deposited in PubMed Central and not automatically retrievable from the publisher's website). All the to be checked against the full text markers in the issue are in those two cards, except one in card I-04 concerning supplementary materials.
- Subjects from chapter 09 not covered by this issue, which deserve an issue of their own: cryptosporidiosis and giardiasis (the number-one risk in animal-assisted interventions, already touched on in CRL-C-05 for the neonatal side); dermatophytosis; rabies; cystic echinococcosis; Q fever as a subject in its own right, with the diagnostic problem of complement-fixation-type assays; leptospirosis; bluetongue and Schmallenberg disease as vector-borne diseases; West Nile; and surveillance of contact-with-animals outbreaks at educational farms, for which US data exist at a scale incomparable with anything European.
- Nothing normative is asserted independently. Where references to European regulations appear (Commission Implementing Regulation (EU) 2018/1882, Commission Delegated Regulation (EU) 2020/688), they are reported as statements by the authors of card CRL-I-02, who cite them in their discussion. No regulatory text was opened during this session, and no legal consequence is inferred.
- No data on Italian prevalence were sought beyond the two carded studies (I-03 and I-04). What Central and Southern Italian camelids carry, this issue does not know.
|---|---|---|
| CRL-I-01 | 13 | non letto (abbonamento) |
| CRL-I-02 | 0 | letto |
| CRL-I-03 | 10 | non letto (CC BY, non depositato in PMC) |
| CRL-I-04 | 2 (nome del corrispondente; materiali supplementari) | letto |
| CRL-I-05 | 0 | letto |
The tally of markers
A marker signaling a datum to be retrieved is counted as open. Mentions where the marker appears to say that it does not apply (I-02, field 6; I-05, field 27) are not counted: those two are closures, not openings.
| Card | to be checked against the full text open | Full text |
|---|---|---|
| CRL-I-01 | 13 | not read (subscription) |
| CRL-I-02 | 0 | read |
| CRL-I-03 | 10 | not read (CC BY, not deposited in PMC) |
| CRL-I-04 | 2 (name of corresponding author; supplementary materials) | read |
| CRL-I-05 | 0 | read |
| Issue total | 25 | 3 of 5 read |
The tally is not a defect to hide: it is the measure of how much it costs, today, to verify the literature on camelids. An open article is read and carded in full; a closed article produces thirteen question marks, and none of those question marks is the authors' fault.
Without specific CLSI breakpoints for SACs, those defined for cattle and humans were used».

The three things this issue puts into the hands of an Italian veterinarian
First — a negative test on a camelid is not proof of absence, and in this species the reason is structural, not incidental. Camelids produce two types of IgG: mammalian-type IgG and heavy-chain antibodies (HCAb), which according to the literature cited in CRL-I-02 can represent up to 75% of total serum IgG. Every indirect assay validated on another species therefore works on a target that in the camelid is, three-quarters of the time, of a different shape. The consequences are measured in the two opposite directions, in the same issue: in CRL-I-01 a commercial assay missed all twenty alpacas infected with M. bovis; in CRL-I-02 the ELISA recommended by the European reference laboratory produced 36 non-negative sera out of 739 in disease-free herds, with only 3 confirmations out of 19 re-tested animals. Operationally: a positive serology in a camelid is repeated and confirmed with a different method before being reported; a negative serology does not close a question.
Second — there are no CLSI clinical breakpoints for South American camelids, and the antibiogram that arrives tomorrow is read against bovine or human thresholds. The authors of CRL-I-04, an Italian study, write this in a line of methods that almost no one reads: «Without specific CLSI breakpoints for SACs, those defined for cattle and humans were used». This is not a reason to ignore the antibiogram — it remains the best tool available — but to avoid reading that «S» or that «R» as pharmacological certainty, and to prefer, where it exists, a pharmacokinetic datum obtained in the species.
Third — a camelid that looks perfectly well can be an infectious camelid, and the two things cannot be distinguished by eye. In CRL-I-05, nine alpacas replicated and shed infectious virus of MERS-CoV through the nasal route, transmitted it to their pen mates, and had no fever, no nasal discharge, no decrease in appetite, and no macroscopic lesions at necropsy. In CRL-I-02, in ten German herds of animals selected because they were healthy, 232 fecal samples out of 716 carried stx genes. In CRL-I-01, twenty alpacas that tested negative on the official test were all infected with Mycobacterium bovis, and the first two died fourteen months after arrival. Operationally: clinical observation is a wide-mesh net, and what passes through it is precisely what biosecurity exists to stop. Quarantine on entry, barriers toward other species, dedicated clothing for those entering, and hand washing on leaving the animal area: four measures that cost almost nothing and that, according to CRL-I-04, are absent in Italian herds — dedicated clothing and footwear are required of the veterinarian in 2 of 12 farms and of no external visitor, while 10 of 12 farms purchase animals frequently.
